Neurofibromatosis Type 1 (NF1) is a genetic disorder that affects the nervous system through the formation of tumors or neurofibromas. Encoded by the NF1 gene, neurofibromin functions as a tumor suppressor by converting active GTP-Ras to inactive GDP-Ras. 20% of NF1 patients carry a genomic nonsense mutation, which generates a premature termination codon (PTC) in the NF1 mRNA, leading to the formation of a truncated neurofibromin protein. Termination at PTCs can be suppressed by the incorporation of an amino acid at the site of the PTC, also known as readthrough. Human cytomegalovirus (HCMV) is a ubiquitous virus, infecting greater than 50% of most populations worldwide. Although rarely causing symptomatic infection in immunocompetent individuals, HCMV can cause life-threatening and severe end-organ disease in immunocompromised hosts including the developing fetus. HCMV can cross the placenta and is the most common viral infection of the developing fetus. Most newborn infants with congenital HCMV infection (cCMV) exhibit no clinical evidence of infection; however, up to 15% will develop long term neurodevelopmental sequelae including sensorineural hearing loss (SNHL). Acute myeloid leukemia (AML) is a blood cancer marked by the rapid growth of abnormal myeloid cells, driven by various genetic mutations. AML accounts for approximately 1% of all cancers and presents complicated prognosis and treatment. Menin inhibitors have shown potential in targeting AML. However, resistance to these inhibitors presents a significant clinical obstacle, as patients treated with menin inhibitors are increasingly developing resistance to the treatment. Using CRISPR-Cas9 mediated screening, we identified an epigenetic regulator contributing to resistance against menin inhibitors in AML. The complex nature of parallel algorithms in the Message Passing Interface (MPI) standard challenges the ability of trivially visualizing them. Within MPI libaries (e.g., MPICH, Open-MPI), there are many collectives that offer different solutions for communicating data between processes. The collective MPI_Alltoall uses various techniques such as the spread-out and Bruck algorithms to have processes transfer data between all processes. Each technique within MPI_Alltoall is best suited for different scenarios. The spread-out algorithm uses a linear number of iterations, in process count P, while the Bruck algorithm is logarithmic. Electrical excitation and mechanical function are integral to the physiologic activity of both the heart and stomach. Electrical depolarization through the myocardium triggers contraction in the heart. When normal sequences of electrical activity are disturbed, the heart’s mechanical function may also be disturbed, affecting its ability to pump blood effectively, sometimes fatally. Similarly, electrical waves called slow waves initiate gastric peristalsis. Abnormalities in slow wave propagation patterns can disturb the stomach’s ability to mechanically process gastric contents, sometimes resulting in delayed gastric emptying, nausea, and vomiting. The neuropathology includes the formation of extracellular amyloid-β plaques and intracellular neurofibrillary tangles of phosphorylated tau. Additionally, neurons become functionally impaired before degenerating and dying. In recent years, nanobodies have become important theranostic molecules, and research has focused on improving their pharmacokinetics and binding for novel targets. Nanobodies are characterized by their high kidney uptake, leading to potentially unsafe exposure of the kidneys when delivering cytotoxic payloads. Researchers hypothesize that positively charged amino acids contribute to this kidney uptake. To address this, we assessed the impact of substituting lysine residues within the 2Rs15d HER2 nanobody scaffold to alanine on its in vivo pharmacokinetics using positron emission tomography (PET) imaging. Psilocybin and other classic psychedelics may have potential for treatment of fibromyalgia, a chronic condition characterized by widespread musculoskeletal pain and other symptoms including fatigue, cognitive difficulties, and sleep problems. In this exploratory, descriptive study, an online survey was distributed among individuals with fibromyalgia to gather reports of any changes in pain related to past classic psychedelic use. Sixty-four participants with fibromyalgia completed the study, with most having used psilocybin mushrooms or LSD. This thesis focuses on two key questions: 1) how are bone development and regeneration regulated by the CREB3L1 transcription factor; and 2) what is the mechanism of action of the endoplasmic reticulum resident protein JAGN1, which when dysregulated, causes severe congenital neutropenia (SCN)? CREB3L1 regulates bone development and regeneration in mammalian models by facilitating the expression of COL1A1 and components of the secretory pathway required for collagen secretion. We generated two zebrafish models of Creb3l1 dysfunction (creb3l1ΔbZIP/ΔbZIP and creb3l1TA+/TA+). Colonial objects in Donne’s poems embrace multifaceted attributes, offering diverse reading perspectives. Considering their multifaceted aspects, it becomes impossible for a reader to ignore his poems’ interdisciplinary qualities. They often embrace philosophical, religious, political, and historical connotations. The colonial objects, appearing as “black” characters, silent women, or colonized territories in his poems, generate East-West binaries, Western subject consciousness, Eastern [mis]representations, world polarizations, and color contrasts. Ovarian cancer is the 5th leading cause of cancer deaths for women, primarily due to treatment resistant and/or recurrent disease. Most cases of ovarian cancer are not diagnosed until distant metastases have formed, which drastically reduces overall survival. The median age at diagnosis is 63 and most cases occur in patients who are peri- and post-menopausal. There is a critical gap between the clinical presentation of ovarian cancer and the preclinical models used to study the disease.
