Epigenetic Regulation Of Cancer Testis Antigens In Hepatocellular Carcinoma, Rinrada Klankajorn

Chulalongkorn University Theses and Dissertations (Chula ETD)

Hepatocellular carcinoma (HCC) represents one of the most common causes of cancer-related death worldwide, with few available treatment options. Cancer-testis antigens (CTAs), notably from the MAGE gene family, have opened new possibilities for improving immunotherapy in HCC due to their tumor-specific expression and high immunogenicity. However, the mechanisms regulating MAGE gene expression are largely unknown. This study explored mechanisms controlling the expression of MAGE-A3 and MAGE-C2. We utilized HCC organoid cultures derived from patient HCC tissues known to represent cancer epigenetic status more accurately than cell lines to study the MAGE gene family expression patterns and promoter methylation patterns. Our …

The Impact Of N-Glycans On The Immune Response Of Plant-Produced Sars-Cov-2 Rbd-Fc Proteins, Theerakarn Srisangsung

Chulalongkorn University Theses and Dissertations (Chula ETD)

N-glycosylation is a crucial post-translational modification that significantly affects immune recognition. The plant-based platform, especially Nicotiana benthamiana, offers scalable and cost-effective production of therapeutic proteins with glycosylation patterns similar to mammalian cells. However, plant-specific N-glycans, such as α(1-3)-fucose and β(1-2)-xylose (XF), have the potential to trigger allergic reactions. This study investigates how various N-glycan structures on the RBD-Fc fusion protein influence the immune response in mice. The RBD-Fc fusion protein was used for vaccine formulation, produced in both wild-type (WT) and glycoengineered (∆XF) N. benthamiana plants. N-glycan analysis showed a predominance of mannose (Man) in RBD-Fc with an ER retention …

Gene Expression By Nanostring Rna Profiling Of Monocytic Myeloid-Derived Suppressor Cells In Knee Osteoarthritis, Varote Shotelersuk

Chulalongkorn University Theses and Dissertations (Chula ETD)

Osteoarthritis (OA) is a widespread degenerative joint disease marked by cartilage destruction. This study investigates the role of monocytic myeloid-derived suppressor cells (M-MDSCs) in OA, focusing on their gene expression profiles and their relationship with body mass index (BMI) in knee OA patients. Previous research has shown a positive correlation between M-MDSCs and BMI in OA. We explored the role of M-MDSCs derived from the infrapatellar fat pad (IPFP) in OA patients.Using NanoString technology, we examined the transcriptional profile of M-MDSCs from the IPFP of knee OA patients undergoing total knee replacement surgery, categorized into different BMI groups. Flow cytometry …
