The purpose of this study is to compare the facial morphologies of an adult African-American population to an adult Caucasian-American population using three-dimensional surface imaging. Materials and Methods: Three-dimensional facial images were captured via a stereo-photogrammetric camera system (3dMDfaceTM system). One hundred images of each population (African-American and Caucasian-American) were taken. Subjects were between 19-30 years of age, normal BMI, and no gross craniofacial anomalies. All facial images were aligned and combined using Rapidform 2006 Plus Pack 2 software to produce a male and female facial average for each population. The facial averages were then superimposed for comparison and the …

Effects Of Light Spectra And Uv Radiation In Relation To Dietary Carotenoids On The Sea Urchin Lytechinus Variegatus, James Christopher Taylor Jan 2013

Effects Of Light Spectra And Uv Radiation In Relation To Dietary Carotenoids On The Sea Urchin Lytechinus Variegatus, James Christopher Taylor

All ETDs from UAB

An important factor in the marketability of sea urchin gonad is color. A marketable color is suggested to be dependent upon the in vivo accumulation of red and yellow carotenoid pigments within the gonad. Carotenoids obtained through the diet can be modified in sea urchins, but the function of these carotenoids is unknown. We hypothesize that differential light exposure can affect carotenoid deposition and, as a consequence, gonad color in sea urchins. We performed two experiments to determine if light spectra affect gonad color of Lytechinus variegatus. In the first experiment, urchins were fed a formulated feed supplemented with or …

Plga Nanospheres For Sustained Release Of Rifampicin, Alyssa Brianna Terry

All ETDs from UAB

Although Tuberculosis (TB) is often regarded as a disease of the past, it is still one of the leading killers of people worldwide. TB is caused by Mycobacterium tuberculosis, a bacterium that mostly infects the lungs but can also infect other organs throughout the body. One goal for TB treatment is to create a biocompatible, biodegradable drug delivery system that employs the use of multiple drugs while simultaneously relieving the patient from the burden of self-medicating. For this study, we optimized a procedure for making poly(lactic acid-co-glycolic acid) (PLGA) nanospheres and loading Rifampicin, one of the most potent and least …

Carbon Dioxide Enhanced Oil Recovery From The Citronelle Oil Field And Carbon Sequestration In The Donovan Sand, Southwest Alabama, Konstantinos A. Theodorou Jan 2013

Carbon Dioxide Enhanced Oil Recovery From The Citronelle Oil Field And Carbon Sequestration In The Donovan Sand, Southwest Alabama, Konstantinos A. Theodorou

All ETDs from UAB

Capturing carbon dioxide (CO2) from stationary sources and injecting it into deep underground geologic formations has been identified as a viable method for reducing carbon emissions to the atmosphere. Sedimentary rocks, such as sandstones overlain by shales or evaporites, are the preferred formations because their morphology and structure provide pore space, and containment for the long term storage of CO2. Sandstone formations have also served as repositories to migrating hydrocarbons, and are the sites of many oil recovery operations. For many depleted oil reservoirs, secondary waterflooding recovery methods are no longer efficient or economically viable, hence the application of tertiary …

The Origin, Regulation, And Function Of Tbet In Effector Cd4 T Cells During Chronic Autoimmune Disease, Wen-I Yeh Jan 2013

The Origin, Regulation, And Function Of Tbet In Effector Cd4 T Cells During Chronic Autoimmune Disease, Wen-I Yeh

All ETDs from UAB

Multiple Sclerosis (MS) is an autoimmune disease resulting from aberrant immune cells causing demyelination. We utilize the well-studied mouse model, experimental autoimmune encephalomyelitis (EAE), to decipher the underlying pathogenesis of MS. Effector CD4 T-cells play a critical role during EAE because the disease is induced by adoptively transferring myelin-specific CD4 T-cells into naïve mice. Classically, effector CD4 T-cells have been divided into distinct subsets based on their functional properties: Th1 cells produce IFNγ, Th2 cells secrete IL-4, IL-5, and IL-13, Th17 cells make IL-17A, IL-17F, IL-21, and IL-22, and regulatory T cells produce IL-10. Th1 cells were originally identified as …

Regulation Of Stim1 By O-Glcnacylation In Cardiomyocytes, Xiaoyuan Zhu-Mauldin Jan 2013

Regulation Of Stim1 By O-Glcnacylation In Cardiomyocytes, Xiaoyuan Zhu-Mauldin

All ETDs from UAB

In non-excitable cells, STIM1 and Orai1 are the primary mediators of store-operated Ca2+ entry (SOCE); however little is known about their roles in cardiomyocyte Ca2+ handling. We have previously shown that acute increases in the levels of O-GlcNAcylation (the attachment of O-linked N- acetylglucosamine (O-GlcNAc) to target proteins) are cardioprotective through attenuating Ca2+-mediated injury and increased O-GlcNAcylation blunts SOCE in cardiomyocytes. Therefore, the goals here were to determine the regulation of STIM1 and STIM1-mediated SOCE by O-GlcNAcylation in cardiomyocytes. Immunohistochemical analysis demonstrated the co-localization of STIM1 with SERCA and RyR in adult cardiomyocytes. NRVMs were transfected with the eYFP-tagged STIM1 …

The Role Of Nr4a3 And Its Agonists In Skeletal Muscle Glucose Transport And Insulin Sensitivity, Qinglan Liu Jan 2013

The Role Of Nr4a3 And Its Agonists In Skeletal Muscle Glucose Transport And Insulin Sensitivity, Qinglan Liu

All ETDs from UAB

Insulin resistance plays a central role in the pathogenesis of Type 2 Diabetes (T2DM) and involves diminished stimulation of glucose transport in skeletal muscle. Improvements in insulin sensitivity are essential for both prevention and treatment of T2DM. However, the limitation of current medications highlights the need for alternative molecular regulators of insulin action and developing new insulin sensitizers. Our laboratory has initially identified roles for the orphan nuclear receptor NR4A3 in insulin-sensitization and stimulation of glucose transport. However, the cellular effect and downstream mechanisms of NR4A3 and its agonists (e.g. 6-mercaptopurine/6-MP) on insulin effector system in skeletal muscle remain unknown. …

Investigation Of Death Receptor-5 Mediated Apoptosis And Inhibition Of Wnt/Ss-Catenin Signaling In Basal-Like Breast Cancer Stem Cells, Angelina Ix-Ik' Londono-Joshi

All ETDs from UAB

Basal-like breast cancers (BLBC) display aggressive clinical behavior attributed to the presence of cancer stem cells (CSCs). TRA-8, a monoclonal antibody that binds specifically to TRAIL death receptor 5, preferentially induces apoptosis in BLBC subtype cells; however it is unknown if TRA-8 is also cytotoxic to the CSC population. Moreover, some BLBC cell lines are resistant to TRA-8, and overcoming resistance is critical for clinical translation of this therapy. One potential approach to overcome TRA- 8 resistance is to target the Wnt/ß-catenin pathway. LRP6 is a cell surface receptor that is an indispensable element of the Wnt/ß-catenin signaling pathway that …

Blood Pressure And Tobacco Exposure Among Rural Adolescents Ages 15-18, Luz Huntington-Moskos

All ETDs from UAB

High blood pressure is a prevalent precursor to cardiovascular disease. An estimated 3% of US adolescents have hypertension. In addition, prehypertension is predictive of hypertension in adolescents, with progression between these stages at approximately 7% per year. Tobacco use/exposure is strongly linked to cardiovascular risk and disease in adults. Further, rural communities have higher tobacco use prevalence and fewer community policies restricting tobacco use. Little is known about the effects of tobacco exposure on blood pressure and the mediating effects of inflammation (salivary C-reactive protein (CRP)) in rural adolescents. The purpose of this study is to determine the relationships between …

Targeting Ovarian Cancer Through Epigallocatechin Gallate And Sulforaphane Combinational Treatment, Huaping Chen

All ETDs from UAB

The cellular development of resistance to chemotherapy contributes to the high mortality noted in patients affected by ovarian cancer. Novel compounds that can enhance efficacy of conventional chemotherapy or target both drug-sensitive and -resistant ovarian cancers directly are highly desired. Previous epidemiological studies indicate that consumption of green tea and cruciferous vegetables is inversely associated with occurrence of ovarian cancer. Therefore, revealing the effects and mechanisms of major components of green tea (epigallocatechin gallate, EGCG) and cruciferous vegetables (sulforaphane, SFN) on ovarian cancer cells will provide necessary knowledge for developing potential novel treatments for the disease. Since drug resistance generation …

Blood Pressure Control And Perceived Stress, Job Satisfaction, Life Satisfaction, And Perceived Health Status In Municipal Workers, Debra D. Baldwin

All ETDs from UAB

Although hypertension is highly prevalent, associated with serious health consequences and treatable, fewer than half of those with hypertension have achieved blood pressure control (Centers for Disease Control and Prevention [CDC], 2011b). Limited studies have examined the influence of perceived stress, job satisfaction, life satisfaction, and perceived health status may have on blood pressure control, although epidemiological evidence reveals psychological factors can mediate and negatively influence health behavior leading to hypertension (Blanchflower & Oswald, 2008; Kulkarni, Farrel, Erasi, & Kochar, 1998; Walcott-McQuigg, 2000). The purpose of this study was to explore the prevalence of hypertension, obesity, and blood pressure control …

The Molecular Basis Of Male Sex Determination During Temperature-Dependent Sex Determination, Kayla Bieser Jan 2013

The Molecular Basis Of Male Sex Determination During Temperature-Dependent Sex Determination, Kayla Bieser

All ETDs from UAB

The red-eared slider turtle (Trachemys scripta) possesses temperature-dependent sex determination (TSD) in which the incubation temperature of the developing embryo determines gonadal sex. Although a number of mammalian gene homologues have been identified in the sex determination/differentiation cascade of reptiles with TSD, the exact trigger(s) is not well understood. As such, this dissertation research addresses the mRNA expression profiles of putative sex-determining/differentiation genes (Dmrt1, Sox9, Amh, Lhx9, and Foxl2) with an emphasis on the male cascade in embryonic gonads of T. scripta. Additionally, this research addresses the influence of exogenous estrogen and aromatase inhibitors on gene expression. Lastly, this dissertation …

The Role Of Il-21 And Il-17 In Regulating Follicular T Helper Cells In Germinal Center Response Of Autoimmunity, Yanna Ding Jan 2013

The Role Of Il-21 And Il-17 In Regulating Follicular T Helper Cells In Germinal Center Response Of Autoimmunity, Yanna Ding

All ETDs from UAB

Overreactivity of the germinal center (GC) is a central feature of autoantibody-mediated autoimmune diseases. Follicular T helper (Tfh) cells are the primary T helper subset that migrates to the incipient GC and forms close contacts with GC-B cells to promote GC formation and help GC-B cell differentiation, resulting in high-affinity antibody production. Increased levels of interleukin (IL)-21 and aberrant accumulation or function of Tfh cells have been associated with autoimmune disease severity in humans and lupus-prone mice. Follicular regulatory T (Tfr) cells are the regulatory T cell subset that also localizes in the GC and inhibits GC B cell differentiation. …

All ETDs from UAB

The Cahaba prairie clover (Dalea cahaba) is one of eight species endemic to the dolomitic glades of Bibb County, Alabama. The evolutionary history of this species is currently unknown. This study tests the hypothesis that the species is a result of adaptive radiation from a widespread congener. The phylogenetic histories of several eastern species of Dalea are reconstructed using two genes that have previously been informative in plant molecular systematic studies: the internal transcribed spacer (ITS) and the matK gene. Morphological similarities between D. cahaba and a Texas-endemic species, D. tenuis, have prompted questions of whether similar traits imply common …

Determining Whether Sequence Differences In Gfa Gene Cause Alexander Disease, Lital Nehama Silverman Webster Jan 2013

Determining Whether Sequence Differences In Gfa Gene Cause Alexander Disease, Lital Nehama Silverman Webster

All ETDs from UAB

Alexander disease (AxD) is a fatal astrogliopathy usually caused by mutations in the GFA gene, which encodes glial fibrillary acidic protein (GFAP). In this document, two unusual sequence differences in GFA that are associated with AxD are characterized. The first difference is the only reported case of an AxD-causing mutation occurring in a non-coding region of GFA, c.(619-3C>G), resulting in the largest known in-frame deletion in GFAP due to abnormal splicing in a small fraction of the GFA transcripts. The second difference is the farthest reported C-terminal point mutation in GFAP, g.1277A-->T, which produces a Q426L coding change. …

Role Of Kca3.1, A Calcium-Activated Potassium Channel, In Cell Migration In The Adult Brain, Kathryn L. Turner Jan 2013

Role Of Kca3.1, A Calcium-Activated Potassium Channel, In Cell Migration In The Adult Brain, Kathryn L. Turner

All ETDs from UAB

A limited amount of migration occurs in the adult brain, including migration of neuroblasts during adult neurogenesis and pathological migration of primary brain tumors. Ion channels are commonly utilized during migration to allow shape and volume changes required for cell movement, and we specifically examine the role of KCa3.1 in the adult brain. These channels are well-suited to facilitate movement because they respond to common migration signals, i.e. intracellular Ca2+ changes, and efflux K+, allowing the outflow of water and cell volume regulation. Our findings suggest that KCa3.1 is highly expressed in migratory cells, adult neuroblasts and glioma cells, but …

The Impact Of Fine Sensor Spacing On Complex Tissue Impedance Spectra Measurements In Rabbit Ventricular Epicardium, Charlotte Mae Kent Waits

All ETDs from UAB

Our research group recently developed an instrumentation and signal processing approach that overcomes many of the obstacles associated with using small sensors to complete complex composite impedance (uCI) measurements, whose amplitude and phase depend upon the tissue's resistive and capacitive properties. The goal of this study was to test the feasibility of using smaller electrodes with finer separations than sizes traditionally employed in cardiac electrophysiologic studies with whole hearts to resolve uCI spectra in terms of magnitude (|uCI|) and phase (

The Role Of Pneumococcal Choline-Binding Protein A (Pcpa) In The Mechanism Of Virulence And Protective Immunity, Melissa Marie Walker Jan 2013

The Role Of Pneumococcal Choline-Binding Protein A (Pcpa) In The Mechanism Of Virulence And Protective Immunity, Melissa Marie Walker

All ETDs from UAB

pcpA is under control of the Mn 2-dependent repressor PsaR. In the high manganese environment of mucosal sites pcpA gene expression is repressed and at the low manganese environment of the lung and blood derepression of pcpA gene expressionis observed. Previous data has shown that immunization of mice with rPcpA elicits protection against pneumonia and sepsis but had no effect on nasal colonization. We and others have shown that pcpA expression significantly enhances adherence of pneumococci to lung epithelial cell lines and although it is a virulence factor for pneumonia in mice there is no biologically relevant role for PcpA …

Synthesis Of Ph-Responsive Multilayer Hydrogels And Amphiphilic Block Copolymers, Yun Wang Jan 2013

Synthesis Of Ph-Responsive Multilayer Hydrogels And Amphiphilic Block Copolymers, Yun Wang

All ETDs from UAB

The research presented in this thesis is focused at the design and fabrication of pH-responsive layer-by-layer (LbL) hydrogels and the synthesis of the ABA block copolymer poly(2-methyloxazoline)-block-poly(dimethylsiloxane)-block-poly(2-methyl-oxazoline) (PMOXA-b-PDMS-b-PMOXA). Ultrathin stimuli-responsive layer-by-layer hydrogels, as compared to bulk hydrogels, can respond to external stimuli in a short time range (usually within seconds) which renders them particularly useful in sensing. Moreover, layer-by-layer technique allows the hydrogel film to be deposited with controlled thickness growth. However, the fabrication of LbL hydrogels with controllable cross-linking density and swelling degree still remains challenging. In chapter two, novel pH-sensitive LbL hydrogels based on poly(4-vinyl pyridine) (P4VP) were …

Anti-Group B Streptococci Monoclonal Antibodies Protect Against Invasive Aspergillosis, Rebekah Wharton

All ETDs from UAB

Aspergillus fumigatus is a ubiquitous saprophytic fungus which can potentially cause life-threatening infections in immunocompromised individuals. Although a serious threat, current treatments are largely ineffective at reducing morbidity and mortality. To this end, we have characterized a monoclonal antibody generated against Group B Streptococci, type Ib, which also recognizes Aspergillus fumigatus conidia and hyphae. We show that this antibody appears to bind a similar epitope on both pathogens and provides a significant degree of protection in a murine model of invasive aspergillosis via a mechanism that appears to be dependent on complement. The studies we performed using this antibody should …
