The physiologic demands of exercise constitute a form of stress which is accompanied by modulation of immune responses. However, numerous studies in many different model systems have failed to clearly define the effects of exercise on immunity. Furthermore, the role of the neuroendocrine system in exercise-induced immunomodulation has been largely unexplored. To address these issues, we evaluated the effect of acute exercise stress on 3 indices of immune function in horses, before and after completion of a 12 week treadmill-based physical conditioning program. The complete nucleotide sequences of the hemagglutinin/esterase (HE) genes of human coronavirus (HCV) strain OC43 and bovine respiratory coronavirus (BRCV) strain G95 were determined from single-stranded cDNA fragments generated by reverse transcription of virus-specific mRNAs and amplified by polymerase chain reaction. An open reading frame of 1272 nucleotides was identified as the putative HE gene by homology to the bovine coronavirus HE gene. This open reading frame encodes a protein of 424 amino acids with an estimated molecular weight of 47.7 kDa. Ten potential N-linked glycosylation sites were predicted in the HE protein of HCV-OC43 while nine of them were …

Specific and sensitive amplification of major outer membrane protein (MOMP) gene (ompA) DNA sequences of Chlamydia species with various MOMP genotypes was achieved by a two-step polymerase chain reaction (PCR). Degenerate, inosine-containing oligonucleotide primers homologous to the 5' and 3' ends of the translated regions of all chlamydial MOMP genes were used in a PCR to amplify a DNA fragment of approximately 1,120 bp. A portion of this DNA fragment was amplified in a second genus-specific reaction that yielded a DNA fragment of approximately 930 bp. The mechanism by which a horse conceptus-derived immunosuppressive factor (HCS) of M(r) >100 000 inhibits lymphocyte proliferation was investigated. The factor was obtained from the culture supernatants of 20-day-old horse conceptuses; activity, identified by reduced uptake of [3H]thymidine by mitogen-stimulated lymphocytes, was greatest (P0.05). Data from a fluorescence-activated cell sorter indicated that supplementation with HCS reduced the number of T cells in phytohaemagglutinin-stimulated cultures and suppressed proliferation of T and B cells in pokeweed-mitogen-stimulated cultures compared with controls. Specific and sensitive amplification of major outer membrane protein (MOMP) gene DNA sequences of Chlamydia psittaci was achieved in a two-step polymerase chain reaction. First, oligonucleotide primers specific for 5' and 3' nontranslated regulatory regions of the MOMP gene were used in a polymerase chain reaction to amplify a DNA fragment of approximately 1,400 bp. A portion of this DNA fragment was amplified in a second reaction using a degenerate oligonucleotide primer specific for a DNA sequence contained within the 1,400-bp DNA fragment and one of the first-step primers. One solution to overcome the shortage of hospital nurses is to establish and implement clinical career ladder programs. The purpose of this study was to examine hospital clinical nurses' perceptions of ladder programs as a job enrichment strategy and to determine individual and work-related variables contributing most to nurses' participation or nonparticipation in available clinical ladder programs. A random sample of 600 clinical nurses employed full time in five regional medical center hospitals located in Louisiana and Mississippi were the study subjects. Respondents were 106 (88.3%) of the 120 ladder program participants and 385 (80.2%) of the 480 nonparticipant nurses. …

Calyptospora funduli is a pathogenic coccidium that infects the liver and pancreas of estuarine killifishes belonging to the genus Fundulus. Experimental infection of the atypical fish host Rivulus marmoratus with this intracellular protozoan parasite caused the development of hepatic macrophage aggregates (MA's) and granulomas. The Rivulus/Calyptospora host-parasite system thus represents a new laboratory model for investigations of MA biology. In the present study, this model was used to: (1) examine the morphology and sequential development of the parasite-elicited hepatic MA's in R. marmoratus, (2) define the role that these structures play in the pathogenesis of an infectious disease process, and …

This dissertation discusses modifications of the internal architecture of production systems that could significantly increase the execution performance of production system programs. This increased efficiency is achieved in part by modifications to the matching step of the execution cycle. Rather than checking all possible instantiations, we propose to consider only a small subset of the potential instantiations which are the "best candidates" for firing (according to the conflict resolution scheme). The increased execution efficiency provided by this matching strategy is compounded by modifying the OPS5 conflict resolution strategies. We propose a goal-directed (look-ahead) conflict resolution strategy which will still retain …

The Carl D. Perkins Act of 1984 provides significant financial support for the enhancement and expansion of secondary and post-secondary vocational education programs in the United States. Individual states are required to develop a State Plan for the use of Carl D. Perkins vocational funds. The formulation of this State Plan requires the development of Local Plans on the part of local education agencies who wish to participate in this program. The purpose of this study was to develop a Toddler Behavior Screening Inventory (TBSI) to rapidly assess maternal-perceived toddler behavior problems. The TBSI is a 40-item socially valid measure intended for use in clinical settings as it will aid child care professionals in planning parent-child intervention strategies. Empirical methods were followed to construct the TBSI. The item pool was generated primarily by 187 mothers with toddlers and relevant professionals. Items were reviewed and formatted before 312 mothers rated the initial 93-item TBSI in terms of behavior frequency and problem severity in order to eliminate items. Forty items were retained based …

Previous investigations have characterized both the genomic and antigenic variations in equine infectious anemia virus (EIAV) surface glycoproteins that arise during replication of the virus. DNA sequence analyses have defined regions of conserved and variable sequences localized in the EIAV envelope glycoproteins. These studies indicate that EIAV persistence and periodicity of clinical disease result from the relatively rapid evolution of antigenic variants which allows the virus to temporarily circumvent the host immune response. Vibrio anguillarum exhibits species-specific antigens on the protein core of the polar flagellum (H) and H-determinants which are expressed by heterologous Vibrio species. Monoclonal antibody (MAb) which reacted with flagella core protein by ELISA were affinity purified from protein A-sepharose. Staphylococcus aureus Cowan 1 cells armed with anti-H MAb coagglutinated each of the 10 V. anguillarum O-antigen serovars within 1 to 2 minutes, as well as V. ordalii isolates. These findings suggest these two vibrios express similar if not identical species-specific H-determinants. The anti-H reagent did not coagglutinate 20 heterologous Vibrio species. The effects of in utero and neonatal exposure to maternal Brugia pahangi infections on the development of homologous infections, immune responses, and pathologic lesions was studied in age-matched progeny of infected and uninfected female jirds. High IgG antibody titers to B. pahangi antigens were present in the sera of progeny from infected mothers. These neonatal titers to homologous antigens were shown to be of maternal origin, and did not alter the cellular responses of uninfected progeny as measured by in vitro antigen-stimulated blastogenesis and in vivo pulmonary granulomatous inflammatory responses to antigen-coated beads. This study was divided into: (1) a descriptive study of the farm resources, management practices, veterinary services utilization, and socio-psychological characteristics of Louisiana ranchers in relation to beef cattle performance, (b) a classification of the ranchers according to their utilization of veterinary services with the objective of identifying producer characteristics that are associated with frequent, moderate, and non-use of veterinary services, and (c) identification of factors associated with continuing or ending beef cattle production. The intensity of management, herd health and veterinary services utilization was quantified by scoring and ranking indicator variables that defined these composite variables. Most ranchers had …

Prevalence of mastitis and efficacy of intramammary treatment were determined in unbred and primigravid heifers (n = 116) from 4 dairy herds. Of 97 heifers from which secretion samples were obtained, intramammary infections were detected in 96.9% of animals and 74.6% of quarters. Clinical symptoms were observed in 29% of heifers. Staphylococcus aureus infections were detected in 37.1% of animals. Teat canal colonizations were confirmed in 93.1% of heifers and 70.7% of quarters. The most common isolates from mammary glands were Staphylococcus chromogenes, Staphylococcus hyicus, and Staphylococcus aureus. Average somatic cell counts in secretion from infected and uninfected quarters were …

This experiment was conducted to further define epidemiological events involved in the induction, duration and maturation phases of inhibited development of Ostertagia ostertagi in naturally infected calves and to characterize some aspects of the immune response associated with the inhibition phenomenon. During March, April and May 1988, 3 groups of 18 calves were grazed on the same experimental pasture for 3 weeks. At the end of each grazing period, 2 calves of each group were slaughtered for analysis of worm population; the remaining calves were divided into 2 subgroups of 8 calves. A genuswide protein antigen extracted from Legionella pneumophila serogroup 1 (strain Philadelphia 1) cells was enriched by differential pelleting and ammonium sulfate precipitation and subsequently purified with a combination of high-performance size-exclusion and ion-exchange chromatography. The protein has an apparent molecular weight of 650,000 before and 63,000 after urea (5 M) treatment, as determined by size-exclusion chromatography. These proteins resolved to a single band of 60,000 after sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The urea-treated protein had an isoelectric point of 5.8. This purified 60-kilodalton protein reacted with a convalescent-phase serum sample from a patient with legionellosis and rabbit immune sera …

A study was conducted to determine the prevalence of Campylobacter jejuni in various avian species and humans. Cloacal swabs derived from birds submitted to the Louisiana Veterinary Medical Diagnostic Laboratory for necropsy examination during the period February 1987 through July 1988 were routinely cultured for C. jejuni using Butzler's selective medium. Campylobacter jejuni was isolated from 10.1% of the cases examined. The isolation rate was 25.2% in Galliformes, 12.9% in Anseriformes, 8.3% in Columbiformes, 7.7% in Falconiformes, and 0.6% in Psittaciformes. None of the other orders of birds examined yielded C. jejuni. Prevalence data demonstrated that Galliformes are important reservoirs …

Throughout history, people speculated that an association existed between nutrition and disease prevention. Recently, studies have examined the influence of single nutrients on immunological mechanisms, but with limited focus on marginal deficiencies. This project investigated the effects of severe, marginal, and adequate levels of dietary magnesium and copper on immune function. Three experiments were conducted, with 100 gm male rats consuming one treatment diet for three, four or eight weeks. Mineral levels used were: (1) magnesium--50 ug/gm, 160 ug/gm, 280 ug/gm, 400 ug/gm; (2) copper--0.5 ug/gm, 2.0 ug/gm, 3.5 ug/gm, 5.0 ug/gm; or (3) a combination of the severely deficient …

The composition of a forest of relicts of cultivation in four uninhabited valleys in Kohala, Hawaii, is documented. A general hypothesis is made that arboreal distribution patterns are a function of both historical land use and ecological interaction since abandonment. The physical and historical geography of the valleys was investigated. Climate varies little, but distinct geomorphic zones offer differing biological environments. Prehistoric land use consisted of taro patches with intercropped banks. Talus slope gardens supported the Polynesian tree crops 'ohi'a 'ai (Eugenia malaccensis), kukui (Aleurites moluccana), 'ulu, (Artocarpus incisus), ti (Cordyline terminalis), and noni (Morinda citrifolia), important in today's flora. …

Three Shetland ponies (A, B and C) were inoculated with plasma from a pony with clinical signs of equine infectious anemia (EIA) and observed for 165 to 440 days. Clinical signs of EIA marked by high fever (39.5$\sp\circ$C) lasting 2-4 days were observed 2-3 weeks after infection and one to four such episodes occurred in each pony. EIAV was reisolated from plasma by end-point dilution in fetal equine kidney (FEK) cells and a plasma viremia 10$\sp{3.5}$ TCID$\sb{50}$/ml was observed during each episode. The pathogenesis of equine herpesvirus type 1 (EHV-1) and type 2 (EHV-2) respiratory infection was studied in cell and organ cultures by infectivity assays following experimental infections (growth kinetics), and by light, scanning and transmission electron microscopy. The cell cultures included fetal donkey and pony alveolar macrophages (FDAM/FPAM), fetal equine kidney (FEK), rabbit kidney (LLC-RK(,1)), and african green monkey kidney (VERO) cells. Organ cultures were derived from respiratory tissues (nasal, tracheal and pharyngeal mucosae) of seronegative donkey (F1D1) and pony (F1M1) fetuses, and of a seropositive adult pony (A1). Antibody responses to the apparently species specific phenolic-glycolipid-1 (Phen-Gl-1) antigen of Mycobacterium leprae were examined in humans and armadillos using enzyme-linked immunosorbent assays (ELISA). Statistical definitions for the interpretation of positive and negative reactions were derived. A retrospective serological survey of armadillos indicated that leprosy in the wild armadillo is a naturally acquired zoonosis. Presently 12.5% of the armadillos in 2 parishes in south central Louisiana have detectable IgM antibodies to Phen-Gl-1. Approximately 2.7% of these histologically exhibit clinical disease. Recombinant DNA technology was used to produce a probe for detection of Clavibacter xyli subsp. xyli, the sugarcane ratoon stunting disease agent. C. xyli, subsp. xyli chromosomal DNA was digested with the restriction enzyme Pst 1, inserted into the plasmid pUC 19, and transformed into Escherichia coli JM 83. Recombinant clones containing six different sizes of C. xyli subsp. xyli DNA sequences were obtained. A total of 402 bovine embryos, non-surgically collected on day 6.5-7.5 after estrus, were utilized in evaluation of fluorescein diacetate (FDA) for estimating embryo viability in vitro and to investigate the effect of Bovine Viral Diarrhea (BVD) Virus on embryonic development during in vitro culture. Fluorescent assessment, using UV microscopy (440-480 nm) of embryos after staining in FDA (2.5 (mu)g/ml phosphate buffered saline: PBS), was found to be a rapid, non-toxic and accurate method for evaluation of viability. Serratia marcescens LRL781, a gram-negative bacterium, produces an extracellular protease. Evidence is provided for an outer membrane-associated, larger molecular weight, enzymatically inactive precursor of the protease. Antiserum was prepared against the extracellular protease, which was purified from the supernatant fraction of S. marcescens LRL781 cultures. Column adsorption of the antiprotease serum was performed using a slurry of live cells and cellulose. The resulting effluent was serologically inactive against protease preparations and chromium chloride-coupled protease-rabbit erythrocytes. Whole washed cells agglutinated in the presence of the antiserum. Cells were uniformly labeled with ferritin conjugated goat anti-rabbit IgG, indicating that the protease precursor …

A canine rotavirus isolate caused clinical disease, small intestinal lesions, serum antibody response, and shedding of rotavirus in the feces in newborn gnotobiotic dogs. The results of this study demonstrated this rotavirus was a pathogen in neonatal gnotobiotic dogs. The pathogenesis of the intestinal lesions was similar to the pathogenesis of rotaviral enteritis in other species. In three consecutive experiments, twenty puppies were derived by caesarean section and transferred to sterile isolators. The interaction of reovirus infection and manganese, biotin, niacin, choline, or folic acid fed to chickens at a dietary level of 20%, 100%, or 200% of the National Research Council Nutrient requirements of poultry-1977 (NRC-77) (Nat. Acad. Sci., Washington, D.C.) was evaluated in five studies. Responses measured were the incidence and severity of tenosynovitis, the incidence of nutrient deficiency lesions, bone shearing strength, and immunologic responses. The severity of tenosynovitis was greater in male chickens than female chickens fed varying dietary levels of manganese, biotin, choline, or folic acid.
