Although research on Autism Spectrum Disorder (ASD) continues to increase, there is not yet a “cure” for the disorder. There is a lack of consensus regarding most effective treatments, much less an accepted treatment plan or course. Families may feel alone and lost in the treatment decision process. In order to best assist caregivers, psychologists must understand the selections caregivers make, their sources of information, and what influences them in their decision-making process. The objective of this dissertation is to understand the functions of Escherichia coli YqjA and YghB, which are members of the conserved DedA/Tvp38 membrane protein family. YqjA and YghB are inner membrane (IM) proteins with multiple predicted membrane–spanning domain, sharing 62% amino acid identity and partly overlapping functions. Simultaneous in-frame deletions of these two genes in a strain named BC202 results in various phenotypes including cell division defects, temperature sensitivity, and sensitivity to drugs and alkaline pH. The cell division defect of BC202 is due to the inefficient secretion of periplasmic amidases by the twin arginine transport (Tat) pathway into …

Hybrid core-shell nanoparticles have attracted attention due to their unique characteristics which combine properties of the inorganic core and organic shell in a way that generates new properties which do not exist for the two individual parts. This makes them promising candidates for the design of stimuli-responsive materials, chemo- and biosensors, and various biomedical applications. In this dissertation research, we designed and prepared a series of hybrid environmentally responsive nanoparticles where fluorescent block copolymers, including various combinations of polythiophene (PT), poly(p-phenylene) (PPP), poly(3-hexylthiophene) (P3HT) and polyallene (PA), were grafted on the surface of inorganic nanoparticles using surface-confined Kumada catalyst-transfer polymerization. …

Disturbances are a common occurrence in coastal ecosystems and can provide opportunity for adaptation and renewal in healthy systems; hurricanes bring mineral accretion to a marsh, floods provide a pulse of freshwater and nutrients to estuaries, and fires increase species diversity and abundance in forests. Humans, however, have depleted the resiliency of many coastal systems via top down and bottom up mechanisms, leaving these ecosystems more vulnerable to both natural and anthropogenic disturbances. This research explores virtual reality as a medium for live concert performance. I have realized compositions in which the individual performing on stage uses a VR head-mounted display complemented by other performance controllers to explore a composed virtual space. Movements and objects within the space are used to influence and control sound spatialization and diffusion, musical form, and sonic content. Audience members observe this in real-time, watching the performer's journey through the virtual space on a screen while listening to spatialized audio on loudspeakers variable in number and position. Use of pesticide has become part of modern day agricultural practice. Some pesticides can remain in the environment for decades and contaminate surface water that is used for irrigation of produce. Two studies were conducted- first to examine possible pesticide residue in surface water and some fruits, vegetables and cereals in Louisiana; and second was to alleviate possible pesticide residues in the water using zeolite filtration. How do many species live in a certain place? How does species composition changes among habitats? And what mechanisms decide species distribution? These are fundamental questions in community ecology. I first investigated ant diversity in two coastal ecosystems (dunes and wetlands) in the northern Gulf of Mexico, and then used the distribution patterns to infer assembly processes that structure ant communities in coastal areas. Specifically, the following hypotheses are tested: (1) coastal systems support lower ant diversity due to the unsuitable environment; (2) species living near the seashore are a subset of those exist near inland; (3) deterministic processes are …

Rickettsia parkeri is an emerging human pathogen and spotted fever group Rickettsia that is transmitted via Amblyomma maculatum (the Gulf Coast tick) in the United States. Since these ticks must feed for several days in order to molt to the next life cycle, they must be able to counteract the host immune response. Despite this fact, there have been few studies that evaluate the immunomodulatory effect of this vector and the resultant influence on rickettsial disease. The Mexican rice borer, Eoreuma loftini (Dyar) (Lepidoptera: Crambidae), is an invasive pest of sugarcane, Saccharum spp.; rice, Oryza sativa; and other graminaceous crops along the U.S. Gulf Coast. Identification of E. loftini sex pheromones led to the development of pheromone baited traps. Studies were conducted to determine strategies for the use of E. loftini pheromone traps in invasive species monitoring and pest management. A two-year field study demonstrated that E. loftini pheromone traps attract males from distances of up to 100m. A behavioral assay observed that detection of the pheromone by E. loftini males occurs at ≈48m from the …

Species interactions play a prominent role in the establishment and spread of many invasive species. However, rarely are invasions studied in more than a direct pairwise species context, or with consideration to how species interactions can vary biogeographically. I used field surveys combined with common garden and greenhouse experiments to investigate how multitrophic above- and belowground interactions influence plant invasions at large spatial scales. I focused on comparisons between sympatric native and invasive lineages of Phragmites australis, a wetland grass distributed throughout North America. End rot development in sweetpotato is caused by several pathogens and exacerbated by unfavorable environments. A critical need exists to know what factors in the environment trigger end rot development and how to manage the crop to minimize the incidence of end rots which are the overall objectives of this study. This research was divided into three studies: 1) Effect of environmental factors on expression of end rot in sweetpotato roots. Spotted fever group (SFG) Rickettsia are primarily associated with their reservoir host and vector, the tick. Rickettsial colonization and maintenance within the arthropod is a key component of vector competence and pathogen transmission to the mammalian host. Contemporary detection of novel tick hosts for rickettsial species, combined with an unprecedented rise in human cases of SFG rickettsiosis, necessitates a deeper understanding of tick/Rickettsia interactions. Cardiovascular function is under the influence of autonomic nervous system, both of which can be assessed non-invasively. The purpose of this dissertation was to examine these non-invasive markers of cardiovascular and autonomic function and their relationships with exercise training, glucose ingestion and hydration status. A series of three studies were conducted to gain insight to various influences on cardiovascular and autonomic function. The first study examined the influence of exercise training of brachial artery flow-mediated dilation (BAFMD) using meta-analytic techniques. The development of biocompatible polymers using thiol-acrylate Michael addition reactions between not only acrylates and thiols but also between amines and acrylates were demonstrated. The synthesis of an in situ tertiary amine catalyst initiates the propagation steps of the polymerization process by subsequently reacting with trimethylolpropane tris (3-mercaptopropionate) (TMPTMP). A few notable characteristics of this reaction are tunable gel times, the capability of reaching high polymer conversion and suitable mechanical strength. Common bottlenose dolphins (Tursiops truncatus; dolphins) inhabiting Mississippi Sound (MSS) in the north-central Gulf of Mexico (GMx) are considered a part of a single stock of dolphins that includes Bay Boudreau and Lake Borgne by the National Marine Fisheries Service (NMFS). MSS is bounded by the mainland (north) and several barrier islands (south). Dolphins inhabiting coastal waters directly south of the barrier islands constitute part of the Northern Coastal Stock. Abundance in MSS has been reported to fluctuate seasonally, with higher abundances of dolphins estimated in summer versus winter. Purpose: To determine if bolus electron conformal therapy (BECT) combined with volumetric modulated arc therapy (VMAT) and flattening filter free volumetric modulated arc therapy (FFFVMAT (6x and 10x)) can maintain equal or better dose coverage than standard volumetric modulated arc therapy (VMAT) while reducing doses to organs at risk (OARs). Methods: BECT+VMAT, FFFVMAT (6x and 10x), and VMAT treatment plans were produced for ten post-mastectomy radiotherapy (PMRT) patients previously treated at our clinic. Approaches to prepare spatially selective surfaces were developed in this dissertation for constructing nanopatterns of organic thin film materials. Nanoscale surface patterns were prepared using immersion particle lithography and scanning probe lithography combined with organothiol chemistry. Organic thin films and nanomaterials can be patterned with tunable periodicities and designed shapes by selecting the diameter of mesospheres used as surface masks or scanning probe lithography, respectively. The surface platforms of well-defined nanopatterns are ideal for high resolution investigations using scanning probe microscopy (SPM). Approximately 2,300 petroleum platforms are currently operating in the Gulf of Mexico (GOM), and are estimated to provide an additional 4 to 12 km2 of artificial reef habitat. The ecological role of platforms has often been overlooked and little is known about the effect of artificial light from the active platforms on surrounding fish communities. Tar is inevitably produced from biomass thermochemical processes, and is often disposed as an industrial waste, leading to environmental pollution. As a result of its high carbon content, tar was expected to be a promising precursor for manufacturing carbon materials. Consequently, low-cost porous carbon nanofibers (CNFs) using tar, polyacrylonitrile (PAN), and silver nanoparticles was fabricated through electrospinning and subsequent stabilization and carbonization processes. The continuous electrospun nanofibers were obtained with diameters ranging from 392 to 903 nm. The addition of biomass tar resulted in increased fiber diameters, reduced thermal stabilities, and slowed cyclization reactions of PAN in the as-spun nanofibers. …

Genomic imprinting is a mechanism that targets epigenetic modifications to regulate gene transcription to express a gene from only one of its two parental alleles. Imprinted genes are typically clustered together and are involved in developmental regulation of the fetus. The paternally expressed gene 3 (Peg3) domain represents one such imprinted gene cluster involved in fetal growth regulation and maternal caring behavior. The DNA of eukaryotic cells does not exist in free linear strands; it is tightly packaged and wrapped around nuclear proteins in order to be accommodated it inside the nucleus. The basal repeating unit of chromatin, termed the nucleosome, provides the first level of compaction of DNA into the nucleus. Nucleosomes are interconnected by linker DNA and associated linker histones to form 30 nm fibers. Lignocellulosic biomass contains cellulose and hemicellulose which are composed of hexose and pentose sugars. These sugars can be used in the sustainable production of fuels and chemicals. However, the recalcitrant nature of lignocellulosic biomass makes this conversion a challenging process. An effective pretreatment can remove lignin, solubilize the hemicellulose, decrease cellulose crystallinity, and prepare the biomass for enzymatic hydrolysis and conversion into green renewable chemicals. Development of resistant cultivars is the main control measure against sugarcane brown rust caused by Puccinia melanocephala. Durability is uncertain, since the pathogen possesses adaptive ability to overcome host plant resistance. A differential gene expression study utilizing suppressive subtraction hybridization was conducted to improve understanding of brown rust resistance mechanisms in sugarcane. In view of its mountainous terrain, peninsular shape, and status as an agglomeration of formerly separate landmasses, the island of Sulawesi, Indonesia, presents a fertile opportunity for examining biogeographical processes. Previous work on Sulawesi primates and amphibians suggested the existence of distinct areas of endemism (AoEs), but the relevance of these zones to a broader set of taxa has not been widely investigated, nor the reasons for their existence fully explored. Here, we use population genetic analyses of the endemic Sulawesi shrew, Crocidura elongata, to assess biogeographical partitioning according to the putative AoEs.
