Bacillus anthracis is a bacterial pathogen of great importance, both historically and in the present. This study presents data collected from several investigations and indicates that B. anthracis virulence is associated with the clonality and virulence of plasmids pXO1 and pXO2. Guinea pigs vaccinated with Anthrax Vaccine Adsorbed were challenged with 20 B. anthracis isolates representative of worldwide genetic diversity. These same isolates were characterized with respect to plasmid copy number by using a novel method of quantitative PCR developed for rapid and efficient detection of B. anthracis from environmental samples. Most spontaneously occurring mutations that cause extensive herpes simplex virus type 1 (HSV-1)-induced cell fusion are single amino acid changes within glycoprotein K (gK). Despite the strong genetic association of gK with virus-induced cell fusion, its direct involvement in cellular membrane fusion has been controversial, largely due to previously unsuccessful efforts to detect gK expression on virion and cellular surfaces. Clinical, serological, and pathological abnormalities observed in Holstein cows naturally infected with bovine lentivirus 1 bovine immunodeficiency virus (BIV) and other infections were progressive and most commonly associated with weight loss, lymphoid system deficiency, and behavioral changes. Clinical evidence of meningoencephalitis was dullness, stupor, and occasional head or nose pressing postures. The polymerase chain reactions associated the BIV provirus with the lesions in the central nervous system and lymphoid tissues. Multiple concurrent infections developed in retrovirally infected cows undergoing normal stresses associated with parturition and lactation. An energy balance was developed for heated and unheated earthen aquaculture ponds to 1) determine the relative importance of energy transfer mechanism affecting pond temperature; 2) predict pond temperatures; 3) estimate the energy required to control pond temperatures, and 4) recommend efficient heating and cooling methods. PHATR (Pond Heating and Temperature Regulation), a computer program using 4th order Runge-Kutta numerical method was developed to solve the energy balance using weather, flow rate and pond temperature data. By comparing measured and modeled pond temperatures, the average difference (the average bias) was 0.5°C for unheated ponds and 2.4°C for heated ponds. The …

Historians typically agree that reconciliation between the white North and South took place between the period of 1898 (Spanish-American War) and 1913 (before World War I). To test this hypothesis and identify when reconciliation took place in Baton Rouge, Louisiana, I will use the burial of R. L. Pruyn in the Baton Rouge National Cemetery. Pruyn served as a U.S. soldier during the Mexican War and a Confederate soldier during the Civil War. Forty-one (10 primiparous and 31 multiparous) Holstein cows were used in an experiment to determine the effects of prepartum dietary energy (PDE) and supplemental Ca-propionate on the incidence of ketosis in transition dairy cows and to evaluate the usefulness of urine pH as a predictor for ketosis. Treatment factors were prepartum dietary energy concentration (normal vs. high) with or without 113.5g Ca-Propionate/day (Nutro Cal®, Kemin Industries, Inc., Des Moines, IA). All cows were fed the same basal diet postpartum. Ca-propionate supplementation continued postpartum. Cows were individually fed and dry matter intakes (DMI) were measured daily. Vaccination programs are designed to protect an animal from infection, however, depending upon the age and health of the animal vaccination may not stimulate a protective humoral response. It is possible that, as in the human and mouse models, geriatric equines may be less responsive than their younger counterparts to current vaccination protocols. In the Maya area, archaeological excavations have yielded an abundance of skeletal material from a multitude of sites throughout Belize, Guatemala, Mexico, El Salvador and Honduras. In 1979, a series of excavations led by Heather McKillop at Moho Cay, Belize, revealed a number of human interments. This thesis analyzes the Moho Cay skeletal collection composed of remains from eight discrete Late Classic (A.D. 600-800) burials and nine other archaeological units. Skeletal and dental analysis of the remains included assessing minimum number of individuals per burial and establishing age and sex for each individual. The bones were also examined for signs …

To study the protective responses of cyathostome-infected ponies, two challenges were performed employing animals with different histories of exposure to these parasites. The hypothesis developed and to be tested in these experiments was that ponies that had longer exposure to cyathostome contaminated pastures would express acquired resistance to infection. The assumption behind this hypothesis was that helminth-naïve ponies infected with cyathostomes would eliminate the infection using only innate immune responses. Whereas previously exposed ponies would eliminate the infection with acquired immune responses, and these would be more effective in ponies with longer exposure to cyathostomes. Inflammation is a component of the innate immune response. However, severe or prolonged inflammation can be detrimental. Dietary n-3 fatty acid supplementation has proven clinical benefits in chronic inflammatory diseases, most likely due to reduced synthesis of inflammatory mediators and inhibition of lymphocyte proliferation and function. The purposes of this study were to characterize alterations in inflammatory mediator production, and lymphocyte proliferation, in dogs fed a diet consisting of 0.65% n-3 fatty acids (DMB) with an n-6:n-3 fatty acid ratio of 3.4:1. Shiga-toxigenic Escherichia coli (STEC) O157:H7 is an important human pathogen. Dairy cattle, beef cattle, and white-tailed deer (WTD) are involved in STEC O157:H7 transmission to humans. We conducted cross-sectional studies in Louisiana, using sensitive microbiological methods, in dairy cattle, beef cattle, and WTD to investigate the epidemiology of STEC O157:H7. Results of the dairy studies are as follows. In the point prevalence study, summer-time prevalence in herds was 38.5%, with a cow-level prevalence of 6.5%. Among positive herds, cow-level prevalence ranged from 3%-34.6%. Three of five herds sampled in the longitudinal study were positive. Cow-level prevalence increased during spring and …

Louisiana Gulf Coast oysters (Crassostrea virginica) were subjected to thermal shucking treatments to effect adductor muscle release from both left and right valves. The oysters were instrumented with thermocouples to monitor and record process temperatures in the oysters and on the shell. Following treatment, the oysters were evaluated for relaxation and release of the adductor muscle, meat quality and texture, and for the effect of the treatments on the storage life of the oysters as measured by total microbial plate counts. Psychosocial factors are thought to influence health through primarily direct physiological mechanisms or the alteration of health related behaviors. Three factors hypothesized to negatively impact health include arousal, life stress, and depressive symptomatology. One recent theorist suggests that the interaction between psychological stress and stress hormones on the neuroendocrine system may result in adverse changes to body composition, most notably the increased deposition of visceral adipose tissue (Bjorntorp, 1993). It is important to be able to predict the potential spread of water borne diseases when building dams or redirecting rivers. This study was designed to test whether the use of a growing degree day (GDD) climate model and remotely sensed data (RS) within a geographic information system (GIS), could be used to predict both the distribution and severity of Schistosoma haematobium. Streptococcus iniae causes meningoencephalitis and death in cultured fish species and soft-tissue infection in humans. We recently reported that S. iniae is responsible for local tissue necrosis and bacteremia in a murine subcutaneous infection model. The ability to cause bacteremia in this model is associated with a genetic profile unique to strains responsible for disease in fish and humans (J. D. Fuller, D. J. Bast, V. Nizet, D. E. Low, and J. The genus Brucella consists of bacterial pathogens that cause brucellosis, a major zoonotic disease characterized by undulant fever and neurological disorders in humans. Among the different Brucella species, Brucella melitensis is considered the most virulent. Despite successful use in animals, the vaccine strains remain infectious for humans. To understand the mechanism of virulence in B. melitensis, the proteome of vaccine strain Rev 1 was analyzed by two-dimensional gel electrophoresis and compared to that of virulent strain 16M. At the beginning of the epidemic, HIV/AIDS was affecting mainly gay men. As the disease began to spread, more diverse populations were affected. The experiment was conducted at Southeast Research Station in Franklinton, Louisiana between March and June, 2001. The primary purpose of this study was to determine the relationships between selected institutional and personal demographic factors, knowledge, observation of environmental risk factors, and the degree of nursing student compliance with standard and transmission-based precautions for the prevention of transmission of infectious diseases. A four part, 89 item questionnaire was administered to students enrolled in clinical courses at three associate degree and four baccalaureate nursing programs in order to measure knowledge, compliance, environmental risk factors, and demographics. This dissertation focuses on Lavinia Lloyd Dock's (1858-1956) re-envisioning of nursing and caring as social responsibility and the implications of this conceptualization for democracy. Dock was an American nurse, educator, settlement worker, suffragist, pacifist, social activist, writer, and historian. Her conception of holistic welfare embodied a 'new ideal' of society (Dock 1907, p. 899), a new understanding of democracy, and an expression of citizenship based on social responsibility for the welfare of others. Dock's idea of democracy embraced women's values and ways of being in the world; disputed universal, individual rights; and privileged communal values, collaboration, inclusion, and diversity. A series of experiments were conducted to produce transgenic goats by somatic cell nuclear transfer (NT). In all experiments, donor cells were electrically fused to enucleated metaphase II oocytes, then chemically activated. In a preliminary study to evaluate embryonic development following NT with proliferating or quiescent fibroblast or cumulus cells, significantly more embryos reconstructed with quiescent cumulus cells fused (77%) compared with proliferating cumulus cells (41%), proliferating fibroblasts (36%) or quiescent fibroblasts (37%). Improved development to the eight- to sixteen-cell stage was observed when fibroblast cells were serum starved (serum starved 39% vs. serum fed 15%). However, there was no …

Substantial funding has established the Mississippi Sandhill Crane National Wildlife Refuge in Southern Mississippi and an artificial insemination / breeding facility at the Audubon Center for Research of Endangered Species in New Orleans, Louisiana to promote species recovery. To facilitate detection of glycoprotein K (gK) specified by herpes simplex virus, a 12-amino-acid epitope tag was inserted within gK domain III. Recombinant virus gKprotC-DIII, expressing the tagged gK, was isolated. This virus formed wild-type plaques and replicated as efficiently as the wild-type KOS virus in Vero cells. Anti-protein C MAb detected high-mannose and Golgi complex-dependent glycosylated gK within cells as well as on purified virions. An outbreak of Fusobacterium necrophorum-induced septicemia occurred in a group of 40 captive wild-caught pronghorns (Antilocapra americana). Primary pododermatitis or necrotic stomatitis progressed to produce fatal septicemia with metastatic lesions in the forestomachs, lung, liver, and cecum in 38 of the animals. Two remaining animals were euthanatized because of chronic pododermatitis. Housing the animals in a pasture previously used by bovids and heavy rains with persistence of ground water pools in the pasture were contributing factors in the pathogenesis of this outbreak.

Previous studies from our laboratory indicated that a 28-amino-acid carboxyl-terminal truncation of gB caused extensive virus-induced cell fusion (Baghian et al., 1993, J Virol 67, 2396-2401). We tested the ability of additional truncations and mutations within gB to cause cell fusion in the recently established virus-free cell fusion assay (Turner et al., 1998, J. Virol. 72, 873-875). Deletion of the carboxyl-terminal 28 amino acids of gB (gBΔ28), which removed part of the predicted α-helical structure H17b, caused extensive cell fusion.
