Lithium, a glycogen synthase kinase 3 (GSK3) inhibitor, ameliorates experimental autoimmune encephalomyelitis (EAE), the murine model of multiple sclerosis (MS) in different mouse models. However, the mechanism of lithium's attenuation of EAE and the role of GSK3 in these processes are currently unknown. IFN-g producing Th1 cells and IL-17-producing Th17 cells are both effector cells in MS and in EAE models. However, relapsing remitting MS patients segregate into Th1 or Th17 serum cytokine signatures and each is differentially responsive to IFN-ß therapy, a first line therapy for MS. Recent literature indicates associations between inflammation, aging, and cognitive function. Inflammatory processes have also been implicated in the onset of pathological and neurodegenerative conditions. Current study aims were to examine the associations between inflammatory biomarkers and cognitive function in older adults, and to explore the interrelationships of inflammatory biomarkers, depressive symptoms, health conditions, and cognitive function. Two studies were utilized for the current project: 1) a cross-sectional sample of sedentary older adults aged 65+ from the Physical and Cognitive Exercise Study (PACES; n=54); and 2) a longitudinal study (UAB Study of Aging) of community-dwelling older African Americans and Caucasians (n=393). …

Atherosclerosis is a chronic inflammatory disease of the vessel wall and underlying cause of most cardiovascular disease. Much is understood about the nature of atherosclerosis development, including contributions from leukocytes that drive the disease process forward. The general process by which leukocytes are trafficked to sites of atherosclerosis development, and sites of inflammation in general, is understood and relies upon endothelial expressed protein adhesion molecules which serve as ligands for cognate leukocyte receptors. The goal of this dissertation is to investigate applying currently available Frequentist spatial analysis methods to cardiovascular MRI (cMRI) data. This data is typically analyzed using methods dependent summary statistics; our goal is to analyze using the full dataset. In our first paper, we explore application of spatial covariance structures to cMRI data via a generalized linear model. MicroRNAs are endogenous non-coding RNA (~22nt) that bind to the 3' untranslated region (UTR) of target messenger RNA (mRNA) and facilitate translational repression and degradation. Previous studies have demonstrated the essential role of miRNA in many biological processes, including skeletal development and tooth formation. Dlx3, a homeobox gene, is critical for craniofacial patterning and postnatal skeletal development. Mutations in the Dlx3 gene in humans result in two autosomal dominant phenotypes known as tricho-dento-osseous syndrome (TDO) and amelogenesis imperfecta with taurodontism (AIHHT). Retinitis pigmentosa is one of the most common inherited blinding disorders, affecting 1 in 4000 individuals world-wide. Approximately 30% of retinitis pigmentosa cases are dominantly inherited and approximately 30% of these dominant mutations are found in rhodopsin. Most rhodopsin mutations result in misfolded protein aggregates and subsequent apoptosis while others have been shown to result in apoptosis associated with mislocalization, diminished/enhanced activity, or faulty protein-protein interactions. This work serves to test for mechanisms by which the naturally-occurring Ter349Glu rhodopsin mutation causes rod cell death. Mycoplasmas are host-specific human and animal pathogens within the class of wall-deficient bacteria named Mollicutes and have smaller sizes and genomes than the walled bacteria. Mycoplasma genome sequences reveal few if any genes for synthesis of a glycocalyx. Nevertheless, we find that mycoplasmas produce glycolipids, glycoprotein, and polysaccharides. Using gas chromatography and mass spectrometry, glucose, mannose, rhamnose, and galactose were detected in the glycolytic species Mycoplasma pneumoniae and Mycoplasma pulmonis. All of these sugars except galactose was found in Mycoplasma arthritidis. Rhamnose was in the rare D configuration in the glycolytic species and in both D and L forms in …

Part of the challenge is the large amount of sensory information received by the brain. Depending on individu-al goals, the brain must select and react to task appropriate stimuli while ignoring other irrelevant sensory input. The purpose of this study was to compare ocular component dimensions and treated minus control eye differences measured with the Lenstar optical biometer to those measured with A-scan ultrasound in the tree shrew model of refractive development and induced myopia. A-scan ultrasonographic measures (A-scan) were taken with a 15 MHz focused pulser/receiver and compared with measures made with the Lenstar LS900 optical biometer (Haag-Streit, Basel, Switzerland). Innovative therapeutic approaches are necessary to combat these devastating cancers, and oncolytic herpes simplex virus type-1 (oHSV) deleted of the γ134.5 neurovirulence gene is a promising adjunctive therapy. The primary mechanism of tumor clearance by oHSV is lytic replication specifically within malignant cells. oHSV also stimulates tumor infiltration of cytotoxic immune effector cells that can participate in tumor clearance. O-GlcNAc has emerged as an important regulator of cellular processes such as cell signaling, transcription, translation, apoptosis, and cell cycle regulation, among others. Social control theorists posit that crime and delinquency occur among members in society because an individuals' behavior is not well regulated. Recent control theories have emphasized the importance of high self-control and strong bonds to society in deterring criminal behavior. However, one of the original theories of self-control has been largely ignored among scholars. Developed by Walter Reckless in the 1960s, containment theory suggests that individuals are pushed and pulled into crime. This study investigated factors that may influence antenatal care (ANC) utilization and their association with adverse pregnancy outcomes; defined as low birth weight, stillbirth, preterm delivery or small for gestational age among pregnant women in Kumasi. Astrocytes are the most numerous cells in the brain and play a critical role in maintaining homeostatic extracellular potassium ([K+]e). This process is mediated, in part, by a glial-specific, inwardly rectifying potassium channel, Kir4.1. Pharmacological inhibition, knock down, or complete knock out of this channel results in astrocytes with increased membrane resistance, depolarized resting membrane potential, and altered extracellular potassium dynamics. Subsequent to the dysregulation of [K+]e, Kir4.1 knockout (KO) animals suffer from ataxia, seizures, and early postnatal death. Numerous studies have shown that small soluble oligomers of Aß have higher neurotoxicity than large, insoluble aggregates and fibrils. Brain tissue analysis has shown that excessive metal ions including zinc, copper and iron are co-localized with the plaques. The purpose of this qualitative multiple case study was to explore the experiences and instruction of kindergarten teachers in an urban school district who are teaching students who were previously enrolled in a Head Start program. Eleven kindergarten teachers who taught in an urban school district in the southeastern United States participated in this study. Data sources collected for this multiple case study included: (a) semi-structured interviews, (b) observations, and (c) curriculum documents. Results from the study revealed mixed experiences teaching former Head Start students. Estrogen replacement therapy (ERT) has been shown to improve memory function in menopausal women, but increases hormone sensitive cancers, prompting a more targeted investigation of estrogen's action in learning and memory to develop more targeted therapeutics. Estrogen improves performance on the novel object recognition task by increasing phosphorylation of extracellular signal-regulated kinase (pERK) within minutes in the hippocampus. Acetylcholine receptors (AChRs) are involved in visual processing and are expressed by inner retinal neurons in all species studied to date (Keyser et al., 2000; Dmitrieva et al., 2007; Liu et al., 2009), but their distribution in the mouse retina remains unknown. Reductions in alpha 7 nicotinic AChRs (nAChRs) are thought to contribute to memory and visual deficits observed in Alzheimer's and schizophrenia (Coyle et al., 1983; Nordberg et al., 1999; Leonard et al., 2006). This dissertation research focuses on how variants in various host genes are associated with clearance of HPV in three longitudinal cohorts from the United States. I first examined the definition of the intermediate phenotype to cervical cancer, HPV persistence and then I incorporated this phenotype in all my three aims by examining variations in xenobiotic metabolism genes in HIV-1 negative and immune-related genes in HIV-1 positive females and how these contribute to HPV infection outcomes.
