This work develops computational methods for identifying molecular signatures from high-throughput genomic data and for modeling long non-coding RNA (lncRNA) sub-cellular localization. The response of multiple myeloma to CB-6644, a selective RUVBL1/2 complex inhibitor with potential anti-tumor activity, is analyzed to identify drug-responsive pathways and molecular signatures. Conventional gene set enrichment analysis (GSEA) often excludes low-expression genes. Here, phenotype comparison is reformulated as a supervised machine learning problem: genes most informative for discrimination are first selected using a machine learning approach, and GSEA is then applied to these machine-learning derived gene sets. Applied poultry nutrition and feed manufacture science can be described as a “three-legged stool” supported by pellet quality, feed hygienics, and nutrient availability, where imbalance in any one component can compromise overall bird performance. The research presented herein primarily addresses feed hygienics and nutrient availability, with pellet quality serving as an underlying theme, to better understand how processing conditions and dietary formulation influence performance and nutrient utilization.

Habitat fragmentation has the potential to isolate localized populations, particularly those which are understood to have limited dispersal capabilities. This isolation can reduce genetic diversity through the effects of inbreeding depression, and small populations are more susceptible to the impacts of genetic drift. In this study, I aim to understand the level of genetic connectivity between known occupied patches of Cheat Mountain salamanders (Plethodon nettingi; CMS) on managed public land. CMS are a federally threatened, high-elevation, West Virginia endemic species associated with red spruce (Picea rubens) forests. Invasive plants alter forest ecosystems by reducing species richness, habitat heterogeneity, and nutrient cycling as they rapidly displace native plants. Invasive plant composition can influence soil health through altering soil microbial associations and communities. By studying the mechanisms in which invasives alter above- and below-ground communities, we can better understand whole-ecosystem impacts and inform management decisions. This study examined the relationships between co-occurring invasion on above and belowground communities of mixed hardwood forests of north central West Virginia. It may come as a surprise to learn that the U.S. Food and Drug Administration (FDA) has done just that as a condition for allowing access to some pharmaceutical products that create a serious risk of birth defects. “Cryptids” as a theme within Appalachian (more specifically West Virginian) folklore has gained increased interest in recent years, yet limited scholarly attention has been paid as to why. Along similar lines, many West Virginians have reclaimed the West Virginia regional identity. In this study, I collected and analyzed the opinions of West Virginians on the West Virginia identity and on cryptids to argue that the popularity of cryptids within folklore (a reclamation) has coincided with the reclamation of the West Virginian identity. This Essay draws upon two decades of the author’s research on rural United States communities to explicate ways in which the regulatory state has intervened in rural areas and economies, often with the claim of protecting land, natural resources, and/or wildlife. The outcomes are frequently disastrous for the human populations of those communities and generally are not planned for or mitigated by policy. The Norman invasion of Italy in the eleventh century was a process that required political support and legitimacy for its leaders, the Hauteville dynasty. The members of this dynasty either commissioned or financed three individuals as they wrote three respective histories of the Norman conquest in order to accomplish this legitimation. Maternal electronic cigarette (ECIG) use during pregnancy raises concerns about potential risks to offspring, yet its behavioral effects remain unclear. The present studies were designed to investigate how prenatal exposure to ECIG aerosols, with and without nicotine, influences the behavior of Sprague-Dawley rat offspring. Dams were exposed to aerosols containing base liquid alone, aerosols containing base liquid with 18 mg/ml nicotine, or filtered air (control) throughout gestation. Following postnatal day 65, male offspring from 17 litters completed lever-press acquisition (Experiment 1), sustained-attending (Experiment 2), and nicotine self-administration (Experiment 3) tasks. Volunteers are the heart of the land-grant outreach model. They bring education and resources to communities across West Virginia. Volunteers have been relied upon in many aspects of Extension programming and current budget conditions have reduced the level of staffing within WVU Extension. There may be an increased demand for volunteers to help with programming. The purpose of this study was to investigate the relationship between motivation and the perceived level of well-being within the WVU Extension volunteer programs. Robots provide a unique proving ground for testing scientists’ understanding of neuromuscular systems. Synergistically, mimicking biological control laws and mechanics could offer robotics the robust and adaptive locomotion of animals. Previous work has utilized the wealth of research on insect biomechanics to develop scaled robotic models but doing so presented a problem: while these robots’ larger scale has simplified manufacturing, the scale has also shifted its mechanics from those of the modeled organisms. Animals have evolved sensory systems to help decipher important environmental signals that are vital for their survival. Feedback pathways are necessary to allow these signals to stand out in a noisy background, enabling the animal to focus on them more effectively. The electrosensory system of Apteronotus leptorhynchus allows the fish to sense the modulations caused by other conspecifics to allow for proper responses during social interactions. Electroreceptors detect these EODs, and afferent nerves project these signals to the ELL, where electrosensory information is projected to other parts of the brain. Protein biomarker detection is a critical analytical method for quantifying proteins in biological samples. It is relevant not only in medical diagnostics and research but also numerous other fields such as environmental monitoring, pharmacology, food safety, and biotechnology. Sensitive detection at picogram-per-milliliter (pg/mL) levels is essential for diagnosing diseases, monitoring progression, and tailoring treatments, especially for low-abundance biomarkers like troponin, interleukin 6, carcinoembryonic antigen (CEA), activin A, and vascular endothelial growth factor (VEGF). Despite how simple walking may seem, the locomotor control system is structurally and functionally complex. Its hierarchical organization of supraspinal and spinal networks with forward and feedback pathways has many interactions at multiple levels that are dependent on the dynamics of a high-dimensional musculoskeletal system. Having a comprehensive understanding of sensorimotor integration within a healthy locomotor control system is crucial for understanding changes to the system due to age or neurologic disease and developing effective technologies to recover mobility in those populations. Corollary discharge circuits (CDCs) are conserved mechanisms for sensory modulation during species specific behavioral programs. Many behaviors require multiplexed differential sensory modulation for proper execution. Co-transmission of fast and slow neurotransmitters allows for distinct modulation of different downstream targets within one sensorimotor circuit. However, studying co-transmission within the context of modulatory circuits like CDCs requires complete knowledge of behavioral circuits and precise tools for individually determining the implications of each transmitter on its targets. American chestnut (Castanea dentata) and various oak species (Quercus spp.) are impacted by several economically important canker pathogens in the fungal order Diaporthales, namely Cryphonectria parasitica and Endothia gyrosa (Cryphonectriaceae). Cytospora canker caused by several closely related fungi in the fungal genera Cytospora and Valsa (Cytosporaceae) have also been implicated in causing canker diseases of Quercus and Castanea in Asia, Europe and North America. Rare earth elements (REEs) are used in high-tech applications due to their electromagnetic and optical properties. As global demand for REEs surges, the limitation of primary mineral deposits has increased interest in alternative sources. One such source is acid mine drainage (AMD), which, although typically low in REE concentration, offers a promising avenue for recovery. Among the emerging recovery methods, ion exchange has shown promise due to its effectiveness at low concentrations and lower operational cost than conventional techniques such as acid leaching and solvent extraction.

Descending neurons (DNs) play a critical role in sensorimotor integration by relaying motor information to the ventral nerve cord (VNC). Within the VNC, local interneurons, other DNs, and ascending neurons (ANs) make circuits with sensory and motor neurons to modify flies’ behavior. One important AN pair in Drosophila Melanogaster is mesothoracic ascending histaminergic neurons (MsAHNs), which are known to convey predictive wing motor signals to the brain. The propensity to attribute incentive salience to reward-related cues, or sign-tracking (ST) has been linked to addiction in animal models and some initial human data. The current study explores the role of sign-tracking (ST), as a potential risk factor for problematic slot machine gambling, which abundantly features audiovisual reward-related cues, henceforth sensory features. A sample of healthy participants without gambling problems (n = 30) completed a Pavlovian-to-Instrumental Transfer (PIT) paradigm to assess ST propensity and played on a realistic slot machine simulator consisting of two versions: with sensory features (SF+) and without (SF-). Tsetse flies (Diptera: Glossinidae) are the vector of trypanosomes (Trypanosoma spp.), which are responsible for African trypanosomiasis, also known as sleeping sickness. African trypanosomiasis is a significant health and economic burden yet remains a neglected tropical disease. As an obligate blood feeder, tsetse flies are reliant on B vitamin supplementation from intracellular Gammaproteobacteria, Wigglesworthia glossinidia. Folate (B9) is essential for tsetse development and reproduction, as well as DNA methylation in animals.
