The Influence Of Processing On The Chemical And Electrophoretic Patterns Of Water-Soluble And Salt-Soluble Proteins In Blue Crabs., Orthnell George Dowdie

Raw and processed Blue crab meat were investigated to determine the influence of processing temperature on the distribution of water soluble and salt soluble proteins, using sodium dodecyl sulfate polyacrylamide gel electrophoresis in conjunction with photodensitometry. Analysis of the raw crabmeat protein showed 30 water soluble and 45 salt soluble protein subunits. Salt soluble proteins were composed of subunits whose molecular weights were much more variable than the water soluble protein subunits. Salt soluble proteins were found to have molecular weights of 12,000-160,000 daltons, whereas the water soluble subunits were of molecular weights 12,000-130,000 daltons. There were significant differences (P < 0.001) in the changes between the number of bands present in the raw crabmeat and the number retained after processing at 100(DEGREES)C; however, the level of significance was only changed to (P < 0.05) when raw crabmeat was compared to crabmeat processed at 121(DEGREES)C in steam. Salt soluble proteins were more heat labile than the water soluble proteins. At 70(DEGREES)C, the total level of salt soluble protein was reduced 57% compared to the levels in raw meat, whereas, the water soluble proteins were reduced only 13%. Processing temperatures were found to significantly influence (P < 0.05) the pH of the processed meat as well as the water in which the crabs were processed. Correlation coefficients of r = 0.7604 and r = -0.6401 were found between changes in pH and basic and acidic groups. These coefficients were significant at (P < 0.05). Analysis of Blue crab hemolymph for copper revealed the copper content to be approximately eight times greater than quantities found in the meat. Nine proteins were found in the hemolymph with 3 of the proteins being complexed with copper (hemocyanins). The hemocyanins had molecular weights of 58,000, 71,000 and 95,000 daltons. The subunit with a molecular weight of 58,000 daltons contained approximately 65% of the total copper in the hemolymph.

It has been a consistent epidemiological finding that women are more prone to develop symptoms of depression than men. The present investigation examines women's greater vulnerability to depression in terms of Beck's cognitive theory of depression. Sixty women were divided into a depressed group and a non-depressed group on the basis of criteria scores on Scale 2 of the MMPI and on the Beck Depression Inventory. The groups did not differ significantly in age, education or employment status. Rhodopseudomonas viridis grows by means of nitrogen fixation under anaerobic, photosynthetic conditions. In batch culture, nitrogenase activity was highest at early-logarithmic phase, lower during mid- to late-logarithmic phase, and nearly zero during stationary phase. Nitrogen-fixing cells were morphologically and ultrastructurally similar to non-nitrogen-fixing cells as determined by electron microscopy. Electron spin resonance (esr) spectroscopy of nitrogen-fixing whole cells yielded g4.26 and g3.66 signals indicating the presence of nitrogenase molybdenum-iron (MoFe) protein. Ammonia switch-off occurred upon addition of 0.2 mM NH(,4)Cl, however, nitrogenase activity did not reappear for nearly four hours. Two hundred raw milk samples were collected and stored in sanitized plastic containers which were transported in ice to the laboratory. Three 100 ml sub-samples were aseptically removed from each container. One sub-sample (Fresh) was analyzed for Standard Plate Count (SPC) and adenosine-5'-triphosphate (ATP) content. The two remaining sub-samples underwent preliminary incubation; one sub-sample was incubated at 12.8(DEGREES)C for 18 h (PI 12.8) and the other at 7.2(DEGREES)C for 48 h (PI 7.2). Following incubation, SPC and ATP content were determined. Correlation of ATP content with SPC-Fresh, SPC-PI 12.8 and SPC-PI 7.2 was 0.72, 0.84 and 0.89 with log(,10) transformed …

Epibenthic (trawlable) macrofauna and hydrographic variables were sampled at ten stations in the Calcasieu estuary, Louisiana, from June, 1974, through February, 1976. The 100 otter trawl samples collected contained 32,221 individuals belonging to 63 invertebrate and 68 fish species. The 10 most abundant fish and invertebrate species represented 95.3 and 90.0%, respectively, of the total number of individuals collected. Seasonal variation in numbers of species and individuals were similar, with significant increases in number of species during spring and early summer months. A microbial and enzymatic assessment protocol for determining the environmental effect and fate of these manufactured chemicals in coastal wetlands was devised. A method was developed for the direct determination of mercury in water and biological samples using a unique carbon bed atomizer for atomic absorption spectroscopy. The method avoided sources of error such as loss of volatile mercury during sample digestion and contamination of samples through added reagents by eliminating sample pretreatment steps. The design of the atomizer allowed use of the 184.9 nm mercury resonance line in the vacuum ultraviolet region, which increased sensitivity over the commonly used spin-forbidden 253.7 nm line. The shortage of training programs available to CES in energy education makes it difficult to properly prepare agents for energy conservation work. A canine rotavirus isolate caused clinical disease, small intestinal lesions, serum antibody response, and shedding of rotavirus in the feces in newborn gnotobiotic dogs. The results of this study demonstrated this rotavirus was a pathogen in neonatal gnotobiotic dogs. The pathogenesis of the intestinal lesions was similar to the pathogenesis of rotaviral enteritis in other species. In three consecutive experiments, twenty puppies were derived by caesarean section and transferred to sterile isolators. The effects of sensitivity of heart rate feedback on bidirectional heart rate control were examined by comparing two groups of 10 subjects who received feedback signals of either high or low sensitivity. Subjects in the high sensitivity condition received a feedback signal in which 1 cm of feedback meter needle movement was equivalent to an 8.57 beat-per-minute (BPM) change in momentary heart rate. The low sensitivity condition received a feedback signal which was one-half as sensitive, i.e. 1cm = 17.14 BPM change. The work presented here is based on the Zimm-Bragg model for the helix-coil transition in polypeptides. Biologically active peptides discussed include: (beta)-endorphin, (beta)-lipotropin, glucagon, secretin, vasoactive intestinal peptide, and the binding or B subunit of cholera toxin. Random copolypeptides studied include copolymers of (omega)-hydroxyethyl and (omega)-hydroxybutyl-L-glutaminyl residues, and ionic copolypeptides containing L-glutamyl and either L-alanyl or L-tyrosyl residues. Residues 17-29 of (beta)-endorphin are expected from calculation to be alpha helical in the presence of anionic lipids. This portion of the chain forms an amphipathic helix which would be expected to interact favorably with a membrane containing anionic lipids. The order …

Eighteen male subjects participated in an investigation to examine the effects of static, dynamic, and combination work (static and dynamic) on the variables heart rate, systolic and diastolic blood pressure, pressure rate product, and maximal oxygen consumption (VO(,2)(' )max). The subjects, who were all free from any cardiovascular problems, were divided into 3 groups based on their maximum oxygen consumption levels. Microbial production of methane from alkaline sweet potato wastes was studied. Assessment of methane production potential was based on total COD of the wastes. A single-stage and a two-stage system were studied. In both systems, to ensure stable operation and high performance, methane fermenters had to be initially seeded with large quantities of methane formers. A 50% inoculum (based on total fermenter volume) was found to be most effective. Methane formers tended to aggregate to form spherical particles which had extremely high settling rates, this eliminated the requirement of cell recycle. The achievement of the goals of General Systems Theory are, in part, dependent on the development and use of a language which will facilitate relevant communication across disciplines so that structural similarities which are hidden by the use of specialists' languages can be examined. This study explores various dimensions of the language associated with GST. The major vocabulary components associated with GST were identified using four criterion. Sperm incorporation in Lytechinus pictus and Strongylocentrotus purpuratus eggs incubated (10 min) and fertilized in the presence of 2 x 10('-6) M cytochalasin B (CB) and 1 and 10 mM ammonium chloride (NH(,4)Cl) occurs in the presence of limited microvilli and microfilament organization and cortical exocytosis. CB inhibition on sperm incorporation is time related and continues to occur long after sperm-egg fusion. In S. purpuratus, at least 30% incorporation is observed in all CB treated eggs. Sperm do not incorporate until 2.5 min after insemination. Maximum incorporation occurs between 7 to 10 min. Percent incorporation approached 60% when eggs were …

The peripheral electrophysiological manifestations of levels of cognitive processing and memory performance were investigated by recording heart rate, skin conductance, skin temperature and electromyogram measures during a three phase verbal task. Subjects processed words at three cognitive levels (phonetic, low semantic, high semantic), and physiological recordings were made during cue covert processing and verbalization phases. Three colored lights were used to cue subjects to the appropriate processing level for each word. An incidental memory task was given following the processing tasks. In laboratory studies, 24 hr LC(,50) values obtained for Ambush('(REGTM)) and Pydrin('(REGTM)) on 18-27 mm juvenile red swamp crawfish, Procambarus clarkii (Girard), in clear water were 0.49 and 0.37 ppb, respectively. LC(,50) values derived for 10-20 mm channel catfish fry, Ictaluras punctatus Rafinesque, were 1.81 and 1.83 ppb, respectively for Ambush('(REGTM)) and Pydrin('(REGTM)). The addition of alluvial soils to test waters had no apparent effect on toxicity to crawfish but did reduce toxicity to channel catfish. The studies presented in this work relate to conformational studies of polymers and biopolymers. Polyethylene is used as a model chain to develop a general formalism to characterize helical molecules with varying degrees of flexibility. The average projection of a unit vector along the last bond onto a unit vector along the first bond is the principal quantity calculated for modeling purposes. This thesis describes the late Pleistocene (Rancholabrean) vertebrate paleontology of Louisiana through the use of vertebrates as paleoenvironmental indicators. This method of interpretation is established through morphological description and evaluation, taphonomy, systematic paleontology, and in some cases, the use of modern analogs.
This study compares and contrasts Louisiana with other southeastern states during the late Pleistocene. This comparison shows that Louisiana did not vary significantly from the rest of the southeastern United States during the late Rancholabrean. This work also shows that the most common habitats of Louisiana at this time were open temperate woods, grasslands, and marshes. Geographically, most …

The present study was an experimental investigation of the effects of a sex education workshop in an adolescent in-patient psychiatric population. Eleven adolescent patients (seven males, four females) participated in the eight session sex education workshop (SEW). The specific activities of phosphofructokinase, aldolase, and pyruvate kinase was diminished in crude extracts from skeletal muscle of streptozotocin diabetic rats, whereas the activities of glucosephosphate isomerase and phosphoglucomutase were not changed. The decreased phosphofructokinase activity was shown to be independent of allosteric control, and it could be reversed by insulin treatment. A rapid and mild method for the purification of phosphofructokinase from rat skeletal muscle were developed. The partially purified enzyme from muscle of normal and streptozotocin diabetic rats showed identical Michaelis constants for ATP and equal sensitivity to inhibition by excess of ATP. With autoradiography and fluorescent stain techniques, the degree of ethylation of Drosophila melanogaster sperm following treatment with (1-('3)H)-ethyl-methanesulfonate (EMS) or (2-('3)H)-diethylnitrosamine (DEN) was shown by different levels of silver grains on sperm. The degree of ethylation of sperm cells treated with the mutagen in different germ cell stages of development (sperm, spermatids, spermatocytes, or spermatogonia) was determined by varying the time of sampling. A marked improvement in health status for black adults took place over the last decade in the United States. Life expectancy for black men increased 4.6 years between 1968 and 1978, while for black women the increase was 5.7 years. Turpentine-induced inflammation was used as a model for the study of toxic neutrophils in the dog. Changes in the number of circulating toxic neutrophils paralleled changes in clinical signs, body temperature, total leukocyte count, and neutrophil count. Light microscopic appearance of neutrophils from turpentine-injected dogs was similar to toxic neutrophils seen clinically in dogs with severe bacterial or other inflammatory diseases. Ultrastructural studies showed that foamy vacuolation of the cytoplasm was due to irregular, electron-lucent areas which often contained membrane remnants and myelin figures. Increased cytoplasmic basophilia was due to the retention of rough endoplasmic reticulum and polyribosomes in mature …
