In this report, placement of Rickettsia felis in the spotted fever group (SFG) rather than the typhus group (TG) of Rickettsia is proposed. The organism, which was first observed in cat fleas (Ctenocephalides felis) by electron microscopy, has not yet been reported to have been cultivated reproducibly, thereby limiting the standard rickettsial typing by serological means. To overcome this challenge, several genes were selected as targets to be utilized for the classification of R. felis. Peptides and peptide analogs are central to the understanding of biological and biochemical phenomena as well as in the development of new pharmaceutical agents. This research encompasses two classes of peptide mimetics: phosphonopeptides and short, amphipathic peptides rich in alpha,alpha-disubstituted amino acids (alphaalphaAA). The focus of the first part of the peptide analog project was on the preparation of sterically-hindered phosphonamide and thiophosphonamide dipeptides. Conditions were developed, based on our P(III) synthetic approach, for the preparation of the phosphonamide dipeptides, Cbz-CyhGly psi [P (O) (OpNb) NH] Trp-NH2 and Cbz-CyhGly psi [P (O) (OpNb) NH] Trp-OMe, which were previously unattainable by …

Previous abusive clinical trials have caused several obstacles in recruiting African Americans for clinical trials today. The memory of the Tuskegee Syphilis Study alone remains a hard pill to swallow and is a constant hindrance to recruiting potential African Americans specifically males, for clinical trials. This study explores how advertising and company promotion may be used to address social issues. In particular, I examine how the clothing company United Colors of Benetton attempted to position itself within the AIDS crisis. Specifically, this study investigates through case studies how the Benetton Group used activist advertising and company promotion to bring about social change concerning AIDS-related issues. I also attempt to account for the controversy that the promotion elicited through establishing the anomalous status of the company's promotion within traditional advertising efforts. Our model examines the allocation of medical research funds at the National Institutes of Health using public interest theory, incremental budgeting theory, and special interest group theory. We use the allocation of research funds among various diseases and measures of the burden of disease on the population to test the hypothesis that the N.I.H. is allocating funds on a pure public interest basis, to test for incremental budgeting effects, and to test for the influence of lobbying and other political variables representing special interest groups. T cell immunity to lytic proteins of herpesviruses is important in host control of infection. We have characterized the cytotoxic T lymphocyte (CTL) response to 5 human herpesvirus 8 (HHV-8) homologues of lytic proteins in HHV-8-seropositive individuals. HLA class I-restricted, CD8+ CTL responses to ≥1 HHV-8 lytic protein were detected in all 14 HHV-8-seropositive study subjects tested, with or without human immunodeficiency virus type 1 (HIV-1) infection, but not in any of 5 HHV-8-seronegative individuals. A multiplex amplification and detection platform for the diagnosis of Mycobacterium bovis and Brucella abortus infection simultaneously in bovine milk and nasal secretions was developed. This system (designated the bovine pathogen detection assay [BPDA]-PCR) consists of duplex amplification of species-specific targets (a region of the BCSP31K gene of B. abortus and a repeat-sequence region in the hsp65 gene of M. bovis, respectively). This is followed by a solid-phase probe capture hybridization of amplicons for detection. Epidemiological studies have failed to show any important impact of human immunodeficiency virus (HIV) infection and disease on leprosy, in contrast to the well documented increase in incidence and morbidity with other mycobacteria in these patients, notably Mycobacterium tuberculosis. The early events following exposure and repeated exposure to Mycobacterium leprae (ML) have not still been studied. Previous studies of first inoculations of ML in the skin of rhesus monkeys have indicated delayed recruitment of CD4+ cells and delayed interleukin (IL) 2 secretion in skin inoculation sites in simian immunodeficiency (SIV) positive animals. Research suggests that individuals with Posttraumatic Stress Disorder (PTSD) have significant dysregulation in a number of physiological indices, especially the sympathetic nervous system (SNS) and hypothalamic-pituitary-adrenal (HPA) axis. Biological theories of PTSD propose that prolonged SNS and HPA axis dysregulation places individuals with PTSD at risk for development of medical morbidity and impairments in health status. Bartonella henselae is a zoonotic agent in which the domestic cat serves as the natural reservoir, and humans acquire potentially serious infections associated with this microorganism. The purpose of this research is to contribute to the understanding of the pathogenesis of B. henselae in the domestic cat using a molecular approach. Using sequence differences in a portion of the I 6S rRNA gene between B. henselae genotype I, and B. henselae genotype II, a nested polymerase chain reaction (nPCR) was designed and used to investigate various phases of feline bartonellosis. The goal of this work was to evaluate the feasibility of using antimicrobial peptide gene transfer to enhance oyster immunity. The specific objectives were to: (1) compare the efficiency of collagenase, pronase, and trypsin to dissociate atrium, ventricle and mantle into individual cells for the initiation of primary cell culture; (2) develop cryopreservation conditions to preserve the dissociated cells; (3) establish a serum-free transfection system; (4) evaluate inducible promoter function using the established serum-free transfection system; (5) evaluate the effects of the transfer of antimicrobial peptide genes on oyster immunity. Cancer is one of the most serious health problems in the U.S. Louisiana mortality rates for all sites of cancer have exceeded national rates since 1950. Even though concern about the issues of environmental cancer is growing, conclusive results have not been reported. This research was part of a multidisciplinary effort aimed at the elucidation of the mechanism of membrane lysis by model peptides. The focus of the dissertation was to characterize the average size dimensions of model cell membranes before and after being treated by model peptides. Experiments with neutral dioleoylphosphatidylcholine (DOPC) vesicles and negatively charged dioleoylphosphabdylcholine/dioleoylphosphabdylglycerol (DOPC/DOPG) vesicles were conducted using the model peptides (KLAKKLA) 3 and (KLGKKLG)3. Evaluation Of Insecticide Resistance In Louisiana Adult Mosquitoes And The Efficacies Of Selected Larvicides., Stacy Lee Bearden

Pesticide resistance in Louisiana mosquitoes has never been examined on a state-wide basis. In 1996, first component of this monitoring program examined selected pesticide efficacies and documented current pesticide resistance. Ground ULV-applied FyfanonRTM (96.8% malathion and Pyrocide RTM (96.8% + 1% natural pyrethrum) were applied to caged Culex quinquefasciatus adults (Denham Springs [Livingston Parish] and Lake Charles [Calcasieu Parish]) and Aedes sollicitans (Vermilion Parish). FyfanonRTM caused significantly different (p < 0.05) mortalities among each species and strain within 24hrs (95% Aedes, 48% Denham Springs, and 14% Lake Charles). Pyrocide RTM also produced significantly different mortalities (99%, 70%, and 38%, respectively). Malathion and permethrin were topically applied to 3--5 day old adult mosquitoes from 14 parishes. St. Martin parish Cx. quinquefasciatus was most susceptible to malathion (2.58 mug/mul), and the Vermilion strain was most resistant (417.05 mug/mul). Resistance is based on a ≥5-fold resistance ratio. Calcasieu parish strain was least resistant to permethrin (1.93 mug/mul), and the St. Tammany parish strain was most resistant (40.86 mug/mul). Detecting pesticide resistance by bottle bioassay technique was compared to topical bioassay. Both techniques were effective in detecting resistance for selected Aedes, Anopheles, and Culex species in Louisiana. Ground ULV 97% malathion was applied to caged Cx. quinquefasciatus adults from a mosquito abatement district, agrichemical farmland, and a susceptible colony. No significance (p > 0.05) existed in mosquito mortality between the agrichemical area and the abatement district. Social factors are associated with a wide variety of health outcomes. Social epidemiology has successfully used the traditional methods of surveillance and description to establish consistent relations between social factors and health status. Epidemiology as an etiologic science, however, has been largely ineffective in moving toward causal explanations for these observed patterns. Using the counterfactual approach to causal inference, the authors describe several fundamental problems that often arise when researchers seek to infer explanatory mechanisms from data on social factors. Contrasts that form standard causal effect estimates require implicit unobserved (counterfactual) quantities, because observational data provide only one exposure state …

Human immunodeficiency virus type 1 (HIV-1) infection is dependent on susceptible host cells that express both CD4 and chemokine co-receptors. The co-receptor CCR5 is associated with primary infection by macrophage-tropic virus isolates, whereas CXCR4 is commonly associated with T cell- and dual- tropic viruses. Once infected, lymphocytes and macrophages may replicate HIV- 1 or harbor latent virus, depending on environmental factors and cellular activation. Immune activation is often associated with viremia, which is consistent with enhanced infection and viral replication in activated cells harboring virus. Bartonella henselae is the causative agent of human cat scratch disease as well as several serious sequelae of infections, including bacillary angiomatosis and bacillary peliosis. Conflicting reports describe the pathogenesis of B. henselae in the cat. In this study, we characterized a strain of B. henselae termed LSU16. This strain was isolated on rabbit blood agar from a naturally infected 10-month-old female cat during a recurrent episode of bacteremia. The bacterial species was confirmed by PCR- restriction fragment length polymorphism analysis. The eastern oyster, Crassostrea virginica, supports a valuable industry in the United States and Louisiana. In recent years this industry has been plagued by diseases, creating interest in the use of gene transfer to produce oysters capable of eliminating pathogens. The goal of this work was to develop techniques for laboratory research of eastern oysters, and for transfer of genes to oyster gametes, larvae, and adults. Cell culture provides an effective alternative to work in whole animals. Food, which ensures the sustenance of life, has also been implicated in the development or prevention of numerous diseases including cancer. Cancer may be a result of hereditary factors or genomic instability induced by DNA damage, from the intrinsic chemistry of cells or extrinsic factors like aflatoxin B1. Aflatoxin B1, produced by Aspergillus flavus and Aspergillus parasiticus, is a common contaminant of corn and cottonseed and a proven mutagen and animal carcinogen. Since the first European settlement was established in the middle of Indian territory in 1714, the Red River Valley had experienced a dramatic transformation until the end of the antebellum period. The trajectory of the evolution can be divided into five vertical cross sections. The formative phase (1714--1803) features the colonial regimes in political arena, less-specialized regional economies, nascent urban units laden heavily with defensive functions, and coerced labor system. The settlement system resembled a semi-primate pattern with Natchitoches on the summit. The period of the Territory of Orleans (1804--1812) corresponded to the transition phase. Infection with HIV-1 results in pronounced immune suppression and susceptibility to opportunistic infections (OI). Reciprocally, OI augment HIV-1 replication. As we have shown for Mycobacterium avium complex (MAC) and Pneumocystis carinii, macrophages infected with opportunistic pathogens and within lymphoid tissues containing OI, exhibit striking levels of vital replication. Our knowledge of the disease burden components of tropical populations is fragmentary. Historically, the infectious diseases have been emphasized but, as some populations have undergone socio-economic changes, vital statistics have described a change in the pattern of disease. The picture is of a decline in infectious and a rise in chronic non-communicable disease. We focus here on the emergence of chronic cardiovascular diseases, and use hypertension as the paradigmic example. Early blood pressure surveys showed a virtual absence of hypertension among rural Africans and moderate prevalences in the Caribbean. Prevalence was highest among US and UK blacks. In a recent …

Studies have shown that individuals with Alzheimer's disease have a greater number of automobile crashes than normal elderly controls. Assessment of driving ability is usually conducted by use of an on-the-road examination. These examinations are costly, time intensive, and sometimes dangerous. Finding other measures that are predictive of driving ability will enable screening of patients to decrease the number of on-road examinations. Alzheimer's disease patients and normal elderly control subjects were administered neuropsychological measures as well as the Driver Performance Test (DPT) and Driver Risk Index (DRI), both videotaped tests of driving knowledge and risk assessment.
