Novel Dnm1l Variants Impair Mitochondrial Dynamics Through Divergent Mechanisms, Jdc Pereira Baptista Dec 2022

Novel Dnm1l Variants Impair Mitochondrial Dynamics Through Divergent Mechanisms, Jdc Pereira Baptista

Peninsula Medical School

Imbalances in mitochondrial and peroxisomal dynamics are associated with a spectrum of human neurological disorders. Mitochondrial and peroxisomal fission both involve dynamin-related protein 1 (DRP1) oligomerisation and membrane constriction, although the precise biophysical mechanisms by which distinct DRP1 variants affect the assembly and activity of different DRP1 domains remains largely unexplored. We analysed four unreported de novo heterozygous variants in the dynamin-1-like gene DNM1L, affecting different highly conserved DRP1 domains, leading to developmental delay, seizures, hypotonia, and/or rare cardiac complications in infancy. Single-nucleotide DRP1 stalk domain variants were found to correlate with more severe clinical phenotypes, with in vitro recombinant …

A Global Building Occupant Behavior Database, B Dong, Y Liu, W Mu Dec 2022

A Global Building Occupant Behavior Database, B Dong, Y Liu, W Mu

School of Art, Design and Architecture

This paper introduces a database of 34 field-measured building occupant behavior datasets collected from 15 countries and 39 institutions across 10 climatic zones covering various building types in both commercial and residential sectors. This is a comprehensive global database about building occupant behavior. The database includes occupancy patterns (i.e., presence and people count) and occupant behaviors (i.e., interactions with devices, equipment, and technical systems in buildings). Brick schema models were developed to represent sensor and room metadata information. The database is publicly available, and a website was created for the public to access, query, and download specific datasets or the …

Sources Of Copper Into The European Aquatic Environment, Sean Comber, Genevieve Deviller, Iain Wilson, Adam Peters, Graham Merrington, Pasquale Borrelli, Stijn Baken

School of Geography, Earth and Environmental Sciences

Chemical contamination from point source discharges in developed (resource-rich) countries has been widely regulated and studied for decades; however, diffuse sources are largely unregulated and widespread. In the European Union (EU), large dischargers report releases of some chemicals, yet little is known of total emissions (point and diffuse) and their relative significance. We estimated copper loadings from all significant sources including industry, sewage treatment plants, surface runoff (from traffic, architecture, and atmospheric deposition), septic tanks, agriculture, mariculture, marine transport (antifoulant leaching), and natural processes. A combination of European datasets, literature, and industry data were used to generate export coefficients. These …

‘Gentle Humour’ To ‘Savage Satire’: Austen Obituaries On Her Death, Its Centenary And Bicentenary, A Bautz

School of Law, Humanities and Social Sciences

A ‘modest and retiring lady novelist’, ‘the adored aunt of boisterous nephews and nieces rather than a blue stocking’ to some, to others an author with a ‘strong feminist streak’, ‘biting irony’, and ‘imperishable wisdom’: this essay investigates cultural perceptions of Austen at three points in time as evidenced through obituary articles published on her death in 1817, at the 1917 centenary, and at the bicentenary in 2017. Rather than focusing on how her texts are interpreted by readers or critics of her novels, by viewers and consumers of tv, film, and multimedia adaptations, websites and other media, the chapter …

Democracy And Schooling: The Paradox Of Co‐Operative Schools In A Neoliberal Age?, Tom Woodin, Cath Gristy

Plymouth Institute of Education

From the first co-operative trust school at Reddish Vale in Manchester in 2006, the following decade would witness a remarkable growth of ‘co-operative schools’ in England, which at one point numbered over 850. This paper outlines the key development of democratic education by the co-operative schools network. It explains the approach to democracy and explores the way values were put into practice. At the heart of co-operativism lay a tension between engaging with technical everyday reforms and utopian transformative visions of an educational future. A new arena of debate and practice was established with considerable importance for our understanding of …

AbstractMore than two billion people worldwide have suffered thyroid disorders from either iodine deficiency or excess. By creating the national map of groundwater iodine throughout China, we reveal the spatial responses of diverse health risks to iodine in continental groundwater. Greater non-carcinogenic risks relevant to lower iodine more likely occur in the areas of higher altitude, while those associated with high groundwater iodine are concentrated in the areas suffered from transgressions enhanced by land over-use and intensive anthropogenic overexploitation. The potential roles of groundwater iodine species are also explored: iodide might be associated with subclinical hypothyroidism particularly in higher iodine …

Standardized Multi-Omics Of Earth’S Microbiomes Reveals Microbial And Metabolite Diversity, Jp Shaffer, L-F Nothias, Lr Thompson, Earth Biome Project 500 Consortium

School of Biological and Marine Sciences

Despite advances in sequencing, lack of standardization makes comparisons across studies challenging and hampers insights into the structure and function of microbial communities across multiple habitats on a planetary scale. Here we present a multi-omics analysis of a diverse set of 880 microbial community samples collected for the Earth Microbiome Project. We include amplicon (16S, 18S, ITS) and shotgun metagenomic sequence data, and untargeted metabolomics data (liquid chromatography-tandem mass spectrometry and gas chromatography mass spectrometry). We used standardized protocols and analytical methods to characterize microbial communities, focusing on relationships and co-occurrences of microbially related metabolites and microbial taxa across environments, …

The twentieth century started with engineers’ new concepts and ideas to develop the built environment, with ideas ranging from reducing weight to automated industrialisation, enhancing energy efficiency, etc. However, they failed to predict the negative impact of automation on the environment and its resources. Therefore, future technologies proposed by engineers should consider the adverse effects on nature and the surrounding ecosystem. Using the science of ecology and integrating it into building typology can help avoid unwanted scenarios. Such technologies prioritise the performance of buildings and cities by using simulation tools to explore more ideas for designing and testing their performance. …

Strengthened Multi-Stakeholder Linkages In Valuation Studies Is Critical For Improved Decision Making Outcomes For Valuable Mangroves – The Malaysian Case Study, Soon Loong Lee, Amy Yee Hui Then, Hong Ching Goh, Caroline Hattam, Andrew Edwards-Jones, Melanie C. Austen

School of Biological and Marine Sciences

Mangrove forests in Southeast Asia are continuously declining as a result of unsustainable practices, partly due to limited recognition of the value of mangrove services in land use decision making. Valuation practitioners have assumed that monetary valuation should inform local and national decision makers to ensure sustainable management of mangrove resources. For ecosystem service valuation to be of use to decision makers, best practices should be adhered to such as having straightforward policy questions and strong stakeholder engagement from the onset of valuation studies, suitable choice of valuation methodologies, and the ability to effectively demonstrate causal links between drivers of …

‘Gentle Humour’ To ‘Savage Satire’: Austen Obituaries On Her Death, Its Centenary And Bicentenary, Annika Bautz

School of Law, Humanities and Social Sciences

This chapter invez stigates cultural perceptions of Austen at three points in time as evidenced through obituary articles published on her death in 1817, at the 1917 centenary, and at the bicentenary in 2017, in order to trace shifting trends in the cultural appreciation of the author. The vantage point here is that of the newspaper obituarist or columnist who may or may not have read Austen ahead of their commissions. Turning to non-specialists can reveal much about an author’s cultural status outside of the academy, where assumptions may be just as, if not more, entrenched. Half of the obituaries …

Withdrawing Antipsychotics For Challenging Behaviours In Adults With Intellectual Disabilities: Experiences And Views Of Experts By Experience, G De Kuijper, J De Haan, S Deb, R Shankar

Peninsula Medical School

People with intellectual disabilities (PwID) are frequently prescribed long-term antipsychotics for behaviours that challenge (BtC) despite the lack of proven effectiveness and the increased risks for side effects of these medications in this population. National and international good clinical practice guidelines recommend deprescribing antipsychotics for BtC, which is often not successful due to environmental and other factors. The involvement of all stakeholders, including PwID, is crucial for deprescribing. However, studies showed that PwID and/or their families are often not involved in decision-making regarding the (de)prescribing of antipsychotics despite their desire to get involved. Moreover, studies on the views of PwID …

Agri-food supply chain (AFSC) resilience is receiving increasing attention as AFSC stakeholders perceive its benefits in recovering from unexpected disruptions. However, which resilience capabilities are more effective in mitigating AFSC risks remains unclear. To address this gap, this article presents a novel AFSC risk and resilience analysis based on a systematic literature review (SLR). In total, 95 journal articles on AFSC risk and resilience management published between 2004 and 2020 are analyzed to identify key risks and resilience capabilities in AFSCs, the relationships, correlations and causalities between them, and research gaps and future research directions in the field. Surface faulting earthquakes are known to cluster in time from historical and palaeoseismic studies, but the mechanism(s) responsible for clustering, such as fault interaction, strain-storage, and evolving dynamic topography, are poorly quantified, and hence not well understood. We present a quantified replication of observed earthquake clustering in central Italy. Six active normal faults are studied using 36Cl cosmogenic dating, revealing out-of-phase periods of high or low surface slip-rate on neighboring structures that we interpret as earthquake clusters and anticlusters. Our calculations link stress transfer caused by slip averaged over clusters and anti-clusters on coupled fault/shear-zone structures to viscous flow laws. …

Future Of Maritime Autonomy: Cybersecurity, Trust And Mariner's Situation Awareness, Misas Jd Palbar, R Hopcraft, K Tam

School of Engineering, Computing and Mathematics

As technology evolves, the level of automation in the maritime industry grows, and will continue to grow significantly, as the industry strives for fully autonomous vessels. Current maritime operations on board rely on a mixture of automated processes, human decision making, and human interventions. In the future autonomy may lead to the removal of the mariner physically on board, increasing remotely operated vessels. The remote nature of these operations will subject mariners to new operational risks, such as a potential reduction in Situational Awareness (SA) and/or cyber threats. This article will discuss the importance of SA in maritime operations, and …

Using The Mental Health Wellness Framework In Dental Practice, A Plessas, R Witton, M Paisi, F Ellwood, E Mccoll

Peninsula Dental School

Dentistry is recognised as a stressful profession and dentists perceive their profession to be more stressful than other healthcare professions.1 An alarming upwards trend in deterioration of mental health and wellbeing in the profession has been observed in recent years, with up to 87% of UK general dental practitioners reporting symptoms indicative of burnout.2 The COVID-19 pandemic has further heightened anxiety levels in all members of the dental team and especially in frontline dental nurses.3,4,5 In a study conducted at the dental department within Guy's Hospital in London at the beginning of the pandemic, nearly a quarter of dental nurses …

School of Geography, Earth and Environmental Sciences

Direct links between carbonaceous chondrites and their parent bodies in the solar system are rare. The Winchcombe meteorite is the most accurately recorded carbonaceous chondrite fall. Its pre-atmospheric orbit and cosmic-ray exposure age confirm that it arrived on Earth shortly after ejection from a primitive asteroid. Recovered only hours after falling, the composition of the Winchcombe meteorite is largely unmodified by the terrestrial environment. It contains abundant hydrated silicates formed during fluid-rock reactions, and carbon- and nitrogen-bearing organic matter including soluble protein amino acids. The near-pristine hydrogen isotopic composition of the Winchcombe meteorite is comparable to the terrestrial hydrosphere, providing …

The Hidden Cost Of A Smartphone: The Effects Of Smartphone Notifications On Cognitive Control From A Behavioral And Electrophysiological Perspective, Joshua D. Upshaw, Carl E. Stevens, Giorgio Ganis, Darya L. Zabelina

School of Psychology

Since their release in 2007, smartphones and their use have seemingly become a fundamental aspect of life in western society. Prior literature has suggested a link between mobile technology use and lower levels of cognitive control when people engage in a cognitively demanding task. This effect is more evident for people who report higher levels of smartphone use. The current study examined the effects of smartphones notifications on cognitive control and attention. Participants completed the Navon Letter paradigm which paired visual (frequent and rare target letters) and auditory (smartphone and control sounds) stimuli. We found that overall, participants responded slower …

Transposable Elements Maintain Genome-Wide Heterozygosity In Inbred Populations, Kort H De, Sylvain Legrand, Olivier Honnay, James Buckley

School of Biological and Marine Sciences

Elevated levels of inbreeding increase the risk of inbreeding depression and extinction, yet many inbred species are widespread, suggesting that inbreeding has little impact on evolutionary potential. Here, we explore the potential for transposable elements (TEs) to maintain genetic variation in functional genomic regions under extreme inbreeding. Capitalizing on the mixed mating system of Arabidopsis lyrata, we assess genome-wide heterozygosity and signatures of selection at single nucleotide polymorphisms near transposable elements across an inbreeding gradient. Under intense inbreeding, we find systematically elevated heterozygosity downstream of several TE superfamilies, associated with signatures of balancing selection. In addition, we demonstrate increased heterozygosity …

Acute cardiorespiratory breathlessness accounts for one in eight of all emergency hospitalizations. Early, noninvasive diagnostic testing is a clinical priority that allows rapid triage and treatment. Here, we sought to find and replicate diagnostic breath volatile organic compound (VOC) biomarkers of acute cardiorespiratory disease and understand breath metabolite network enrichment in acute disease, with a view to gaining mechanistic insight of breath biochemical derangements. We collected and analyzed exhaled breath samples from 277 participants presenting acute cardiorespiratory exacerbations and aged-matched healthy volunteers. Topological data analysis phenotypes differentiated acute disease from health and acute cardiorespiratory exacerbation subtypes (acute heart failure, acute …

Social interactions evolve continuously. Sometimes we cooperate, sometimes we compete, while at other times we strategically position ourselves somewhere in between to account for the ever-changing social contexts around us. Research on social interactions often focuses on a binary dichotomy between competition and cooperation, ignoring people’s evolving shifts along a continuum. Here, we develop an economic game – the Space Dilemma – where two players change their degree of cooperativeness over time in cooperative and competitive contexts. Using computational modelling we show how social contexts bias choices and characterise how inferences about others’ intentions modulate cooperativeness. Consistent with the modelling …

Widespread, and sometimes inappropriate use of psychotropics in adults with intellectual disability has been an international concern. These medicines have been used to treat mental health conditions, but also, controversially, some types of behaviours not necessarily associated with the diagnosis or in the absence of a relevant diagnosis. Results from the Intellectual Disability Supplement to the Irish Longitudinal Study on Ageing (IDS-TILDA) study of older adults with intellectual disability in Ireland revealed that 60% were taking psychotropics in 2010. In the intervening decade changes in regulations, policy, and increased decongregation of people with intellectual disability have taken place likely influencing …

Surface water flooding is an issue which has required an increased level of addressment over the past two decades, with the methods used to combat flood events seeing an evolution. This evolution has been influenced heavily by multi-scale legislations and policies, which has pushed for more holistic methods for pluvial flood management. This review will analyse how Bristol City Council have implemented these multi-scale pieces and what has been put in place to encourage sustainable flood management. This will be done through a purposive review of the literature and an extensive review of legislation and policies on a national, European, …

Express: A Curse Of Knowledge Or A Curse Of Uncertainty? Bilingualism, Embodiment, And Egocentric Bias., S Samuel

School of Psychology

The ability to adopt others’ perspectives – our ‘Theory of Mind’ – underpins social interaction. Nevertheless, adults are imperfect perspective takers, demonstrating egocentric biases. Here, a series of experiments assessed whether 1) embodying an agent’s physical perspective (working out whether he held something in his left or right hand), or 2) being bilingual, would benefit perspective taking. Participants were shown a scenario in which an agent puts a ball in one of four boxes. When he returns later, the boxes have been rearranged. Participants then judge how likely the agent is to look in each of the four boxes first. …

A Roadmap To Understanding Diversity And Function Of Coral Reef-Associated Fungi, Anna Roik, Miriam Reverter, Claudia Pogoreutz

School of Biological and Marine Sciences

Tropical coral reefs are hotspots of marine productivity, owing to the association of reef-building corals with endosymbiotic algae and metabolically diverse bacterial communities. However, the functional importance of fungi, well known for their contribution to shaping terrestrial ecosystems and global nutrient cycles, remains underexplored on coral reefs. We here conceptualize how fungal functional traits may have facilitated the spread, diversification, and ecological adaptation of marine fungi on coral reefs. We propose that functions of reef-associated fungi may be diverse and go beyond their hitherto described roles of pathogens and bioeroders, including but not limited to reef-scale biogeochemical cycles and the …
