Pluronic Pf68 Increases Transfection Efficiency In Electroporationof Mesenchymal Stem Cells, Safa Aydin, Mehmet Emi̇r Yalvaç, Ferruh Özcan, Fi̇kretti̇n Şahi̇n

Electroporation is a safe and efficient method to transfect human tooth germ stem cells (hTGSCs), which are mainly mesenchymal stem cells (MSCs). In this study, pluronic F68 (PF68), a nonionic and low-foaming surfactant, was used for the first time to increase the transfection efficiency in electroporation of hTGSCs. Our results revealed that when PF68 was used in the preincubation, electroporation, and recovery medium, it dramatically increased transfection efficiency. Moreover, use of PF68 did not alter the differentiation capacity of hTGSCs, whereas cells electroporated without PF68 had significantly lower expression of Oct-4, pluripotency marker gene, and alkaline phosphatase (ALP) activity upon …

The aim of this research was to investigate the effects of the methanol extract obtained from Lycium barbarum fruits on anxiety, depression-like behaviors, and spatial memory in Wistar albino rats. A total of 28 rats were selected, randomly assigned to four experimental groups, and tested by means of the open field and elevated plus maze tests for anxiety-like behaviors, the forced swim test for depression-like behaviors, and the Morris water maze test for spatial memory. Lipoxygenase (linoleate:oxygen oxidoreductase; EC 1.13.11.12; LOX) is a key enzyme in the signaling pathway leading to the production of biologically active compounds involved in plant growth development and stress responses. In this study the novel LOX enzyme from opium poppy (Papaver somniferum L.) seedlings was purified to electrophoretic homogeneity and characterized. LOX activity during the germination of opium poppy seeds was analyzed, and the highest LOX activity was determined on the fourth day of germination. The present study investigated the effect of different salts on cotton shoot regeneration and transformation efficiency. Two-day-old germinating embryos of a local cotton cultivar (SG-125) were pretreated with 50 mM each of NaCl, CaCl2, and KCl for 60 min. The embryo explants were transformed by cocultivation with Agrobacterium tumefaciens strain LBA 4404 harboring a binary plasmid pTF101.1 that carried the insecticidal gene (cry1Ac) under control of wound-inducible promoter (AoPR1) and bilanafos acetyl reductase (bar) gene for plant selection. During the process of industrial production of biofuels and biochemicals from lignocellulosic biomass, large amounts of waste byproducts rich in xylose are generated, resulting in excessive wastage of natural resources and environmental pollution. In this work, xylose solution from corncob hydrolysate was utilized to produce 2,3-butanediol (2,3-BD), using the strain Enterobacter cloacae CICC 10011 to improve the utilization rate of hemicellulose and reduce environmental pollution. 2,3-BD fermentation conditions were subsequently developed. We investigated the implication of AMP-activated protein kinase (AMPK) in melatonin-induced protection in a renal ischemia/reperfusion (I/R) model. Animals were divided into four groups: rats of the sham group were not subjected to renal I/R. Rats in the I/R group received the vehicle 30 min before renal ischemia for 1 h followed by reperfusion for 6 h. Rats in the Mel+I/R group received melatonin (10 mg/kg) 30 min before the onset of I/R. Salinity is among the significant abiotic stresses adversely affecting plant development and yield for a wide range of crops. For quantitative and qualitative expression studies of proton gradient regulator 5 (PGR5) under salt stress, differential display RT-PCR and rapid amplification of cDNA ends (RACE) were performed on salt-stress responsive Gossypium arboreum. Alignment of genomic and cDNA sequences revealed that the GPGR5 gene comprises a single open reading frame of 96 amino acids and contains no introns. The present study addressed the successful in vitro seed germination, bulblet regeneration, increased bulblet size, hardening, and acclimatization of Persian lily (Fritillaria persica). Seed germination rate was recorded as 86.7% and 96.7% after 2 and 3 months of cold treatment at 4 °C, respectively. Bulblet explants taken from a germination experiment were cultured on Murashige and Skoog (MS) medium supplemented with 0.5-2.0 mg/L thidiazuron (TDZ). This study examined the effect of yohimbine alone and in combination with glibenclamide on ischemia and reperfusion-induced arrhythmias in diabetes. Six minutes of ischemia were produced in 1-week diabetic rats by ligation of the left coronary artery, and 6 min of reperfusion were produced by releasing the artery. A dose of 5 mg/kg of yohimbine and glibenclamide was administered for 7 days prior to the coronary ligation. Sox6, a member of the Sox (Sry-related HMG box) family of transcription factors, plays an important role in embryonic muscle development and adult fast myofiber maintenance. Here we report the expression and purification of a His-tagged version of recombinant porcine Sox6 (pSox6) in Escherichia coli. The recombinant pSox6 was expressed as a C-terminal fusion protein with His6 tag in the E. Coli Rosetta (DE3) host strain. Findings in the last decade suggest that there is a considerable amount of cardiomyocyte turnover in the human heart throughout life, albeit not sufficient for heart regeneration following myocardial infarctions. Only a few species are known to be remarkably efficient in cardiac regeneration. They restore lost cardiomyocytes via a process of cardiomyocyte dedifferentiation, which is followed by robust proliferation of cardiomyocytes and incorporation into the myocardium. Cell transplantation through direct cell injection is not an optimal method due to the lack of cell-extracellular matrix interactions. As a novel approach in skin tissue engineering, gold nanoparticles (AuNPs) were synthesized and incorporated at different concentrations into collagen/PEO nanofibrous matrices in this study. The group containing 14.27 ppm AuNPs (CM-Au) had the best nanofibrous morphology. CM-Au was cross-linked with glutaraldehyde vapor (CM-AuX). All groups were disrupted in collagenase in 2 h, but cross-linked groups and Matriderm1397904493 resisted hydrolytic degradation for 7 and 14 days, respectively. Due to its small pores and dense structure, lower water swelling results (7.26 ± 2.62 g/g) were obtained for CM-AuX than Matriderm (17.51 ± 1.97 g/g). Wound healing is an orderly process driven by numerous cellular mediators that aim to accelerate it. In skin conditions such as leprosy and psoriasis, Centella asiatica, which contains high amounts of triterpenoid saponins (TSs), has been reported to have beneficial effects. Mesenchymal stromal cells (MSCs) have been characterized as an important component of the hematopoietic niche that plays a role in regulating hematopoietic stem cell (HSC) quiescence, self-renewal, and lineage commitment. Macrophages, one kind of innate immune cells, are mainly developed from HSCs in niche. However, how MSCs and their subpopulations regulate macrophage commitment is still unknown. The current study compared the contribution of MSCs and Sca-1+MSCs to macrophage commitment and further modulation on the function of macrophages. We aimed to determine whether increased levels of asymmetric dimethylarginine (ADMA) directly cause hepatic deterioration and whether it is decreased by ascorbic acid (AA) as an antioxidant. The study included three groups: Control (n = 10), ADMA (n = 10), and ADMA + AA (n = 10). ADMA was administered at 2 mg kg-1 day-1 and AA was given at 50 mg kg-1 day-1 for 10 days. Polyhydroxybutyrate (PHB) is a polymer used to restore tissues or regenerate bones. However, its compatibility with human bone marrow-derived mesenchymal stem cells (MSCs) has not been examined. PHB membranes of random (r-PHB) or aligned (a-PHB) electrospun nanofibers that generate bone and spinal axon scaffolds were combined with human MSCs. The adhesion and proliferation of cells on these membranes were examined. The orientation of cells on PHB membranes was analyzed by confocal microscopy and scanning electron microscopy (SEM). Severe graft-versus-host disease (GvHD) is a life-threatening complication after allogeneic hematopoietic stem cell transplantation (HSCT). Bone marrow mesenchymal stem cells (BM-MSCs) display immunosuppressive activity and have been previously shown to improve the outcome of GvHD therapy. We describe the use of umbilical cord mesenchymal stem cells (WJ-MSCs) expanded in serum-free medium for the treatment of acute GvHD (aGvHD) and chronic GvHD (cGvHD) in 10 patients.
