Human epidermal growth factor receptor 2 positive (HER2+) breast cancer is a biologically diverse disease, in which intratumoral heterogeneity plays a significant role in tumor progression. Advanced medical imaging and image processing techniques offer the potential to noninvasively characterize the underlying intratumoral biology. Capturing these complex changes can allow for assessing therapeutic response and eventually guiding personalized therapeutic strategies. This work evaluates single and multiparametric imaging biomarkers to characterize tumor biology, identify treatment-induced changes, and predict response to targeted therapies. Maternal immune activation (MIA) refers to the occurrence of an inflammatory immune response during any stage of pregnancy. This manifests as the upregulation of proinflammatory cytokines and can be caused by air pollution, obesity, or viral infection. Glioblastoma (GBM) is the most common primary brain malignancy in adults and the gradual development of treatment resistance prevents patients from achieving remission. A primary objective in the field is to devise strategies to overcome this. Prior research has established a bidirectional relationship between sleep and pain, yet this relationship has been understudied in people living with HIV. The current study investigated the relationship between HIV status (H+/H-) and insomnia status (I+/I-) on sleep and quantitative sensory testing (QST) variables. The study was comprised of two research visits where participants were screened and completed the QST protocol. Participants completed the Insomnia Severity Index (ISI), Dysfunctional Beliefs and Attitudes about Sleep (DBAS), and actigraphy as subjective and objective measures of sleep. Idiopathic pulmonary fibrosis (IPF) is a progressive, chronic lung disease that causes scarring of the lungs. IPF is often characterized by altered growth factor signaling, excessive extracellular matrix (ECM) deposition, fibroblast hyperproliferation, and dysregulated metabolism. The etiology of IPF is unknown, but fibroblasts and myofibroblasts are the known effector cells in IPF pathology. Current treatments, such as pirfenidone and nintedanib, target growth factor signaling pathways to slow fibrogenesis, but are unable to cure the disease. Being raised in a disadvantaged neighborhood increases the risk of adult obesity, chronic inflammation, and epigenetic aging even when adjusting for family socioeconomic status (SES). Because little is understood regarding the timing of these outcomes through development and possible moderating effects of the family environment, this project investigated the relationship between neighborhood disadvantage and DNA methylation in children and whether this association varies across child age, parenting behaviors, or neighborhood cohesion. Currently there is no known cure for NF1, and more than 3000 germline pathogenic variants (PVs) in NF1 have been observed. The neurofibromin gene encodes the 320kDa NF1 protein which has multiple predicted functions. Flagellins belonging to bacteria from the Lachnospiraceae family are immunodom-inant antigens in inflammatory bowel disease. Flagellin, the basic unit of flagella utilized by bacteria for motility, is recognized by the host via toll-like receptor 5 (TLR5). Due to the diversity of flagellated bacteria, we investigated if this diversity results in distinct interac-tions between the bacteria and the host. Phylogenetic analysis on over 200 flagellin se-quences, primarily from Lachnospiraceae bacteria, revealed two divergent clusters we refer to as Group 1 (G1) and Group 2 (G2). Nonmotor deficits are present in nearly all Parkinson’s disease (PD) patients, with ~80% of these patients developing dementia. Importantly, expression of the severe GBA1 L444P heterozygous mutation increases the risk for Parkinson’s disease devel-opment by 5.43 times compared to mild mutation carriers. Currently, many PD treat-ments predominately focus motor symptoms, while treatments for nonmotor symptoms, such as visual-spatial difficulties and dementia, focus on symptom management and continue to remain elusive. Changes in the strength of GABAergic transmission is heavily influenced by posttranslational modifications and allosteric modulators like benzodiazepines and neurosteroids. O-GlcNAcylation (O-GlcNAc) is a post-translational modification that is tightly regulated by O-GlcNAc transferase (OGT) and O-GlcNAcase (OGA), which add or remove the O-GlcNAc moiety of β–N-acetylglucosamine to serine and threonine residues on proteins, respectively. Multiple System Atrophy (MSA) is a fatal and rare demyelinating synucleinopathy, characterized by the accumulation of alpha-synuclein (α-syn) in glial cytoplasmic inclusions (GCIs) within oligodendrocytes. This condition is marked by neuroinflammation, demyelination, and neurodegeneration in areas showing GCI pathology, involving antigen presentation by myeloid cells and infiltration of CD4+ and CD8+ T cells. Cardiovascular diseases are the leading causes of global death. Due to the limit regenerative capacities of adult mammalian cardiomyocytes (CMs), myocardium is repaired with non-contractile fibrotic scar after a myocardial infarction (MI). Various strategies, including apical resection (AR) and cardiac cell therapy, are being explored to promote heart regeneration after MI. With combination antiretroviral therapy (cART), the population of older people living with HIV (PLWH) is growing. There is a need to better understand everyday functioning, including instrumental activities of daily living (IADLs), to improve real-world outcomes for this clinical population. This dissertation attempts to (1) characterize factors related to self-report and performance of IADLs in PLWH, (2) examine correlates of self-awareness for functional abilities, and (3) apply a theoretical model to examine complex relationships between factors that impact self-awareness for functional abilities. Predicting wind and temperature fields around military vehicles during extended missions is crucial to avoid detectability by infrared (IR) devices. Also, the abrupt shifting of wind direction can have a significant impact on vehicle stability.
