Expression And Purification Of Porcine Pid1 Gene In Escherichia Coli, Huan Wang, Xiaoling Chen, Zhiqing Huang, Bo Zhou, Gang Jia, Guangmang Liu, Hua Zhao

In this study, in order to scale up the production of recombinant porcine phosphotyrosine interaction domain containing 1 (pPID1), a pET-28a (+)-pPID1 expression plasmid was constructed and transformed into Escherichia coli Rosetta (DE3). The recombinant pPID1 was then purified and identified by western blotting, and was also analyzed in vitro for its function. The recombinant protein was tagged with only a His6 tag at its C-terminus, which could be conveniently purified by affinity column. The protein could be induced for efficient expression with 0.75 mM IPTG for 8 h at 30 °C, yielding approximately 3 mg/L. In vitro biological activity …

Orange peel is an agroindustrial waste rich in pectin and known to be an inducer for pectinase production. The use of this low-cost substrate for the production of an industrially important enzyme, polygalacturonase (PG), can be an alternative way to turn this waste into a value-added product, contributing to the reduction of environmental waste disposal problems. This study was conducted to determine the tolerance of safflower (Carthamus tinctorius L.) cultivars (Dinçer, Remzibey-05, and Yenice) against salt stress based on some physiological and biochemical parameters at the vegetative stage. Eighteen-day-old plants were subjected to salt stress [0 (control), 75, 150, 225, and 300 mM NaCl concentrations] for 12 days, which led to a significant decrease in growth parameters (stem growth, fresh weight, and relative water content) and the photosynthetic pigment contents of the safflower cultivars. A variety of chemical and physical methods are utilized for enhancing bacterial competence. This study examines the influence of the polyamines putrescine and spermidine on the efficiency of androgenesis in anther cultures of 2 carrot cultivars, Kazan F_1 and Narbonne F_1, and the effect of putrescine on the process of plant regeneration from androgenetic embryos. In the Kazan F_1 variety, an increase in the number of obtained embryos was achieved using each of the 2 polyamines separately and in combination. In contrast, no beneficial effects of polyamines on the efficiency of androgenesis were observed in the Narbonne F_1 variety. The isolation and phylogenetic analysis of new yeast strains represent an important basis for the development of practical applications in biotechnology, industry, and bioremediation. The yeast strain RG1 was isolated from oil-polluted soil and characterized using morphological observations and the API 20C AUX system. The amplified ITS1-5.8S rDNA-ITS2 region had 653 bp and the restriction profiles obtained with Cfo I, Dde I, Hae III, Hinf I, and Hpa II endonucleases showed high similarities with those from Rhodotorula glutinis reference strains. The gene encoding ribulokinase araB from Anoxybacillus kestanbolensis AC26Sari was cloned and sequenced. The recombinant protein was expressed in Escherichia coli BL21 under the control of isopropyl-\beta-D-thiogalactopyranoside-inducible T7 promoter. The enzyme, designated as AC26RK, was purified with the MagneHis Protein Purification System. The molecular mass of the native protein, as determined by SDS-PAGE, was about 61 kDa. AC26RK was active throughout a broad pH (pH 5.0-10.0) and temperature (50-75 °C) range, and it had an optimum pH of 9.0 and optimum temperature of 60 °C. Conyza blinii H.Lév. is a traditional Chinese medicinal plant that is distributed mainly in southwestern Sichuan and northern Yunnan. Its characteristic product is blinin, which has, among other properties, antigastric ulcer activity, and can serve as a quality-control standard for such medicine. The problem is that C. blinii only produces low yields of blinin. As a diterpene, blinin is likely formed by the methylerythritol phosphate pathway. While 1-deoxy-D-xylulose-5-phosphate synthase (DXS) is the first rate-limiting enzyme in diterpenoid biosynthesis, it is a switch in the pathway. Peste des petits ruminants (PPR) is an important viral disease of sheep and goats and is endemic in all regions of Turkey. In this study, PPR virus infection was investigated by RT-PCR assay based on the fusion (F) gene in PPR-suspected sheep and goat samples. PPR virus RNA was detected in 65 small ruminants (51 sheep, 14 goats) from independent outbreaks during 2008-2012 in provinces in the central and Mediterranean regions and the central-west part of the Aegean region in Turkey. Breast cancer has been treated successfully with selective estrogen receptor antagonists (SERMs) such as tamoxifen, receptor-depleting agents such as fulvestrant, and aromatase inhibitors such as anastrozole. Selective progesterone receptor modulators (SPRMs or PRMs) have not been studied as much and are currently under investigation for inhibition of mammary carcinogenesis in animal models and breast cancer prevention trials in women. Cancer stem cells (CSCs) are a small subset of cancer cells within a tumor that are responsible for tumorigenesis and contribute to drug resistance. The CSC displays an anchorage-independent survival, active DNA-repair capacity, and relative quiescence and is capable of self-renewing and maintaining tumor growth and heterogeneity. At the molecular level, there are several signaling pathways (e.g., Wnt/Beta-catenin, Notch, and Hedgehog) to control CSC properties and alteration of these pathways has been recognized as an essential step for CSC transformation. Although genome-wide RNA interference (RNAi) screens for mitotic genes are not new in the literature, most of them lack the cytological characterization of the cell karyotype as for chromosome integrity. Here, the effects of RNAi on chromosome structure in S2 cultured cells of Drosophila melanogaster were analyzed. The cytological phenotype of 1132 genes selected by coexpression with known mitotic genes was scored. Cytological and statistical analysis of the treated cells allowed the identifying of 81 loci whose inactivation brings a level of chromosome breakage significantly higher than in the control. The tumor suppressor genes (TSGs) ING3 and ING5, members of the inhibitor of growth gene family, are effective in inhibition of cell growth and induction of apoptosis. However, in many cancer types, one of the alleles of a TSG is lost through carcinogenesis, while the remaining allele is usually inactivated through a process called loss of heterozygosity (LOH). We previously identified a panel of GC-induced candidate genes in children with ALL and in other biological systems using whole genome expression profiling. Zataria multiflora Boiss., with the common name Avishan-e-Shirazi, is native to Iran. This herb has been found to possess varied pharmacological properties. In the present study, for the first time, colon chemopreventive effects of Z. multiflora essential oils (0.01% and 0.1% in the diet) in rats treated with 1,2-dimethylhydrazine (DMH) were demonstrated. For this purpose, the oxidative stress/antioxidant parameters (lipid peroxidation, glutathione, superoxide dismutase, catalase, and ferric reducing ability of plasma) concomitant with xenobiotic metabolizing enzymes (CYP450 and GST) were considered. A Revision Of Centaurea Sect. Centaurea (Asteraceae) From Iran, Massoud Ranjbar, Kazem Negaresh

A taxonomic review of Centaurea L. sect. Centaurea (Asteraceae) in Iran was carried out. Seven taxa are recognized and 1 of them is described as a new species: Centaurea joharchii Ranjbar & Negaresh. This new species has been described and illustrated from Khorasan Province, SE Iran. A new combination is proposed: C. kermanshahensis (Wagenitz) Ranjbar & Negaresh. In addition, C. aggregata Fisch. & C.A.Mey. ex DC. subsp. aggregata is typified and C. diffusa Lam. is reported here as a new record for Iran. Finally, a key to all species of the section Centaurea in Iran is provided.

The morphological variability of the baculum (os penis) of 147 adult specimens of species in the genus Nannospalax from 58 localities in Turkey was examined using morphological and numerical taxonomic methods. Significant differences among all of the Turkish species in the genus were determined by morphological and biometrical comparison of the bacula, and the results of this study showed that N. nehringi and N. xanthodon are separate species and that the names are not synonyms. We present a review of technological methods developed in recent years for the purpose of gallium nitride nanostructuring, with the main focus on fabrication of thin GaN membranes. In particular, we report on traditional methods of wet etching undercutting for membrane manufacturing, technologies applied for the fabrication of photonic crystal structures based on GaN nanomembranes, double side processing, and surface charge lithography. Prospects of membrane applications in photonic devices, sensors, and microoptoelectromechanical and nanoelectromechanical systems are discussed, taking into account the advantageous piezoelectric, optical, and mechanical properties of GaN and related III-V nitride materials.

To investigate the expression of epidermal growth factor receptor¬-HER1 (EGFR), cerbB2 (HER2), p16, and p53, as well as the relationship of the expression of these genes with conventional parameters in squamous cell carcinoma (SCC) of the larynx. To demonstrate galectin-3-immunoreactivity in the undiluted essential oil of Origanum hypericifolium when applied to the ultraviolet B (UVB) irradiated skin of mice. Despite the rise in type 2 diabetes prevalence worldwide, we do not have a method for early risk prediction. The predictive ability of genetic models has been found to be little or negligible so far. In this study, we aimed to develop a better early risk prediction method for type 2 diabetes. The Taxonomic Status And Geographic Distribution Of The European Hare (Lepus Europaeus Pallas, 1778) In Turkey (Mammalia: Lagomorpha), Yasi̇n Demi̇rbaş, İrfan Albayrak

This study was based on 125 specimens of hares collected from 61 different localities in Turkey between 2006 and 2010. We present diagnostic characters, pelage coloration, external, cranial and phallic measurements, skull and phallus features, hair structure, and collection localities of hares from Turkey. Additionally, we analyze morphometric variations of 5 regional populations according to the physiography/orography of the study area. Among the morphometric variables, 10 characters (weight, tail length, basal length, condylobasal length, rostral breadth, height of braincase, foramen incisiva length, occipitonasal length, profile length, and mandible length) are found to be significantly different between the populations (ANOVA, P < 0.01). Principal component analysis of 22 morphometric measurements is performed, and 71.56% of total variance is explained through 5 principle components. The Thracian specimen is distinguished from other populations on the first 2 canonical discriminant functions and the cluster dendrogram. The hair structure, as examined by scanning electron microscope, is found to be flattened and imbricate. Our Anatolian specimens are discussed at the subspecies level, comparing them to the relevant literature, and it is concluded that Lepus europaeus in Anatolia is represented by only one subspecies, Lepus europaeus syriacus.

Morphological characteristics of the resting egg, which is important to ensure the continuity of generations, were examined to understand if there were morphologically significant differences among 3 cladoceran species. Resting eggs floating on the water surface were collected by a plankton net with a mesh size of 100 \mum from Mamasın Dam Lake in November 2012 and were left to dry at room temperature in the laboratory.
