Quiescence is a distinct cell cycle phase, termed G0, in which growth, transcriptional and translational activity, and replication are halted. Importantly, quiescent cells are viable and can return to the cell cycle. Quiescence is a feature of all organisms and a key hallmark of stem cells; however, the regulation of quiescence is still poorly understood.
Prior studies have implicated chromatin in the regulation of quiescence. In aim 1 I conducted a comprehensive screen of H3 and H4 histone mutants to identify chromatin features that may affect quiescence. Single-molecule localization microscopy super-resolution methods rely on stochastic blinking/binding events, which often occur multiple times from each emitter over the course of data acquisition. Typically, the blinking/binding events from each emitter are treated as independent events, without an attempt to assign them to a particular emitter. Here, we describe a Bayesian method of inferring the positions of the tagged molecules by exploring the possible grouping and combination of localizations from multiple blinking/binding events. The results are position estimates of the tagged molecules that have improved localization precision and facilitate nanoscale structural insights. Gas-surface scattering involves the conversion of energy in different forms in the impinging molecule, possible bond breaking and forming, and the energy transfer across gas-solid interface. Possible energy dissipation channels include adiabatic energy transfer to the motion of surface phonons and non-adiabatic interactions with surface electron-hole pairs. A complete understanding of the interplay of energy dissipation and physical/chemical changes in the molecule is vital for many important applications such as materials fabrication and heterogenous catalysis.

This research addresses how student participation in Science, Technology, Engineering, and Mathematics (STEM) project-based learning (PBL) education activities encourages underrepresented minority student achievement in STEM career field trajectories. Seven New Mexico high school counselors and 12 STEM organization personnel were interviewed during this study. Their responses represent the nuanced professional voices where New Mexico public education intersects with STEM student interest and cultural influence.
For students, STEM PBL can foster deep integration across educational disciplines and enhance STEM career trajectory interest and readiness. STEM education converged with PBL methodologies has the ability to leverage community support while broadening student networks. …

Quantifying the flow of energy and nutrients through food webs is foundational to understanding the structure and function of ecosystems. Here, I utilize the stable carbon and nitrogen isotope analysis of individual amino acids to trace the movement of essential amino acids through terrestrial and freshwater food webs in New Mexico, USA. I first explore isotopic patterns among co-occurring terrestrial plants and aquatic algae. I then combine this molecular isotopic approach with 16S and 18S rRNA sequencing to demonstrate the importance of gut microbiota as sources of essential amino acids to wild mammalian hosts. There are burgeoning numbers of elderly and a significant number of their adult children provide them with some level of care. To date, there has been little research on elderly parents’ perspective as they communicate with their adult children about the care provided by their children. This dissertation addresses this gap by identifying the nature of the communication, as portrayed in film, which occurs between an elderly parent (EP) and their caregiving adult child (AC).

My thesis focuses on Mary Shepard Greene Blumenschein and Ernest L. Blumenschein, married artists born in the late 1860s. Ernest Blumenschein was an important regional artist and member of the Taos Society of Artists (TSA). Paintings by Blumenschein and other TSA members promoted tourism in the Southwestern United States through annual exhibitions and their use in advertising the Atchison, Topeka and Santa Fe Railway (AT&SF). Mary Greene Blumenschein was an award-winning painter and illustrator whose work focused on images of women at the beginning of the twentieth century, however, she is now a secondary and obscure figure in art history. …

According to self-determination theory, people are intrinsically and/or extrinsically motivated to self-regulate their behavior (Deci & Ryan, 2012). Sport fans watch games for intrinsic rewards such as having fun and supporting a team, which, from the identity fusion perspective (Swann Jr et al., 2009), derive from their visceral sense of oneness with a team. Differential gene expression analysis has the potential to discover candidate biomarkers, therapeutic targets, and gene signatures. How to save money when using an unaffordable sample is a practical question. The case-cohort (CCH) study design can blend the economy of case-control studies with the advantages of cohort studies. But it has not been seen in the medical research literature where high-throughput genomic data were involved.

Measurements of plant water potential provide fundamental insights into how plants are interacting with the environment to manage water needs. However, monitoring water potential is difficult, time consuming, and frequently destructive. Several methods have been in development that attempt to monitor the water status of plant tissues in a simple, continuous, and non-destructive manner. Lead zirconate titanate (PZT) has been a material of interest for sensor, actuator, and transducer applications in microelectromechanical systems (MEMS). This is due to their favorable piezoelectric, pyroelectric and ferroelectric properties. While various methods are available to deposit PZT thin films, radio frequency (RF) magnetron sputtering was selected to provide high quality PZT films with the added capability of batch processing. These sputter deposited PZT films were characterized to determine their internal film stress, Young’s modulus, composition, and structure. Objective: The purpose of this study was to compare the acute physiological, perceptual, and enjoyment responses between a single bout of escalating density training (EDT) and traditional resistance training (TRAD).
Methods: Twelve physically active males (n = 6) and females (n = 6) (age: 21.4 ± 3.0 years; weight: 69.8 ± 7.9 kg; height: 172.9 ± 12.4 cm; maximal oxygen consumption (VO2max): 44.7 ± 9.2 ml · kg-1 · min-1) performed both EDT and TRAD. Schistosoma haematobium, the agent of urogenital schistosomiasis, and related schistosomes are transmitted by members of the genus Bulinus. Each of the 38 Bulinus species vary in their ability to vector schistosome species and non-schistosome trematodes resulting in a patchwork of snail-parasite compatibilities. Accurately identifying snail intermediate hosts and the disease-causing parasites they transmit is critical for snail control strategies and the management of human schistosomiasis. Towards these ends, this thesis identifies bulinid species and the parasites they transmit and implicates certain species in the transmission of S. haematobium. Hummingbirds are the smallest of migratory birds. Their use of hovering flight to feed on nectar results in prodigious rates of energy consumption. At night, when birds are unable to feed, body fat must be used to stay warm. These characteristics make migratory flights to and from northern breeding areas very expensive. To offset these costs, hummingbirds alternate long flights with stopover periods of refueling along the way. Body fat stores, which may account for as much as 40% of a bird's body weight, also provide a buffer if birds find themselves in a nectar "desert" when the need to …

Polyploidy-whole genome duplication-is common in plants. Studies over the last several decades have documented numerous mixed-ploidy populations. Whether arising from secondary contact or via whole genome duplication events, the persistence of mixed populations depends on the ability of the minority cytotype to overcome the negative frequency dependent effects of outcrossing with other ploidies, known as Minority Cytotype Exclusion. One mechanism of overcoming Minority Cytotype Exclusion is microbially-mediated niche differentiation (MMND), wherein cytotypes occupy different niches via interactions with different sets of microbes. RESEARCH QUESTION: Is rigorously selected COVID-19 convalescent plasma with neutralizing anti-SARS-CoV-2 antibodies an efficacious treatment for adults hospitalized with COVID-19?
STUDY DESIGN AND METHODS: This was a multicenter, blinded, placebo-controlled randomized clinical trial among adults hospitalized with SARS-CoV-2 infection and acute respiratory symptoms for < 14 days. Enrolled patients were randomly assigned to receive one unit of COVID-19 convalescent plasma (n = 487) or placebo (n = 473). The primary outcome was clinical status (disease severity) 14 days following study infusion measured with a seven-category ordinal scale ranging from discharged from the hospital with resumption of normal activities (lowest score) to death (highest score). The primary outcome was analyzed with a multivariable ordinal regression model, with an adjusted odds ratio (aOR) < 1.0 indicating more favorable outcomes with convalescent plasma than with placebo. In secondary analyses, trial participants were stratified according to the presence of endogenous anti-SARS-CoV-2 antibodies ("serostatus") at randomization. The trial included 13 secondary efficacy outcomes, including 28-day mortality.

This dissertation evaluates the interconnection between health and political behaviors in a polarized context such as the United States. The COVID-19 pandemic unveiled the current political polarization and the structural health disparities among racial communities. Using the system theory delineated by David Easton, this dissertation demonstrates that health attitudes and behaviors in the electorate can influence voting behaviors, as was the case with the 2020 US presidential election. This dissertation explores how hearing parents with deaf/hard of hearing children come to understand deafness. This mixed methods study used an online survey and multiple case studies (volunteers from survey). Participants were asked about early intervention experiences and beliefs about deafness. The survey had 74 respondents and five families participated in the interviews. Survey participants’ beliefs about deafness were primarily medical, focusing on the perceived barriers caused by deafness and the remediation of those barriers through spoken language options.
