Parenting is a demanding role that encompasses many responsibilities and challenges. When a child is diagnosed with autism spectrum disorder, there are additional demands on the parent's time, energy, and financial resources, which likely leads to increased levels of chronic stress for parents. This dissertation examines the differences in chronic stress levels between parents who have children with autism and parents who do not, using hair cortisol concentration (HCC) as a benchmark. Diagnostic tests can dramatically improve and save lives. Survival rates and patient outcomes related to cancer and other diseases can be improved when routine testing guides personalized treatment. Highly accurate laboratory tests exist for thousands of biomarkers, including those of interest in this dissertation, but cost and complexity limit access to life-saving lab tests in many cases throughout the world. It has been theorized that Quantum Dots (QD) can solve the temperature sensor issue with microfluidic devices. Quantum Dots are nano-sized semiconducting crystals with a radius of an excited electron-hole pair. When a QD is excited by a short wavelength light, the QD produces fluorescent light. As the temperature of the dot changes, the dot expands and shifts the fluorescent spectrum. Quantum Dots are small enough to be placed inside the microfluidic devices, and a spectrometer can measure the change in fluorescent data. Within each cell, genetic material is stored in the form of DNA. This DNA is compacted around proteins to form higher order structures called chromatin. The basic unit of chromatin is a histone protein octamer bound with DNA called a nucleosome. A nucleosome consists of eight core proteins, four pairs of histone proteins H2A, H2B, H3, and H4, wrapped by DNA. The DNA wraps around the histone protein roughly 1.7 times, helping facilitate the compaction of DNA within the nucleus of the cell. The location of nucleosomes makes genetic material that encodes various elements, such as promoters or enhancers, accessible …

Dictyostelium discoideum (Dd) cells are amoeba cells that feed on soil or plant leaf matter bacteria. They divide freely in the presence of food. When starved, cell division is ended, and cells transition from unicellular to multicellular organisms initiated by a chemoattractant, cyclic adenosine monophosphate (cAMP). This study intricately examines cellular sorting within the Dd slug, focusing on individual cell dynamics. Represented as oriented ellipsoids, these cells maintain constant volume through a viscoelastic structure. Applying force to specific semi-axes reveals predictable deformations, highlighting the relationship between force and deformation rate. Human Immunodeficiency Virus (HIV-1) is the causative agent for acquired immunodeficiency syndrome (AIDS). Different strains of HIV-1 have been linked to a long term non-progressor (LTNP) or rapid progressor (RP) form of disease depending on rate of AIDS progression. Viral protein R (Vpr) is a viral protein that has been linked to varying rates of AIDS progression depending on the sequence. In this work, the effects of two polymorphisms, a LTNP and a RP, on different cells are studied. Total knee arthroplasty (TKA) procedures are increasingly common in the US and globally. However, rehabilitation outcomes may be negatively affected by limited access to physical therapy and patients' tendency to not fully adhere to recommended exercises between therapy sessions. This study aims to develop a remote system to aid rehabilitation, building upon recent sensor development at Brigham Young University, providing physical therapy-like biofeedback during unsupervised at-home exercises. Acoustic tweezers have gained substantial interest in biology, engineering, and materials science for their label-free, precise, contactless, and programmable manipulation of small objects. However, acoustic tweezers cannot independently manipulate multiple microparticles simultaneously. This study introduces acousto-dielectric tweezers capable of independently manipulating multiple microparticles and precise control over intercellular distances and cyclical cell pairing and separation for detailed cell-cell interaction analysis. Our acousto-dielectric tweezers leverage the competition between acoustic radiation forces, generated by standing surface acoustic waves (SAWs), and dielectrophoretic (DEP) forces, induced by gradient electric fields. Eukaryotic cells are highly heterogeneous. These cells are arranged into different compartments, carrying out separate functions and facilitating biological processes. Proteins are the effector biomolecules targeted to subcellular locations that help fulfill specific tasks in living organisms. Spatial proteomics can help unravel molecularly how protein abundance and localization are altered in cells, which is not feasible in traditional bulk-scale proteomics. The refugee and newcomer population faces many challenges as they arrive in their host country. Many individuals experience varying degrees of trauma in their country of origin. Trauma can lead to poor mental health outcomes and poor adjustment in host countries. Refugee children in particular may experience post-traumatic stress disorder, depression, and anxiety as a result of traumatic experiences. This dissertation investigates the impact of honeybee introductions on native bee populations, evaluates the efficiency of traditional bee sampling and monitoring methods, and explores the use of drone technology for assessing floral resources on federal lands in the United States. Federal lands, managed for multiple uses, present a valuable opportunity for the summer pasturing of honeybees. These lands produce vast floral resources and are largely free from pesticides and other pollutants that could weaken honeybee populations. Congenital Heart Disease (CHD) is the most common birth defect, affecting approximately 1% of live births. This dissertation explores the genetic mechanisms underlying CHD, focusing on the T-box transcription factors Tbx2a and Tbx2b in zebrafish. Utilizing advanced bioinformatics techniques, we reconstructed the ancestral sequences of these genes to understand their evolutionary trajectory and functional divergence. Our analysis highlights the evolutionary conservation of the DNA-binding domain in tbx2a and tbx2b, suggesting its crucial role in maintaining correct gene expression patterns during development. Liquid microdroplet resonators provide an excellent tool for optical studies due to their innate smoothness and high quality factors, but they can be difficult to control. By using 3D printed mounts to support the droplets, we can obtain precise control over the droplet geometries and positions. We here present our work with oil, water, and ice microdroplets, as well as tools required to enable their study. We first present methods for creating 3D printed mounts for oil microdroplet resonators. The mounts enable precise positioning of the droplets relative to a tapered optical fiber. German abortion law is restrictive for a Western European nation of strong economic standing and with relatively liberal social policy. Additionally, Germany is one of few Western European nations without an embryopathy indication, which allows for exceptional abortions in cases of fetal defects. This thesis addresses the underrepresentation of indigenous Māori people in leadership positions and pursues the need to develop a Māori leadership framework aimed at empowering the next generation of indigenous people. Several frameworks have attempted to exhibit Māori leadership dynamics, each offering unique insights into the cultural and relational aspects of Māori leadership. However, existing frameworks often exhibit limitations in fully encapsulating the breadth of Māori leadership values and practices, necessitating the development of a comprehensive Māori leadership model independent of Western frameworks.
