Translational Control Of Vesicular Stomatitis Virus Protein Synthesis: Isolation Of An Mrna-Sequestering Particle, Craig A. Rosen, Herbert L. Ennis, Paul S. Cohen

An mRNA-ribonucleoprotein particle (mRNP) was found in vesicular stomatitis virus (VSV)-infected Chinese hamster ovary cells. The particle was present 3 and 4.5 h after infection but was barely discernible at 2 h. The mRNP (buoyant density, 1.56 g/cm³), which cosedimented with viral nucleocapsid in a sucrose density gradient at approximately 120 to 160S, was separable from nucleocapsid (buoyant density, 1.31 g/cm³) by CsCl density gradient centrifugation. It contained all five VSV mRNAs and, almost exclusively, viral N protein. Some host mRNA and host protein was also present in the particle. The intact mRNP was incapable of …

Incubation of purified human serum albumin with D-[1-¹⁴C]galactose (5 mM) or D-[1-¹⁴C]glucose (5 mM) in vitro for 7 days under physiological conditions resulted in the time-dependent accumulation of radioactivity into trichloroacetic acid-precipitable material. Comparative studies indicated that the rate of sugar incorporation into albumin increased with increasing pH and temperature of incubation and followed a first order dependence with regard to monosaccharide and albumin concentrations. The extent of nonenzymatic galactosylation of human albumin was approximately 300% greater than the extent of nonenzymatic glucosylation under equivalent experimental conditions. This study is a component of a more comprehensive investigation at the Marine Ecosystems Research Laboratory, (MERL), University of Rhode Island, that was undertaken in order to assess the potential for eutrophication in both its meanings as a natural and man-induced process occurring in the upper portions of Narragansett Bay and its watershed. This assessment required identification and quantification of the various inputs in order to establish the annual loading and eventual effects of the four important nutrients; ammonia, nitrate-nitrite, silicate and phosphate.

This paper will examine the historical impetus for Vessel Traffic System (VTS) development in the United States. Cost benefit techniques utilized to establish the VTS requirements are discussed and the data base upon which the analysis is conducted is critiqued. General Accounting Office criticism of the Coast Guard's VTS development process are analyzed. Finally VTS is examined as a single component in the improvement of port logistics, which must be arrayed against other alternatives to make the most effective use of scarce resources. Historic wrecks exist in many of the navigable waters of the United States, particularly along the Atlantic and Gulf coasts. Attention will mainly focus, however, on the waters off Florida. These often dangerous waters, as part of a long-established trade route, contain more than their share of historic wrecks. As a number of these are Spanish galleons, wrecked on their way to Europe with value cargoes of gold, silver and precious stones, claim to such sites has been the subject of bitter disputes. The conservation measures used in the Interim Fishery Management Plan for Atlantic Groundfish (IFMP) were analyzed. The management techniques used include a minimum mesh size, minimum fish size and haddock spawning area closure. However the mesh regulation does not adequately address the IFMP conservation objectives. Without amendment, the IFMP cannot prevent overfishing as it is required to do by the Magnuson Fishery Conservation and Management Act (MFCMA). The use of a non-numeric Optimum Yield (OI) for groundfish is inappropriate without a definition of overfishing and without a contingency plan to prevent overfishing.

A series of toxins isolated from the dinoflagellates, Gonyaulax spp., sheds new light on the structure of the receptors on excitable membranes. Biosynthetic origins of these toxins and tetrodotoxin, another sodium channel-specific marine toxin, are discussed. A new polyether type of toxin from the dinoflagellate, Gymnodinium breve, are also presented.

The Monitoring Of Clostridium Perfringens Spore Density In Bottom Sediments And It's Applications, David J. Emerson

Clostridium perfringens is an anaerobic, spore-forming bacterium that is consistently associated with human fecal wastes. Two extraction-separation procedures were developed for use in conjunction with the mCP membrane filtration method for the enumeration of C. perfringens spores in bottom sediments. In the simpler of the two procedures a distilled water suspension of the sediment sample is pulse sonicated for 10 seconds and allowed to settle. Portions of the supernatant are removed and assayed by the mCP membrane filtration method. This procedure is recommended for general usage. A more complicated procedure using a two-phase separation system was also developed for use …

Age Related Changes In The Induction Of Hepatic Mixed-Function Monooxygenase System In Miniature Pigs: Effects Of Pentachloroanisole, Pentachlorophenol And Phenobarbital, Alan Paul Agins

Pentachloroanisole (PCA), an environmental degradation product of the biocide pentachlorophenol (PCP), has been detected in the food chain. The metabolic fate of PCA was examined in miniature pig hepatic microsomes, in vitro. The compound was shown to be a substrate for a cytochrome P450-dependent demethylation reaction, which results in the regeneration of the parent compound, PCP. A disproportionately large increase in PCA demethylase activity (PCADM) following pretreatment with phenobarbital suggests that the compound is preferentially metabolized by specific-inducible form(s) of cytochrome P450.

Benthic Filter Feeding: A Natural Eutrophication Control, C. B. Officer, Theodore J. Smayda, R. Mann

The importance of the benthic filter feeding community as a natural control on eutrophication is considered. The important environmental factors favorable for such a control are relatively shallow water depths and a dense benthic filter feeding commmunity of small animals. The criteria are summarized in the equivalence of the water recycling time, τ_ρ, for the benthic community and the time constant, τ_ρ, for phytoplankton growth. The criteria are applied specifically to the conditions that exist in South San Francisco bay.

Nutrient Level In Saudi Arabia Wheat Flour, Abdulrhaman Al-Khalifah

The chemical composition and protein quality of four varieties of wheat flour grown in Saudi Arabia (Rice Mexi-Pak, Red Mexi-Pak, Lugemi, and Moaeah), and one type of home bread (Korsan) made from Moaeah were evaluated. Analysis performed included proximate analysis, thiamine, riboflavin, iron, amino acid composition and protein efficiency ratio (PER). Protein and iron were higher in Moaeah and Lugemi. The protein quality assessed by PER was higkest for Rice Mexi-Pak and lowest for Korsan, the cooked product. Chemical scores based on the essential amino acid content of the proteins were calculated; lysine and/or methionine was the limiting amino acid …
