The Regulation Of Carbonic Anhydrase And Ribulose-1,5-Bisphosphate Carboxylase/Oxygenase Activase By Light And Co2 In Chlamydomonas Reinhardtii, Mamta Rawat, James V. Moroney

We have investigated the regulation of accumulation of ribulose1,5-bisphosphate carboxylase/oxygenase activase and the periplasmic carbonic anhydrase (CA) in Chlamydomonas reinhardtii. In algae, the periplasmic CA is required for efficient CO2 fixation when the CO2 concentration is low. These two proteins are affected differently by the CO2 level in the environment. The steady-state level of the ribulose-1,5-bisphosphate carboxylase/oxygenase activase transcript was only slightly and transiently affected by a reduction in ambient CO2 concentration, whereas the CA transcript level was strongly induced by air containing ambient (350 parts per million) CO2 (low CO2) conditions. Aerobic And Anaerobic Starvation Metabolism In Methanotrophic Bacteria, P. Roslev, G. M. King

The capacity for anaerobic metabolism of endogenous and selected exogenous substrates in carbon- and energy-starved methanotrophic bacteria was examined. The methanotrophic isolate strain WP 12 survived extended starvation under anoxic conditions while metabolizing 10-fold less endogenous substrate than did parallel cultures starved under oxic conditions. During aerobic starvation, the cell biomass decreased by 25% and protein and lipids were the preferred endogenous substrates. Aerobic protein degradation (24% of total protein) took place almost exclusively during the initial 24 h of starvation. Metabolized carbon was recovered mainly as CO2 during aerobic starvation. Wheat forage was harvested at an early head stage of maturity and ensiled in 12 900-kg experimental silos at three percentages of DM (20.8% for direct-cut forage and 27.9 or 39.3% for wilted forage) either with or without application of a lactic acid bacterial inoculant. The objective was to test the efficacy of the inoculant to alter silage fermentation, preservation, and nutritive value of wheat forage ensiled at different moisture percentages because of wilting. Wilting enhanced DM preservation and decreased fermentation end products. Inoculation made the fermentation more homolactic but did not enhance DM preservation. Silage rations (80% DM as …

Incidence of groundwater contamination by agrichemicals in the United States and elsewhere is of concern in terms of the health effects associated with chemical contaminants present in drinking water. The major objectives of this study were: (1) to quantify the mobility of atrazine and nitrates in a Sharkey clay soil in the presence of a shallow water table, and (2) to determine evidence of preferential flow patterns on the mobility of agricultural chemicals in such soils.

Native rat adipocytes and the mouse adipocyte cell line, 3T3-L1, possess transport vesicles of apparently uniform composition and size which translocate the tissue-specific glucose transporter isoform, GLUT4, from an intracellular pool to the cell surface in an insulin-sensitive fashion. Caveolin, the presumed structural protein of caveolae, has also been proposed to function in vesicular transport. Thus, we studied the expression and subcellular distribution of caveolin in adipocytes. We found that rat fat cells express the highest level of caveolin protein of any tissue studied, and caveolin is also expressed at high levels in cardiac muscle, another tissue possessing insulin responsive …

Calcium influx into cultured retinal amacrine cells is followed by a small, slow, inward current that we show here results from the operation of electrogenic Na-Ca exchange. The activity of the exchanger is shown to correlate with the magnitude of the Ca2+ load and to depend on both the Ca2+ and Na+ gradients. Li+ is unable to substitute for Na+ and in the absence of Na+, slow tail currents are almost entirely suppressed. A rapid change in [K+](o) does not affect the activity of the exchanger, suggesting that only Na+ and Ca2+ are transported. The ratio of charge entering as …

The Alu repetitive family of short interspersed elements (SINEs) in primates can be subdivided into distinct subfamilies by specific diagnostic nucleotide changes. The older subfamilies are generally very abundant, while the younger subfamilies have fewer copies. Some of the youngest Alu elements are absent in the orthologous loci of nonhuman primates, indicative of recent retroposition events, the primary mode of SINE evolution. PCR analysis of one young Alu subfamily (Sb2) member found in the low-density lipoprotein receptor gene apparently revealed the presence of this element in the green monkey, orangutan, gorilla, and chimpanzee genomes, as well as the human genome. …

The stringent environmental regulations, rising cost, public concerns, and the limited availability of landfills for disposal of organic wastes have stimulated efforts to identify cost-effective, environmentally sound, alternative waste disposal technologies. The value of organic wastes as soil amendments is limited by their low nutrient value, occasional strong odors and high costs of transportation. Flooding is a common natural stress occurring in wetland/bottomland forests. Submergence, total immersion, is a special case of flooding, which has received limited attention in the literature. This study provides information on the impacts of three submergence treatments on three bottomland tree species. The results indicated that submergence would inhibit shoot and root growth for the three tested species during the submergence period. However, the survival of these species under submergence conditions were different. Taxodium distichum (L.) Rich survived even after 30 days of water coverage and survival continued to be high during the recovery period. Quercus nuttallii Palmer was …

Role Of Erythroid Kruppel-Like Factor In Human Γ- To Β-Globin Gene Switching, D. Donze, T. M. Townes, J. J. Bieker

Erythroid Kruppel-like factor (EKLF) is an erythroid-specific transcription factor that contains zinc finger domains similar to the Kruppel protein of Drosophila melanogaster. Previous studies demonstrated that EKLF binds to the CACCC box in the human β-globin gene promoter and activates transcription. CACCC box mutations that cause severe β-thalassemias in humans inhibit EKLF binding. Results described in this paper suggest that EKLF functions predominately in adult erythroid tissue. The EKLF gene is expressed at a 3-fold higher level in adult erythroid tissue than in fetal erythroid tissue, and the EKLF protein binds to the human β-globin promoter 8-fold more efficiently than …

Several methods of separating the dissolved 4+ and 6+ oxidation states of selenium were investigated. The use of Dowex 1 anion exchange resin in packed columns was not satisfactory as quantitative separation was not obtained. The determination of selenite after formation of a fluorescent complex with 2,4-diaminonaphthalene was quantitative but the procedure was too involved for routine work. Separation was achieved by co-precipitating the selenite from solution with ferric hydroxide precipitate formed between pH 5 and 6. The co-precipitation technique is effective from 4 to 50 degrees centigrade and is unaffected by salinity although phosphates interfere at high concentrations. The …

Three experiments were conducted to evaluate the response of freshwater-oligohaline marsh macrophytes to increased water depth and salinity. In the first experiment, marsh sods containing Sagittaria lancifolia were manipulated in the field to decrease elevation (i.e., increase water depth) by 7.5 and 15 cm. Marsh sods at 15 cm below the marsh surface had the lowest redox potential and highest interstitial water sulfide concentration. Plants in the 15-cm water depth treatment had higher mean and maximum leaf heights than disturbed control plants, but aboveground biomass was not affected. Maize genotype differences in aflatoxin production were observed when kernels of resistant GT-MAS:gk and thirteen susceptible commercial hybrids were inoculated with Aspergillus flavus using a laboratory kernel screening assay. GT-MAS:gk supported the lowest levels of aflatoxin in both intact and endosperm-wounded kernels. Treating intact kernels with KOH effected substantial increases in aflatoxin accumulation in GT-MAS:gk, but only marginal increases in Pioneer 3154. Removing wax from the surface of GT-MAS:gk kernels greatly increased aflatoxin accumulation. These results indicated that GT-MAS:gk resistance was associated with an intact pericarp (wax and cutin layers), acting as a physical barrier, along with internal biochemical factors. …

To study the physiological and biochemical effects of pollen sources on fruit growth and development of southern highbush blueberry (Vaccinium corymbosum hybrids), five-year old 'Sharpblue' plants were grown in a greenhouse for self- and cross-pollination experiments. Cross-pollination with 'Gulfcoast' and 'O'Neal' as pollen sources increased fruit weight by 58.2% and 54.9%, respectively, compared to self-pollination. Coincidentally, cross-pollination doubled the number of mature ovules, and increased their size by 14%. Ovule abortion occurred between 5 and 10 days after pollination (DAP) in both pollination treatments. More abortion occurred in self-pollination (35%), compared with cross-pollination (22% for 'Sharpblue' $\times$ 'O'Neal' and 29% …

A plant based surfactant extracted from fruit pericarps of Sapindus mukurossi (Ritha) is employed for the first time to investigate remediation of contaminated soils. A method for preparing natural surfactant solutions was developed and the solutions were characterized followed by generation and characterization of colloidal gas aphron (CGA) suspensions produced with these solutions. The variation of hexachlorobenzene (HCB) solubility with natural surfactant solutions was estimated and desorption of HCB from soils was investigated with batch and on-dimensional column experiments. This research deals with the dynamic ethnic socio-spatial relations and the transformation of ethnic identities in the early-twentieth-century Honolulu, mainly focusing on the Korean community. Against the widely spread notion that the ethnic relations of Honolulu in those days were little associated with the racist ideology which was prevalent in the contemporary mainland cities, this research shows that white-supremacy ideology had exerted strong influence on the minority groups in Honolulu all the way through their immigration and settling-down process. This dissertation examines the way in which Roland Barthes and Jacques Derrida rework the psychoanalytical constructions of desire through what can be referred to, short of a better word, as play or gaming. Play takes many forms in these two texts; etymological play and structural play are two of the more prominent manifestations of this gaming. New control strategies are based on the model of the process and it is thus necessary to identify the systems to be controlled. It is also often necessary to identify them during closed-loop operation in order to maintain efficient operation and product quality. Some results of multivariable closed-loop identification carried out on a simulated 2 x 2 linear time-invariant system, using two new versions of instrumental variable methods called IV4D and IV4UP as the identification methods, are presented. The purposes of this research were to determine the possibility of using a natural forested swamp for advanced wastewater treatment in coastal Louisiana, and to evaluate the wastewater impact on the ecosystem. The study was performed by comparing two portions of a nutrient deficient swamp separated by a ridge. One portion received secondarily treated municipal wastewater at a rate of $6.3\times10\sp6$ L/day and the other served as a control. Concentrations of BOD$\sb5$, TSS, DO, EC, TDS, pH, N, P, S, CI, K, Ca, Mg, Na and trace metals were monitored. Results showed that the wastewater added nutrients into the swamp. …

A need exists for fish sentinel species for monitoring environmental changes. These studies were performed to examine the suitability of western mosquitofish (Gambusia affinis) as a sentinel species and to explore basic mechanisms of hepatocarcinogenesis. Neoplastic lesions were induced in laboratory-reared mosquitofish fry and adults using single pulse exposures to 1 or 10 ppm methylazoxymethanol acetate. Early changes in the liver of exposed fish included severe cytotoxicity, megalocytosis, and widespread apoptosis. Neoplastic changes were found in the liver in approximately 20% of the exposed fish by 8 weeks after exposure and increased to approximately 70% by 41 weeks. These included …

Studies of recent fish collections from 17 streams on four islands in Hawai'i provided key characteristics for species identification of immature forms of five native species of freshwater gobioids. Young Eleotris sandwicensis lack fused pelvic fins. A suborbital bar identifies Stenogobius hawaiiensis. Awaous guamensis can be distinguished by lack of notches in the upper lip, Lentipes concolor by a single median notch, and Sicyopterus stimpsoni by three notches. Recruitment from oceanic plankton occurred year round but peaked from February to April. Field studies evaluated soybean response to Rhizoctonia foliar blight (RFB) (Rhizoctonia solani AG-1 IA and IB) and interference from common cocklebur, hemp sesbania, or johnsongrass. Soybean maturity was delayed both years by hemp sesbania whether or not RFB was present. Soybean yield was reduced 20% in plots inoculated with the RFB pathogen in 1993, but not in 1994. Soybean yield was reduced only when inoculated plots were infested with common cocklebur and johnsongrass in 1993 and with hemp sesbania and johnsongrass in 1994. This study sought to initiate the process of identifying the rhetoric of mathematics as a distinct field of research, while acknowledging its basis in the rhetoric of science and other literatures. Accordingly, the study started by examining the external basis of the rhetoric of mathematics; in other words, how discourse affects the way in which the culture views mathematics. The primary text for this study was the National Council of Teachers of Mathematics' three-volume Standards for School Mathematics. Reactions of soil arsenic with arsenic addition and the effects of soil arsenic on canola were studied because of a lack of information on this subject. The reactions of different pools of soil arsenic to arsenic addition were studied. The effects of soil arsenic and arsenic form and concentration in solution on canola growth and nutrient uptake were also investigated and an attempt to model arsenic uptake with a mechanistic computer model was made.
