The Toxoplasma Gondii Protein Rop2 Mediates Host Organelle Association With The Parasitophorous Vacuole Membrane, Anthony P. Sinai, Keith A. Joiner
Microbiology, Immunology, and Molecular Genetics Faculty Publications
Toxoplasma gondii replicates within a specialized vacuole surrounded by the parasitophorous vacuole membrane (PVM). The PVM forms intimate interactions with host mitochondria and endoplasmic reticulum (ER) in a process termed PVM–organelle association. In this study we identify a likely mediator of this process, the parasite protein ROP2. ROP2, which is localized to the PVM, is secreted from anterior organelles termed rhoptries during parasite invasion into host cells. The NH2-terminal domain of ROP2 (ROP2hc) within the PVM is exposed to the host cell cytosol, and has characteristics of a mitochondrial targeting signal. In in vitro assays, ROP2hc is …

Solid–state fermentation is experiencing renewed interest for industrial enzyme production. Previous studies on the effects of fungal inoculum size on product yield have focused on spore inoculum. However, some organisms require vegetative inocula. This study investigated the effects of initial inoculum colony age, vegetative inoculum size, and duration of fermentation on the production of fungal biomass and phytase in solid–state fermentation using Aspergillus niger grown on wheat bran and soy meal. Public involvement in highway improvement presents a set of complex problems involving many stakeholders.Since visualizations are increasingly regarded as essential tools in this process, a clearer understanding of the specific merits of advanced visualization techniques and their potential contributions must be developed.As a step towards integrating visualizations into an improved public planning paradigm termed Structured Public Involvement (SPI), this paper investigates the utility and performance of three visualization modes, termed 2D, 3D and VR (Virtual Reality), for a case study highway in Central Kentucky.

The gene sequence for ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) large subunit (LS)εN-methyltansferase (protein methylase III or Rubisco LSMT) from a plant which has a des(methyl) lysyl residue in the LS is disclosed. In addition, the full-length cDNA clones for Rubisco LSMT are disclosed. Transgenic plants and methods of producing same which have the Rubisco LSMT gene inserted into the DNA are also provided. Further, methods of inactivating the enzymatic activity of Rubisco LSMT are also disclosed.

In the present study we evaluated the precision of the ELISA method to quantify caffeine in human plasma and compared the results with those obtained by gas chromatography. A total of 58 samples were analyzed by gas chromatography using a nitrogen-phosphorus detector and routine techniques. For the ELISA test, the samples were diluted to obtain a concentration corresponding to 50% of the absorbance of the standard curve. To determine whether the proximity between the I50 of the standard curve and that of the sample would bring about a more precise result, the samples were divided into three blocks according …

The efficient utilization of federal funds in improving the water quality and aquatic habitat of the region requires a mechanism for assessing and evaluating the impacts of the proposed and ongoing projects as well as some mechanism for prioritizing the allocation of additional funds. In order to evaluate the effectiveness of these projects it is important to provide a formal monitoring and assessment program based on sound scientific principles. This MiniReview updates and expands the MiniReview of aluminium toxicokinetics by Wilhelm et al. published by this journal in 1990. The use of 26Al, analyzed by accelerator mass spectrometry, now enables determination of Al toxicokinetics under physiological conditions. There is concern about aluminium in drinking water. The common sources of aluminium for man are reviewed. Oral Al bioavailability from water appears to be about 0.3%. Food is the primary common source. Al bioavailability from food has not been adequately determined. Industrial and medicinal exposure, and perhaps antiperspirant use, can significantly increase absorbed aluminium. Alfalfa is known as the queen of the forage crops and for good reasons. Alfalfa is an excellent forage for dairy cattle because it provides nutrients needed by dairy cows in a package which is highly digestible and cost effective. Alfalfa can be grazed directly by cattle or harvested as hay, haylage, or batage. Dairy farmers will purchase locally-grown hay if it is high quality (RFV greater than 150). Horse owners frequently mention cleanliness, color and cutting when it comes to choosing hay. Cleanliness should be the most important factor in selecting horse hay. Hay that contains dust or mold can inflame the respiratory tract. It is also possible for hay mold to affect other systems in the horse such as the digestive system and liver. Some horses suffer from a chronic respiratory condition that seriously impairs their ability to breathe normally. This condition, commonly known as heaves, worsens when horses are fed moldy or dusty hay. Horses with heaves are exercise intolerant and thus have little value except …

Bloat refers to excessive accumulation of gas in the rumen. Bloat results when an animal can not eructate or "belch up" gases produced in the process of rumen fermentation. The gas may be in the free form or may be mixed with rumen contents in the form of froth. It occurs both on pasture and in feedlots and can be a major cause of death in cattle wherever intensive farming is practiced. Additional losses can include decreased milk production and reduced rate of gain. Although legumes may increase the opportunity for bloat to occur, fear of bloat should not keep …

We have discussed the importance of stage of maturity and its impact on quality on many occasions. We have presented research on many aspects of hay and haylage management. An overall theme for all the discussions has been "harvest for quality," with emphasis on saving the leaves during the entire harvesting process. Crash data in Kentucky show that the fatal crash rate on two-lane rural roads is substantially higher than on any other type of road. Improvements have been proposed at some locations on this type of road which involve either upgrading the existing two-lane road or adding lanes resulting in a four-lane road. As part of the public information process, highway officials have been asked to document the previous results of this type of construction. Bone Cutting, Placement, And Cannibalism? Middle Preceramic Mortuary Patterns Of Nanchoc, Northern Peru, Jack Rossen, Tom D. Dillehay

Mortuary practices of the Middle Preceramic period (ca. 8500-4000 B.P.) are discussed for the Nanchoc region of the upper Zaña Valley, northern Peru. Careful breaking, cutting, and placement of human bones from adult males during the Las Pircas Phase (8500-6000 B.P. ) gave way to more haphazard breakage and discard during the subsequent Tierra Blanca Phase (6000-5000 B.P.). The evidence of cannibalism is considered. Bone breakage, cutting, and possibly cannibalism is believed to have been part of a broader process of ritualization that mitigated the spiritual danger of the transition from hunting-gathering to horticulture.

Submerged And Solid-State Phytase Fermentation By Aspergillus Niger: Effects Of Agitation And Medium Viscosity On Phytase Production, Fungal Morphology And Inoculum Performance, Maria Papagianni, Sue E. Nokes, Keith Filer

Qualitative relationships between agitation levels and medium viscosity, Aspergillus niger morphology and phytase production were investigated in submerged and solid-state fermentations. Overall phytase production increased with increasing shaker speeds from 150 to 300 rpm, although specific growth rates and phytase production rates were higher at 150 rpm for 72 h from inoculation. Fungal morphology was greatly influenced by agitation with the morphological forms of small pellets and entangled mycelia predominating at 150 rpm, while the free filamentous form was obtained at 300 rpm. Upon inoculation of SSF, increased productivities were obtained from inocula grown at 150 rpm. A shift towards …

Clenbuterol is a β2 agonist/antagonist bronchodilator, and its identification in post-race samples may lead to sanctions. The objective of this study was to develop a specific and highly sensitive serum quantitation method for clenbuterol that would allow effective regulatory control of this agent in horses. Therefore, clenbuterol-d9 was synthesized for use as an internal standard, an automated solid-phase extraction method was developed, and both were used in conjunction with a multiple reaction monitoring liquid chromatography-tandem mass spectrometry (LC-MS-MS) method to allow unequivocal identification and quantitation of clenbuterol in 2 mL of serum at concentrations as low as 10 …

Russian’s second BLUE term goluboj ‘light blue’ constitutes a well-known exception to the Berlin and Kay basic color-term typology. If other Slavonic languages do not have a second BLUE term, then the special position of Russian requires explanation; if there is evidence pointing to two basic terms for BLUE, we have a second set of data for investigating the evolution of this unusual color system. The languages genetically closest to Russian, Ukrainian and Belarusian, are examined. Findings of the list task, a simple and elegant test for psychological salience of color terms, provide strong evidence that Ukrainian and Belarusian have …
