The solubility profiles for metharbital, butabarbital, thiopental and thiamylal were determined in ethanol- water solvent systems at 25 c, in an attempt to define an approximate correlation between therapeutic activity and various solubility relationships. The relative lipophilicity of a drug molecule is an important factor in the physical and chemical processes involved when the therapeutic agent is introduced into a biological system. Binary solvent mixtures aid in delineating the relative polarity, in terms of dielectric constants, of these drugs. Spectrophotometric and, where applicable, gravimetric analyses, were utilized to determine the concentration of drug, in mg./ml., in the 41 solvent systems …

Characterization Of The Effect Of Methylchloroform Inhalation On Hepatic Drug Metabolism In The Rat, Arnold Merwin Olshan

Male rats were exposed to methylchloroform (1,1,1-tricholoethane) vapors (concentration about 2500 PPM) for 24 hours and the effects of this treatment on drug responses and hepatic drug metabolism studied 24 hours later to determining the effect of methylchloroform on inducible hepatic drug-metabolizing enzymes. Exposure to methylchloroform vapor significantly decreased the sleeping time of the rat in response to hexobarbital (120 mg/kg, i.p.) and zoxazolamine (80mg/kg, i.p.). Rat blood and liver methylchloroform content measured immediately following exposure suggested that the effects of methylchloroform were caused by a systemic action. Several 1-alkylbenzoylpyridinium halides containing alkyl side chains of one to fifteen carbons were screened for pharmacological activity. A comparison of potency was based on the ability to produce mortality in 50 percent of mice (LD 5D). The following parameters were investigated in a select number of compounds representative of the series: acute intraperitoneal LD50 in rats; EDSO (hypotensive) in rats; effects on the autonomic nervous system of anesthetized cats and dogs as indicated by alteratio11s in blood pressure; effects en the nictitating membrane in anesthetized cats; direct effects upon blood vessels employing hind quarter perfusion in intact rats; effects on …

Investigation Of The Role Of Dopamine In Maintenance Of Arterial Hypertension, Harold Llewellyn Crossley

The relationship between urinary dopamine levels and arterial blood pressure was studied in normal, unilaterally nephrectomized, and renal hypertensive male albino rats. Experimental hypertension was produced by a unilateral nephrectomy followed by compression of the contralateral renal artery. The two operations were spaced three weeks apart to allow for recovery. Nine weeks after-the-second operation; mean systolic blood pressure of the renal hypertensive control group increased to 170 mm Hg. Mean systolic blood pressure of the diethyldithiocarbamate (DDC)-treated renal hypertensive group decreased abruptly after two weeks of treatment and remained at 105 mm Hg throughout the nine week period.
