Simvastatin Combined With Bone Marrow Stromal Cells Treatment Activatesastrocytes To Ameliorate Neurological Function After Ischemic Stroke In Rats, Yonghua Zhao, Qian Zhang, Zhenwei Chen, Naiwei Liu, Chienchih Ke, Youhua Xu, Weikang Wu
Turkish Journal of Biology
Evidence indicates that reactive astrocytes play an important role in neurological functional amelioration after stroke. Bone marrow stromal cell (BMSC) transplantation-promoted astrocyte-derived neurotrophic factors' expressions resulted in decreasing the number of apoptotic neurons and improving neurogenesis and angiogenesis in the ischemic boundary zone. Recent studies found that simvastatin had neuroprotective activity in acute stroke. However, it is still ambiguous as to whether simvastatin combined with BMSCs can further activate astrocytes, which contribute to neurological functional recovery after ischemic stroke. In our present study, we established a middle cerebral artery occlusion model in rats and found that combination treatment of simvastatin …

Glycation of proteins is a nonenzymatic process in which proteins react with reducing sugar molecules. This process takes place at ?-amino (?-NH+) groups of lysine or hydroxylysine residues as well as α-amino groups of N-terminal residues. In the present study glycation of ?-NH+ groups of lysines was computationally analyzed for 26 proteins based on their 3D structures. We determined the spatial relationship with acidic or basic residues and correlated them with the glycation prediction algorithm Netglycate 1.0 software, which employed primary structure exclusively for glycation site prediction. Of the lysines from 19 of the 20 proteins employed to build the …

The recent progress in DNA sequencing and computational algorithms dramatically heightened the value of single nucleotide polymorphism (SNP) databases. The experimental studies of mutation for the particular gene of interest are laborious and time-consuming. In this study, we used several computational algorithms to investigate the structural and functional consequences of SNPs on the glyceraldehyde 3-phosphate dehydrogenase (GAPDH) gene of zebrafish (Danio rerio). The GAPDH gene is involved in multiple cellular activities, in addition to its association with classical glycolytic pathway. It has most commonly been used as the housekeeping gene for differential gene expression profiling, including western blots. The computational …

PCR is an effective and widely used method for the amplification of target DNA fragments in vitro. The templates that have high guanine-cytosine (GC) content, complex secondary structures, or short tandem repeats are difficult to amplify by the conventional PCR methods. Human genome regulatory regions such as promoters are rich in terms of GC base compositions. To amplify these regions some special modifications or additives are required. Using different PCR strategies such as hot-start/touchdown PCR or the utilization of various additives into the reaction mixture such as organic molecules can improve PCR yield. In the present study we optimized the …

Production Of Recombinant Human Dipeptidyl Peptidase Iv From Sf9cells In Microbial Fermenters, Özlem Ayteki̇n, Sai̇me İsmet Gürhan, Kayoko Ohura, Teruko Imai, Gaye Öngen
Turkish Journal of Biology
The human dipeptidyl peptidase IV (hDPPIV/CD26) is expressed as an immune response in some cancer cells as well as intestine and incretin metabolism, and deficiency of the enzyme leads to metabolic disorders. In the present study, recombinant hDPPIV/CD26 genes were expressed in baculovirus-insect cell systems in a 5-L stirred-tank fermenter. Because of the shear sensitivity of the insect cell line, production from insect cells should be performed in new-generation type bioreactors, which are commonly more expensive than microbial fermenters. To optimize the process, hydrodynamic parameters and oxygen consumption of Sf9 cells at 1.5 L and 3 L were monitored, and …

Turkish Journal of Biology
Crimean-Congo hemorrhagic fever virus (CCHFV) is a deadly tick-borne virus with high mortality rates. Current antivirals lack specificity, making them susceptible to off-target effects and cytotoxicity. There is an utmost need for the identification of active compounds for anti-CCHFV therapies. Inhibition of CCHFV ovarian tumor (OTU) protease by small molecules is an exciting potential antiviral therapy. In this study, computational approaches based on residue homology, the binding coordinates of ligands, and correlation analysis with in vitro data identified the pocket of Y89-W99 as the inhibition site of CCHFV OTU protease. In silico screening of more than 600,000 compounds against this …

Identification Of Root-Knot Nematodes (Meloidogyne Spp.) Fromgreenhouses In The Middle Black Sea Region Of Turkey, Gökhan Aydinli, Sevi̇lhan Mennan
Turkish Journal of Zoology
Root-knot nematodes are among the main important pests causing serious yield losses in greenhouses worldwide, including Turkey. Accurate identification of these pests is essential for determining appropriate control strategies. Identification of Meloidogyne species based on perineal pattern morphology alone is not viable or reliable due to overlaps and high variability among the species. Therefore, our objective was to overcome the difficulties in identifying 90 populations of root-knot nematodes collected from different greenhouses in the Middle Black Sea Region (Samsun, Tokat, Amasya, Çorum, Ordu, and Sinop provinces), using not only morphological (perineal pattern morphology) but also biochemical (esterase phenotype) and molecular …

A 70-day feeding trial was conducted to evaluate the effects of dietary supplementation of selenium nanoparticles (Se-NPs) on physiological and biochemical aspects of juvenile mahseer (Tor putitora) fish. Maintaining a 40% protein level, 2 different experimental diets supplemented with 0% Se-NPs and 0.68 Se-NPs mg kg-1 were fed to triplicate groups of fish. The experiment was conducted in a semistatic condition consisting of 6 fiber tanks. The fish were first acclimatized, and then 30 fish per tank were randomly put into each fiber tank. At the end of the feeding trial, samples were collected from the fish and analyzed. Results …

Mandibular Morphology And Dietary Preferences In Two Pygmy Molecrickets Of The Genus Xya (Orthoptera: Tridactylidae), Katerina Kuravova, Petr Kocarek
Turkish Journal of Zoology
Pygmy mole crickets (Tridactylidae) are an interesting but little-known group of Orthoptera; their food biology and dietary preferences have not been adequately studied. Our study focused on the diet composition and associated functional morphology of the mandibles in two European species: Xya pfaendleri Harz, 1970 and X. variegata (Latreille, 1809). Based on postmortem gut content analyses, the studied species feed mainly on detritus (decaying organic particles), representing 91% and 97% of the diet in X. pfaendleri and X. variegata, respectively. The mandibular structures are associated with the processing of such food. The incisor area is equipped with three robust and …
