A monoclonal antibody (MAb)-based western blot analysis (WB) was developed for detection of a 26-28 kD coproantigen in Fasciola hepatica infected cattle. Using WB the coproantigen was detected in feces from experimentally infected calves with 22 flukes or more. Using the biotin-streptavidin modification of WB, antigen was detected in the feces from experimentally-infected calves with 10 flukes or more as early as 6 weeks post-infection. The MAbs did not react with the ES of Paramphistomum sp. or Moniezia sp, but showed slight cross-reactivity with ES from F. gigantica, and there was moderate cross-reactivity with Fascioloides magna ES. The Cytoplasmic N,N'-Diacetylchitobiase Gene From Vibrio Parahaemolyticus: Sequence Analysis, Protein Secretion, And Secretion System Development., Michael Hongli Wu

The nucleotide sequence of the gene encoding the cytoplasmic $N,N\sp\prime$-diacetylchitobiase from Vibrio parahaemolyticus has been determined. The deduced peptide sequence surprisingly has minimum evolutionary relationship to two other reported $N,N\sp\prime$-diacetylchitobiases from vibrios, except for highly conserved regions which are also homologous with lysosomal beta-hexosaminidases from eukaryotes including humans. In contrast, the other two sequenced chitobiases from vibrios are much more closely related to each other. This 85 kDa cytoplasmic protein, as revealed from the sequence, appears to be a unique protein, lacking a signal sequence and genetically distant from other known enzymes of similar function. Earth system scientists are increasingly using the technologies of Geographic Information Systems (GIS) and Remote Sensing (RS) in their analyses of earth system processes and patterns. These investigations take place over a wide range of scales, from the local to the global. Global change researchers focus on both the physical and human dimensions of changes in the earth's landscapes, which occur across a range of scales and may be scale dependent. Community structure and relative habitat preferences of intertidal fishes from the eastern Canary Islands of Fuerteventura, Gran Canaria, and Lanzarote were examined in a study of 354 tidepools at 33 localities. Habitat use and behavior of the most abundant fish Mauligobius maderensis were examined on Gran Canaria and Lanzarote. Of the 27 species encountered, 11 were common to each of the islands. The most abundant species were: M. maderensis, Parablennius parvicornis, Gobius paganellus, Coryphoblennius galerita, and Chelon labrosus. In a Two-way indicator species analysis Fuerteventura and Lanzarote localities clustered together. The same analysis differentiated between species that inhabited the upper …

I studied the nesting ecology of the White-faced Ibis (Plegadis chihi) during the summers of 1994 and 1995 in three colonies in Lacassine National Wildlife Refuge, southwestern Louisiana. Data were collected on colony-site characteristics, chronology (with abundance) of colony formation, reproductive success, and growth and development of ibis nestlings. Small colonies nested in dense buttonbush (Cephalanthus occidentalis) and water willow (Decodon verticillatus) "islands." The largest colony nested in black willows (Salix nigra) (3.2 ha)--a habitat unique to ibis populations in Louisiana--with nests heights up to 7.5 m. Cattle egrets (Bubulcus ibis) (62%) and ibises (36%) were the most abundant species. …

This study analyzes the personal narrative of Mary Rhyne, an elderly African American domestic worker. The study describes the ethnographic process of collecting, transcribing and interpreting the informant's oral history. Interpretive and theoretical perspectives concerned with African American culture, domestic labor, and feminist practice are used to analyze the narrative. Further, the ethnographic process, the informant's oral "telling," and the events that she recalls and recounts are viewed as, and in terms of, performance. Performance foregrounds the collaborative and "fictional" aspects of the ethnographer-informant relationship. It directs attention toward the "performance event" as a site and situation where the teller …

I examined productivity, survivorship and nest-site habitat characteristics of small terrestrial landbirds breeding in bottomland hardwood forest study sites in Louisiana. Bottomland hardwood forests are the dominate ecosystem of riverine floodplains in the southeastern United States, and these forests support a diverse bird community. Over 80 percent of this forest type has been lost, primarily due to agricultural clearing, and the relative abundance of many bird species that breed in these forests has also declined. The outcomes of 790 nests of 33 species were determined. For most species, productivity was greater in large forest tracts than in small tracts. Predation …

The purpose of this study was to explore the relationship between preservice teachers' expressed beliefs, their practices, and how they described the two. Particular emphasis was given to factors that influenced the preservice teachers' expectations of students. The preservice teachers were in the graduate year of a five year teacher education program that embraced reflective practice. In many ways, the two preservice teachers conformed to the mold of the typical American teacher in that they were middle class, Anglo Saxon, females who were high achievers from small towns. This study has identified environmental factors influencing the distribution of schistosomiasis and the prevalence of the disease in the population in the state of Bahia, Brazil. The Intergraph Modular Environmental Systems nucleus (MGE), a geographic information system (GIS), was applied to 30 municipalities in Bahia to establish a descriptive and quantitative study of the spatial and temporal dynamics of infection. The GIS was constructed by digitizing maps of soil type, vegetation, topology, and hydrologic features including temperature, rainfall, and seasonal pattern of precipitation, prevalence of schistosomiasis, and the distribution of snails, which are the intermediate hosts of the disease. Coastal wetlands worldwide are threatened by sea level rise, which is expected to affect the growth and survival of wetland vegetation by increasing water level and salinity. While the effects of salinity and inundation on adult vegetation of oligohaline marshes have been widely studied, the species composition of the seed bank and its response to elevated salinity and water level have not been examined. Additionally, the role of disturbances of different intensities in structuring marsh plant communities and possible interactions between disturbance and rising sea level have received little attention. Differences in sugar titers, JH synthesis rate and biogenic amine metabolism correlated with three distinct activity groups or mature bees: resting bees, followers and dancers. Dancers had lower trehalose titers than followers or resting bees. In absconding bees trehalose titers were even lower, due to greatly elevated locomotor activity. The collecting and transferring of food by dancers and followers resulted in a higher glucose titer than in resting bees. Followers had a higher-JH synthesis rate than dancers, suggesting that JH could be an internal motivational stimulus to recruit followers to forage. The biology and ecology of the yellowmargined leaf beetle, Microtheca ochroloma Stal, were studied on cabbage, Brassica oleracea var capitata L., collard, B. oleracea var acephala L., mustard, B. juncea Cosson, turnip, B. rapa L., and radish, Raphanus sativus L. The life cycle of the beetle consists of an egg stage, four larval instars, prepupal, pupal and adult stages. There were no significant differences in the effect of host plant on duration of development of immature beetles (p = 0.3353). A feasibility study of using air-lift pumps for water movement, aeration, and degasification in a recirculating aquaculture system was conducted. Fluid delivery tests of various air-lift configurations indicated that water flow is a function of the submergence depth, lift height, and air injection rate. The air-lift units were found to transfer 02 at only 1/5 to 1/2 the rate of comparably operated open-water aeration systems. The standard oxygen transfer rate (SOTR) increased with air injection from 1-7 scftn, however, the energy usage also increased, yielding a reduction in the standard aeration efficiency (SAE) from over 2 to less than 0.5 …

Oxidative Phosphorylation And The Realkalinization Of Intracellular Ph During Recovery From Anoxia In Artemia Franciscana Embryos, Kurt E. Kwast, Joseph I. Shapiro, Bernard B. Rees, Steven C. Hand

The contribution of mitochondrial oxidative phosphorylation to the realkalinization of intracellular pH (pHi) and resynthesis of purine nucleotides during recovery from anoxia was investigated in embryos of Artemia franciscana by assessing the sensitivity of mitochondrial respiration to pH, calculating proton consumption by oxidative phosphorylation, and measuring changes in pHi using 31P nuclear magnetic resonance. Following short-term anoxia, pHi increased from 6.7 to 7.7 during 20 min of aerobic recovery and was temporally correlated with a large increase in ATP. State 3 respiration rates of isolated mitochondria were not substantially compromised at the acidic pH corresponding to the pHi during anoxia …

Increasing economic importance of cotton production in Louisiana, awareness of the financial and environmental benefits of crop rotations have stressed the need for evaluating cotton production in different rotational schemes. The objective of this study was to estimate profitability of alternative crop rotational schemes within a whole farm context. Yield data for 11 alternative production systems involving cotton, soybean, corn, wheat were obtained from the ongoing crop rotation research at the Northeast Research Station during the period 1983 -1993. The primary purpose of this study was to determine the educational value of 4-H activities as perceived by Louisiana 4-H agents. Data were obtained by a researcher designed survey completed by 91 4-H agents during the 1994 Louisiana Cooperative Extension Service’s annual conference. The data showed the 4-H camp was the highest rated educational activity and it was the only activity perceived as having a very high educational value. Twenty activities were rated as high in educational value, 25 as moderate in educational value, and four items as low in educational value. Agents were also asked to estimate the number …

Exocytosis of transmitter at most synapses is a very fast process triggered by the entry of Ca2+ during an action potential. A reasonable expectation is that the fast step of exocytosis is followed by slow steps readying another vesicle for exocytosis but the identity and kinetics of these steps are presently unclear. By voltage clamping both pre- and postsynaptic neurons in an isolated pair of retinal amacrine cells, we have measured evoked synaptic currents and responses to single vesicles of transmitter (minis). From these currents, we have computed the rate of exocytosis during a sustained presynaptic depolarization. We show here …

ID elements comprise a rodent SINE (short Interspersed DNA repetitive element) family that has amplified by retroposition of a few master genes. In order to understand the Important factors of SINE amplification, we investigated the transcription of rat ID elements. Three different size classes of ID transcripts, BC1, BC2 and T3, have been detected In various rat tissues, including brain and testes. We have analysed the nucleotlde sequences of testes- and brain-derived ID transcripts isolated by size-fractlonation, C-talling and RACE. Visualization Of Chromosomal Domains With Boundary Element-Associated Factor Beaf-32, Keji Zhao, Craig M. Hart, Ulrich K. Laemmli

We have purified two proteins from Drosophila that bind to the scs' boundary element of the hsp70 domain at locus 87A7. Their palindromic binding sites (CGATA-TATCG) symmetrically abut the previously mapped hypersensitive site of scs′. We have cloned a cDNA for one of these proteins, BEAF-32 (boundary element-associated factor of 32 kDa). It encodes a novel protein that is bound to scs' but not scs in vivo. Immunostaining localizes BEAF to hundreds of interbands and many puff boundaries on polytene chromosomes, suggesting that a chromosomal domain consists of a band (or puff) and part of the flanking interbands. Gross observation of testicular parenchyma of 1.5 to 2-yr-old horses reveals both light and dark regions. If this gross, differential shading reflects quantitative differences in the development of spermatogenesis and interstitial cell populations, the horse may prove to be a useful model for study of the paracrine relationships associated with initiation of spermatogenesis. The objective of this study was to characterize seminiferous tubules and interstitium of testes with gross, differential shading. Testes with both light and dark regions of parenchyma were obtained from horses 1.5- 2 yr old and compared to parenchyma of fetal, 2-yr old, or 5-yr-old horses. Stereology …

One hundred forty-five boys ages 6 to 11 were administered the Social Skills Rating System and the Matson Evaluation of Social Skills with Youngsters. Subjects were divided into the following groups based on IQ, adaptive behavior, and score on a checklist of ADHD symptoms: normal IQ comparison, normal IQ high-activity-level, normal IQ medicated, mentally retarded comparison, mentally retarded high-activity-level, and mentally retarded medicated. For both normal IQ and mentally retarded children, high-activity-level resulted in higher levels of negative social behavior. Both groups of high-activity level subjects also scored lower on classroom-related social behaviors. Medicated normal IQ subjects displayed social behavior …

The purpose of this study was to describe extension agricultural agents' perceptions of sustainable agriculture in the Southern Region of the United States. The population included Cooperative Extension county/parish agents with major responsibility in agriculture working in the 13 states of the Southern Region of the United States. These individuals were either employed by the thirteen institutions established by the Morrill Act of 1862 (the 1862 Cooperative Extension Services) or by eleven institutions established by the Second Morrill Act of 1890 (the 1890 Cooperative Extension Services). This study adds to the literature on adult gender role by extending adult gender role orientation beyond increases and/or decreases on gender related personality attributes, in particular increases in self-rated Masculine and Feminine ratings. Gender role orientation was examined along three dimensions: gender-related personality attributes of self, attitudes toward gender role issues, and gender-related attributes of others in social contexts. Life stage experience and social cognitive functioning influences on gender role orientation were examined. This project explores the function of body politics in constructing minority identities, or how people's physical and stylistic attributes are invested with meanings about who they are. It is interested in how race and sexual differences are defined in the confluence of discourses around visibility and invisibility. The first two chapters set up the parameters of in/visibility with regard to sexual and racial differences in readings of two paradigmatic texts about visibility, Radclyffe Hall's The Well of Loneliness, which produces the lesbian as visible in the figure of the butch, and Ralph Ellison's Invisible Man, which explores the paradoxical notion …
