Primary cilia are highly specialized organelles that regulate signaling, trafficking, and sensory perception across multiple tissues, including the retina. The small GTPase ADP Ribosylation Factor-Like Protein 13B (ARL13B) is an atypical ARF-family protein enriched in cilia. ARL13B acts as a guanine exchange factor (GEF) for another small GTPase ARF-like Protein 3 (ARL3). Mutations in ARL13B cause Joubert syndrome, a ciliopathy characterized by brain malformation, developmental delays, and retinal degeneration. Despite ARL13B’s known ciliary localization and clinical relevance, ARL13B’s precise functions in cilia remain poorly defined. Small Unmanned Aerial Vehicles have exploded in popularity in the past 15 years among researchers and for industrial uses such as agriculture, search and rescue, and infrastructure inspection. Fixed wing and multirotor designs present two major paths that can be taken with these technologies. Fixed wing platforms have superior aerodynamic efficiency but require large areas for takeoff/landing and cannot hold a constant position in space, limiting their use. Multirotor configurations offer a more versatile platform that can maneuver in 3 dimensions independently but lack the endurance of fixed wing vehicles. Quantification of macronutrients in food requires a large investment in facilities and instrumentation as well as ongoing expenses of labor, reagents, and hazardous waste disposal. These costs pose a significant barrier to determining macronutrient quantities in the thousands of uncharacterized landrace and heirloom crops and discourage traditional crop breeding projects from ensuring that their selected crops maintain desirable macronutrient profiles in addition to target agronomic traits. This thesis will document the process of costume designing for Phoebe in Winter by Jen Silverman. It will examine the process from first read of the script through initial and final design meetings and to execution and production of the design. This will report on challenges specific to this production, such as nudity and violence on an academic stage, and the subsequent solutions. Phoebe in Winter was produced by West Virginia University School of Theatre and Dance in October of 2024. The design process began in February of the same year.

Introduction: Researchers have compared many endodontic root-end filling techniques for porosity, though few have used a micro-computed tomography analysis for comparisons. The purpose of this study was to evaluate total porosity volume of three different bio-ceramics filling materials with two techniques--hand compaction versus hand/ultrasonic compaction at an ultrasonic setting of 5% using Micro-CT Analysis.

Chicken products in grocery stores are contaminated with Salmonella that is ranked number one as a foodborne pathogen, and causes about 1.35 million cases of foodborne illness, 26,500 hospitalizations, and 420 deaths annually. The objective of this study is to determine thermal kinetic parameters of Salmonella surrogate bacteria E. faecium in ground poultry as affected by different temperatures and salt concentrations. The raw chicken was sliced up into a 500 g batch with a total of 6 batches per replicate, ground up in the meat grinder followed by inoculation with E. faecium resistant to 200 ppm Nalidixic Acid. Environmental DNA (eDNA) can serve as a useful monitoring tool for cryptic and imperiled freshwater stream fishes. eDNA can fill knowledge gaps regarding species’ distribution, site abundance, and habitat usage. The last known stronghold of the Diamond Darter (Crystallaria cincotta) in the Elk River of West Virginia provides an excellent test case to examine the utility of this non-invasive method of surveying rare species. Severe acute respiratory syndrome corona virus 2 (SARS-CoV-2) enters human respiratory cells through binding of its spike (S) protein to the ACE-2 receptor, which is highly expressed in lung cells, but is also found in over 150 other cell types encompassing major organs including GI enterocytes. SARS-CoV-2 causes gut dysbiosis in infected individuals and patients with COVID-19 and also coincides with lower alpha diversity of the individual’s gut microbiome. Certain gut microbiome-associated genera are lower in abundance in COVID-19 patients including Lachnospira, Prevetolla, Roseburia, Bifidobacterium, and Faecalibacterium while genera such as Bacteroides, Enterococcus and Proteobacteria are increased during a SARS-CoV-2 …

Sleep plays a crucial role in memory consolidation, neuronal plasticity, and cognitive function. Individuals with autism spectrum disorder (ASD) often experience sleep disturbances, which are positively correlated with symptom severity. However, on the surface, sleep architecture is remarkably normal under baseline conditions in many rodent models of ASD. The goals if this dissertation were to investigate whether regulation of sleep, biochemical signaling during sleep, and downstream consequences of sleep on behavior are altered in two mouse models of ASD. Multiple myeloma (MM), is a hematological cancer originating in plasma cells. Currently multiple myeloma represents a significant clinical challenge due its incurable nature mainly due to inevitable drug resistance. MM is characterized by abnormal plasma cell proliferation within the bone marrow and production of abnormal immunoglobulin resulting in a plethora of physiological abnormalities. Although a rare malignancy, MM accounts for 1% of all cancers and currently has five-year survival rate of around 50%. Beginning in adolescence and throughout adulthood, women are at an increased risk for anxiety and mood disorders compared to men. Several risk factors for these disorders have been identified, but it is not clear how they interact. One such factor is pregnancy. The normative changes to the brain and behavior that occur during and after pregnancy are associated with an increased risk for anxiety and mood disorders. Exposure to adverse childhood experiences (ACEs) has also been associated with an increased risk of mood disorders, and compared to men, women appear to be particularly vulnerable to the lasting effects of stress …

Social-emotional competence is a prerequisite to school performance and participation in daily tasks. Children in West Virginia face social and emotional barriers including the effects of the opioid epidemic leading to children placed in foster care, born with neonatal abstinence syndrome, and being exposed to toxic stress in the home environment. Additionally, the COVID-19 pandemic has impacted children's social-emotional functioning due to isolation periods. This dissertation explores the ecological politics of movement in Romantic literature. It contributes to a growing body of scholarship that investigates the important role that walking played in nineteenth-century literary culture. Many authors from this period cast walking as a practice that frees up not only the body but the mind as well, as the steady rhythm of the feet lulls the walker into a state of self-absorbed introspection and heightened creativity. Edible insects are emerging as sustainable and nutritious alternatives to conventional protein sources. However, concerns about food safety, particularly the presence of pathogenic bacteria such as Escherichia coli and Listeria monocytogenes, remain a barrier to consumer acceptance and industrial use. This study evaluated the effectiveness of thermal processing in inactivating E. coli, L. monocytogenes, and their non-pathogenic surrogates (E. coli JM109 and Listeria innocua) in both fresh and dehydrated forms of Tenebrio molitor (mealworm) and Acheta domesticus (cricket). This dissertation explores how three Irish modernist writers, Elizabeth Bowen, Dorothy Macardle, and James Joyce, incorporate and reimagine tropes from the Victorian Gothic Revival to address Ireland’s uneven modernity in the first half of the twentieth century. I argue that the Irish Gothic tradition is essential for these writers to express their reflection and reaction to ongoing cultural and social turmoil in Ireland’s process of modernization during the foundation of the postcolonial Irish state. The rifle was one of many material objects which colonists of British North America brought with them from Europe. Starting in the early eighteenth century, German-speaking settlers in Pennsylvania began to manufacture rifles from locally made and imported parts. These rifles became associated with other material objects – namely a system of dress, which included wearing a whitetail deer’s tail in the hat, the hunting shirt, and forms of Native American dress – which became emblematic of some British colonists who used rifles in hunting and war. This research introduces an innovative approach for managing autumn olive (Elaeagnus umbellata) on reclaimed surface mines through the integration of machine learning algorithms and unmanned aerial systems. The proliferation of autumn olive presents significant ecological challenges on disturbed landscapes, necessitating more efficient detection and control methods. This study evaluated Random Forest classification and deep learning approaches using the U-Net architecture for identifying autumn olive across four phenological stages using high-resolution imagery. The RGB U-Net model achieved F1-scores of 0.927, 0.912, and 0.871 for autumn olive detection during peak, late, and senescence stages respectively, outperforming both multispectral and vegetation …

Tuberculosis (TB) is caused by Mycobacterium tuberculosis (Mtb), and it has plagued humans for most of recorded history. Despite strong efforts to prevent and treat TB, it remains the leading cause of death due to a pathogen and is most prevalent in certain regions of Asia and Africa. Currently, the only approved vaccine to prevent TB is the live-attenuated Bacillus Calmette-Guérin (BCG) vaccine. Most countries have a universal BCG vaccination policy, but the vaccine ultimately fails to be protective. From 2018 to 2023, Kiko bucks born from January 1st to March 15th were performance tested at West Virginia University to evaluate growth rate, ultrasound carcass characteristics and parasite resistance under an artificial infection. Kiko bucks were submitted by producers from 17 different states and over 5 performance tests, 830 Kiko bucks were evaluated. Hypercholesterolemia, or atherosclerotic cholesterol, is characterized as elevated concentrations of total cholesterol (TC), low-density lipoprotein-cholesterol (LDL-C), and non-high-density lipoprotein-cholesterol (non-HDL-C). According to the Academy of Nutrition and Dietetics (AND), vegan diets have proven to be the most effective diets for improvement of heart disease risk factors. On the other hand, the American Heart Association (AHA) recommends the Mediterranean-style diet as a diet pattern for the prevention of heart disease, stroke, and reduction of risk factors, such as obesity, diabetes, hypercholesterolemia, and hypertension. Foodborne illness outbreaks associated with fresh produce continue to pose a public health risk in the United States. Although the Food Safety Modernization Act (FSMA) Produce Safety Rule (PSR) established science-based standards to prevent contamination, many small-scale farms are exempt from these requirements. Consequently, voluntary adoption of Good Agricultural Practices (GAP) varies widely across regions. Acid mine drainage (AMD) is a widespread issue across the globe, in which some of the most critical cases are created by the mining industry. Researchers have investigated ways to derive benefits from AMD, such as precipitating rare earth elements (REEs) to generate potential revenue from AMD treatment and supplement market demand. However, the bulk of precipitated sludge is composed of iron and aluminum, which also have market value but are discarded in current practice.
