Two-hundred and thirty crude organic extracts from 118 plant species distributed among ten families of higher plants have been evaluated for antimycobacterial activity against Mycobacterium tuberculosis (H$\sb{37}$Rv) and M. avium using the BACTEC 460 radiorespirometric assay. At 100 $\mu$g/ml, twenty-four and ten of the extracts caused more than 95% inhibition of growth of M. tuberculosis and M. avium, respectively. Bioactive chromatographic fractions of Borrichia frutescens (Asteraceae) provided two new triterpenes and one known cycloartenol. In radiorespirometric bioassays against M. tuberculosis, the new (24R)-24,25-epoxycycloartan-3-one and the known (24R)-24,25-epoxycycloartan-3$\beta$-ol exhibited minimum inhibitory concentrations (MICs) of 8 $\mu$g/ml. Efficacy studies are often done on diseased animals while pharmacokinetic studies are done on healthy animals. Disease models are inherently difficult to standardize because of the many variables associated with measuring pharmacokinetic parameters in abnormal animals. Information derived from pharmacokinetic studies with diseased animals could be useful to determine if therapeutic doses of a drug or antimicrobial becomes less effective or toxic due to changes in distribution and clearance. A polymerase chain reaction method for identifying individuals in the Anopheles quadrimaculatus Say sibling species complex was validated for wild mosquitoes from Louisiana and Mississippi. This method distinguished An. quadrimaculatus species A, B, C, and D by detecting species-specific differences in the 2nd internal transcribed spacer of ribosomal DNA and was 100% specific and 95% sensitive. A polymerase chain reaction assay that detects differences in the second internal transcribed spacer of ribosomal DNA was tested for its usefulness in identifying all immature stages of two representatives in the An. quadrimaculatus sibling species complex. Despite the wealth of Mexican archival data pertinent to understanding the ecological interaction between livestock and landscape during the sixteenth century in New Spain, some fundamental methodological issues persist in the use of the mercedes, or land grants. The degree of completeness of the extant mercedes record and the de facto areal extents of the cattle and sheep estancias both remain uncertain. A cartographic method applied to mercedes for the central Veracruz lowlands addresses those issues and suggests a general method for rigorous analysis of the data base. Nonadherence to medical regimens is a critical behavioral threat to the health of HIV-infected individuals. Patients cannot benefit from increasingly efficacious treatments for HIV/AIDs unless they adhere to them. Consistent medical care plays a central role in prolonging life and enhancing quality of life for these individuals. It is therefore essential to understand why many patients do not return reliably for medical services. However, little research has focused on the development of empirically based models of HIV-related nonadherence. The present study investigated the frequency and intensity of behavior problems exhibited by Human Immunodeficiency Virus (HIV)-infected toddlers ages 16 to 40 months by comparing them to HIV-exposed toddlers, toddlers at risk for development delays, and healthy toddlers. The role of maternal factors, such as depression, insularity, and stress, in behavior ratings also was considered. Mothers completed a set of questionnaires and the toddlers' developmental status was assessed by a member of the research team. The extracellular capsule polysaccharide (CPS) of Vibrio vulnificus is a primary virulence factor which allows survival of the bacteria in the human host. To study the genes involved in the production of the polysaccharide capsule, 23 mutants were generated that lost their ability to produce CPS due to the insertion of a mini-transposon into the genome of the encapsulated clinical strain V. vulnificus 1003 (O). The mutants were examined by Southern blot analysis to ensure the transposon had inserted only once in the chromosome. The Htra Stress Response Protease Contributes To Resistance Of Brucella Abortus To Killing By Murine Phagocytes, Philip H. Elzer, Robert W. Phillips, Gregory T. Robertson, R. Martin Roop

Compared with virulent Brucella abortus 2308, the isogenic htrA mutant PHE1 shows decreased resistance to killing by cultured murine neutrophils and macrophages and significant attenuation during the early stages of infection in the BALB/c mouse model. These findings further define the contributions of the htrA gene product to the pathogenesis of B. abortus infections.

An Examination Of The Effects Of The Mother's Education And Household Exposure To Disease On Childhood Diarrheal Morbidity In Sudan., Dawood Hussein Sultan

This dissertation examines the effect of increases in the mother's level of education on the likelihood of diarrheal morbidity among Sudanese children aged 6 to 23 months. Maximum likelihood logistic regression models are used in the analysis. The logit estimates obtained from these models revealed that children of mothers who are illiterate run on average more than 10 times the risk of diarrhea as that run by children to mothers with a senior secondary or higher education. Slight increases in the mother's education, as indicated by an incomplete primary school education, reduce the likelihood of diarrhea by more than 50%. …

Cecropins are basic, amphipathic peptides with antibacterial activity against many Gram-positive and Gram-negative bacteria. By retaining the positively charged, amphipathic characteristic of cecropin B, a cecropin analog S1, in which 54% of amino acid residues were substituted, was synthesized and shown to possess a broad spectrum of antimicrobial activity. To determine expression of the cecropin analog S1 in Sf9 cells, two gene constructs were designed, and inserted into Autographa californica nuclear polyhedrosis virus under the control of the polyhedrin promoter. Rapid, generic, and environmentally friendly methods for extraction and detection of the pesticides atrazine, cyanazine, metribuzin, and simazine were developed for use in monitoring the cause of pesticide related or suspect fish kills. Further, environmental hypoxia and elevated temperature were studied as possible contributors to the toxicity of atrazine in channel catfish (Ictalurus punctatus) and as synergistic factors to be considered in investigating fish kill events. Incurred residues from the exposure of catfish to atrazine alone or hypoxia or elevated temperature and atrazine were also analyzed. Slow-to-Recover (STR) individuals are a subset of the neurologically impaired population who remain non-responsive for extended periods of time before beginning to demonstrate improvement in cognitive, communicative, or motor function. This investigation focuses on the development and implementation of a facilitator-based intervention program carried out with an STR brain injured adult residing in a long-term care facility. The model involved: (1) sensory stimulation techniques to increase responses to external stimuli, (2) augmentative techniques which enabled the subject to control his immediate environment and, (3) training of qualified residents within the facility to act as communication facilitators. Two primer sets were manufactured from the cytadhesin P1 gene of M. Pneumoniae in an attempt to amplify homologous sequences in M. mycoides subsp. mycoides "SC" Type in a search for a P1-like gene in the latter organism. The first primer set MP-2 amplifies a 488 bp fragment in M. pneumoniae. The second primer set MP-3 amplifies a 303 bp fragment bracketing the 13-amino acid binding domain in the same organism. The U3 region of long terminal repeat (LTR) of equine infectious anemia virus (EIAV) contains transcriptional regulatory elements. To identify the elements in prototype (P), fetal donkey dermal cell-adapted (A), and two variant (V) EIAV LTRs, DNase I footprinting, gel shift, and transient gene expression assays were performed. The assays used U937 monocytic cells which can be induced into macrophages by 12-O-tetradecanoylphorbol-13-acetate (TPA). For study of AP1 sites, c-Jun/AP1 was also used. Much of the attention has focused on who provides care and what impact that caregiving has on them. It has been well documented that the patient's family is responsible for much of the caregiving. A need exists for fish sentinel species for monitoring environmental changes. These studies were performed to examine the suitability of western mosquitofish (Gambusia affinis) as a sentinel species and to explore basic mechanisms of hepatocarcinogenesis. Neoplastic lesions were induced in laboratory-reared mosquitofish fry and adults using single pulse exposures to 1 or 10 ppm methylazoxymethanol acetate. Early changes in the liver of exposed fish included severe cytotoxicity, megalocytosis, and widespread apoptosis. Neoplastic changes were found in the liver in approximately 20% of the exposed fish by 8 weeks after exposure and increased to approximately 70% by 41 weeks. These included …

Many unanswered questions on the epidemiology and ecology of eastern equine encephalomyelitis (EEE) still preclude a complete understanding of its natural history. Among these are the distribution and epidemic vectors of EEE virus (EEEV) in different regions, habitats, and foci; its inter-epidemic maintenance, and overwintering mechanisms. The coronavirus strain HECV‐4408 was isolated from diarrhea fluid of a 6‐year‐old child with acute diarrhea and propagated in human rectal tumor (HRT‐18) cells. Electron microscopy revealed coronavirus particles in the diarrhea fluid sample and the infected HRT‐18 cell cultures. This virus possessed hemagglutinating and acetylesterase activities and caused cytopathic effects in HRT‐18 cells but not in MDBK, GBK and FE cells. One of four S‐specific monoclonal antibodies reacted in Western blots with HECV‐4408, BCV‐L9 and BCV‐LY138 but not with HCV‐OC43, and two reacted with BCV‐L9 but not with HECV‐4408, BCV‐LY138 and HCV‐OC43. Comparative resistance or tolerance of four stocks of Apis mellifera L. to Varroa jacobsoni Oudemans, Acarapis dorsalis Morgenthaler, Acarapis externus Morgenthaler and Acarapis woodi (Rennie), was investigated using choice (bioassay) and field experiments. A. m. carnica from Yugoslavia (ARS-Y-C-1), A. m. carnica from Canada (Hastings), F$\sb1$ hybrid between ARS-Y-C-1 and Hastings and a general Louisiana stock were evaluated. A tolerance index showed that Hastings, ARS-Y-C-1 and F$\sb1$ hybrid exhibited some degree of tolerance to V. jacobsoni. These stocks lived longer with higher levels of Varroa infestation in worker pupae and on adult honey bees. Louisiana stock was more susceptible to …

Three amber mutations were introduced proximal to the syn3 locus of the herpes simplex virus type 1 glycoprotein B (gB) gene specifying gB derivatives lacking the carboxy-terminal 28, 49, or 64 amino acids. A complementation system that utilized gBs expressed in COS cells to complement gB-null virus KΔT was established. The 49- or 64-amino-acid-truncated gBs failed to complement gB-null virus KΔT, while the 28-amino-acid-truncated gB complemented KΔT efficiently. Mutant herpes simplex virus type 1 KOS (amb1511-7) specifying the 28-amino-acid-truncated gB fused Vero cells extensively.

Five Macaca mulatta were infected with SIV/DELTA$\sb{\rm B670}.$ Approximately 70 days later, this group (SIV) and five age, sex, weight matched not SIV-infected (NEG) M. mulatta were inoculated orally with approximately $3\times10\sp{10}$ Campylobacter jejuni. Small and large bowel mucosal biopsies, bowel contents, sera, blood, and rectal swabs were collected before and after C. jejuni inoculation. Biopsy cryosections were stained with histologic stains and immunocytochemically for lymphocyte subsets, plasmacytes, macrophages, and B-cells. Coded stained sections were examined semiquantitatively or quantitatively using computerized image analysis. Circulating lymphocyte subsets were determined using FACS analysis. In a Terrebonne Basin (Louisiana) marsh, Cs dating indicated that vertical accretion (0.98cm yr ) was extremely rapid relative to other marshes, but insufficient to counter submergence (1.38cm yr ). Mineral and organic matter accumulation were similar to that in other marshes. This marsh is threatened by a positive feedback loop of plant flooding stress and inadequate vertical accretion. Inadequate plant growth limits vertical accretion, which further increases flooding and decreases plant production. Thus plant production partly determines the degree of submergence some coastal and estuarine marshes will tolerate. -from Authors 137 -1 -1

Polar flagellar core protein was purified from Vibrio parahaemolyticus by differential centrifugation and cesium chloride isopycnic centrifugation. Twelve hybridomas secreting monoclonal antibodies against flagellar core protein (MAb-flc) were obtained from fusions. By chance, one purified flagellar core preparation retained flagellar sheath and as a consequence, two hybridomas were detected which secreted anti-sheath antibodies (MAb-fls). MAb-flc and MAb-fls reacted specifically with their corresponding antigens as demonstrated by immunogold labelling. Coagglutination reagents prepared with MAb-flc and MAb-fls were tested against 34 strains of V. parahaemolyticus and 34 heterologous Vibrio species. A profound decrease in perfusion of the laminar capillaries has been documented during acute equine laminitis, resulting in ischemia of the laminar tissue. Oxygen radicals are believed to mediate ischemia/reperfusion injury. The effects of (1) three hours of surgical ligation of the major blood supply of the equine digit followed by ligature release, (2) resection of the medial and lateral palmar digital arteries, and (3) digital intraarterial infusions of an oxygen radical generating system, were investigated. The goals of this project were to investigate factors that predicted nonadherence to fluid restrictions for patients on hemodialysis, and to evaluate a relatively long-term behavioral intervention to improve adherence to recommended fluid requirements. One hundred forty-one subjects were recruited from outpatient dialysis clinics in Baton Rouge, Louisiana. In the prediction study, several medical and demographic variables as well as psychological variables of health locus of control, depressed mood, and social support were placed into a regression equation to determine the variance of fluid noncompliance predicted. The lentivirus Equine Infectious Anemia Virus (EIAV) causes an acute infection in horses which is characterized by fever, viremia, thrombocytopenia, edema, and hemolytic anemia. Most horses survive the acute disease, but remain persistently infected for life. The exact mechanism whereby horses' control their initial infection is unknown, though it appears likely that cellular immune mechanisms are involved. T lymphocytes have been shown to play a major role in the recovery from a variety of viral infections, though their role in EIAV infection has not been established.
