The etiology of alcoholism continues to be investigated. Heredity and environment have both been implicated as causes; however, the reason some people drink more than others has not been determined by science. Twin and adoption studies in humans yield evidence for genetic factors in alcoholism. Studies of possible markers in humans and studies of laboratory animal models have produced further evidence for the genetic transmission of alcohol preference. This animal study was done to further investigate the effects of heredity and environment on preference for alcohol in laboratory rats. The present report describes the characteristics and regulatory properties of phosphatidylinositol (Ptdlns) 4-kinase activity in rat exocrine pancreas. The membrane associated Ptdlns 4-kinase displayed a broad pH profile with optimal activity at neutral to alkaline pH. Carbachol (CCh) elicits a concentration- and time-dependent increase in Ptdlns 4-kinase activity in homogenates derived from agonist-stimulated acini. This effect was blocked by N-methylscopolamine and mimicked by muscarine. The enzyme had an apparent Km for Ptdlns and ATP of 4 and 60 uM, respectively. CCh caused no discernible change in the Km for either Ptdlns or ATP, but did produce a modest increase in …

Extracellular vesicles (EVs), through their complex cargo, can reflect the state of their cell of origin and change the functions and phenotypes of other cells. These features indicate strong biomarker and therapeutic potential and have generated broad interest, as evidenced by the steady year‐on‐year increase in the numbers of scientific publications about EVs. Important advances have been made in EV metrology and in understanding and applying EV biology. However, hurdles remain to realising the potential of EVs in domains ranging from basic biology to clinical applications due to challenges in EV nomenclature, separation from non‐vesicular extracellular particles, characterisation and functional …

In September, 1992, plague (Yersinia pestis) antibody was found in the blood of a coyote (Canis latrans) (1:128 titer) and a badger (Taxidea taxus) (1:2048 titer) taken near a suspect black-tailed prairie dog (Cynomys ludovicianus) town in Sioux County. However, a 1989-1991 survey of 96 coyotes taken from nine Nebraska Panhandle counties demonstrated no plague antibodies. Passive hemagglutination tests of Nobuto blood-sampling paper proved useful to quickly survey a predator population that is distributed across a broad geographic area.

The purpose of this study was to investigate the effect of experimental alterations in relatively small amounts of excess weight on vertical jump height. The objective was to use the consequences of changes in body weight on vertical jump performance. The subjects were 19 Division I college volleyball players from the University of Northern Iowa. The mean value for age was 20.04 years, the mean height was 176.2 cm, the mean weight was 71.58 kg, and the mean percentage of body fat was 22.60% for all subjects. Epstein-Barr virus (EBV) encodes a protein, ZEBRA, which enables the virus to switch from a latent to a lytic life cycle. ZEBRA expression was studied in lymphoid cells harboring either standard virus or a mixture of standard and defective viruses. ZEBRA protein could not be detected in cells latently infected with standard EBV but was constitutively expressed in cells containing both defective and standard EBV genomes. Experiments indicated that regulation of transcription of the ZEBRA gene (BZLF1) is a pivotal event in the control of EBV replication. ZEBRA specific BZLF1 transcripts were spontaneously expressed in cells harboring defective virus but …

The landmark citation method is a new collection assessment method based on the citation record of a single landmark article. This citation record is developed by identifying sources which cite the landmark article. A bibliography, extracted from the citation record, is then used to complete an assessment of the collection. This method was developed and used to assess the biotechnology collection of the National Library of Medicine. The information gained from this study, in addition to demonstrating the technique, also provided insight into the evolution of the biotechnology literature.

The large T antigen encoded by simian virus 40 (SV40) plays essential roles in the infection of permissive cells, leading to production of progeny virions, and in the infection of nonpermissive cells, leading to malignant transformation. Primary mouse embryo fibroblasts (MEFs) are nonpermissive for SV40, and infection by wild-type SV40 leads to immortalization and transformation of a small percentage of infected cells. We examined the ability of an extensive set of mutants whose lesions affect SV40 large T antigen to immortalize MEFs. We found that immortalization activity was retained by all mutants whose lesions are located upstream of codon 346. …

Many immunization programs in developing countries arr now undergoing a period of rapid utilization. Innovative strategies, such as immunization holidays, are being tried. Political commitment and enthusiasm are high. There is a strong desire by health professionals to protect the lives of many children from the untimely death and/or disability resulting from infectious diseases preventable by vaccination.
Evaluation of an immunization program can benefit other elements of the health system, particularly those that are not amenable to objective evaluation as immunization. In this case, immunization can serve as an indicator of the overall performance of the health system. The problems …

Time courses for ingestion, retention and release via feces of microbial food was investigated using 2 bivalves with different feeding strategies, Potamocorbula amurensis, and Macoma balthica. The results showed 2 pathways for the uptake of food material in these clams. The first is represented by an initial label pulse in the feces. The second pathway operates over longer time periods. Inert 51Cr-labeled beads were used to determine time frames for these pathways. The first pathway, involving extracellular digestion and intestinal uptake, is relatively inefficient in the digestion of bacterial cells by P. amurensis but more efficient in …

In the course of developing strategies to obtain a mutation in the aspartate semialdehyde dehydrogenase (asd) gene of Mycobacterium smegmatis, an efficient transposon trap was constructed which may be generally useful for the identification of transposable elements in mycobacteria. A DNA fragment containing the asd gene was replaced with an aminoglycoside phosphotransferase gene (aph) to generate a Δasd::aph allele. Attempts to replace the wild-type asd gene with the Δasd::aph allele were unsuccessful, suggesting that this deletion was lethal to the growth of M. smegmatis. The plasmid, pYUB215, which …

The purpose of this study was to examine the relationships among variables identified as potentially significant during the transition to second time parenthood. The theoretical framework for this study was based on the Circumplex Model of family systems (Olson, Sprenkle & Russell, 1979; Olson, Russell & Sprenkle, 1983), and a five domain structural model of marital and family adaptation (Cowan & Cowan, 1988).
The convenience sample consisted of 49 Caucasian middle class couples all expecting their second child. This was a one group pretest-posttest design with the birth of the second child functioning as the independent variable. The dependent variables …

Every bulky lesion in DNA can potentially inhibit the Taq DNA polymerase and thereby decrease the amplification produced in the polymerase chain reaction. We investigated the feasibility of using this inhibition to quantify DNA lesions produced by the anticancer drug cisplatin. Products were detected by electrophoresis followed by ethidium bromide staining. Quantitation was obtained by including [32P]dCTP in the amplification reaction and subsequently assessing the incorporated radioactivity. Hamster genomic DNA was platinated in vitro to defined levels and amplified with primers that produce either a 150, 750 or 2,000 base pair fragment. The degree of inhibition of PCR …

Bacteria within the Mycobacterium avium complex are prominent in the environment and are a source of serious disseminated infections in patients with AIDS. Serovars of the M. avium complex are distinguished from all other mycobacteria and from one another by the presence of highly antigenic glycolipids, the glycopeptidolipids, on their surfaces. A genomic library of DNA from serovar 2 of the M. avium complex was constructed in the Escherichia coli-Mycobacterium shuttle cosmid, pYUB18, and used to clone and express in Mycobacterium smegmatis the genes responsible for the biosynthesis of the oligosaccharide segment of the M. avium serovar 2-specific glycopeptidolipid. Rhesus monkeys (Macaca mulatta) reared during the first year of life without social contact develop persistent stereotyped movements, self-directed behaviors, and psychosocial abnormalities, but neurobiological mechanisms underlying the behaviors of socially deprived (SD) monkeys are unknown. Monkeys were reared in total social deprivation for the first 9 months of life; control monkeys were reared socially (SR) with mothers and peers. Subjects were killed at 19-24 yr of age. Because the behaviors of SD monkeys are reminiscent of changes in striatal or amygdalar function, we used immunocytochemistry for substance P (SP), leutine-enkephalin (LENK), somatostatin, calbindin, and tyrosine hydroxylase (TH) to evaluate …

Glucocorticoids (GC) modulate immune function in a number of ways, including suppression of T cell proliferation and other IL-2-mediated T cell functions. These inhibitory effects are similar to those induced by transforming growth factor-beta 1 (TGF-beta 1), a cytokine with potent T cell inhibiting activities. We examined the hypothesis that GC effects may be at least partially achieved through modulation of the expression of the TGF-beta 1 gene in activated T cells. To investigate the role of peptide backbone conformation on the biological activity of chemotactic peptides, we synthesized a unique analog of N-formyl-Met-Leu-Phe-OH incorporating the C α,α disubstituted residue, dipropylglycine (Dpg) in place of Leu. The conformation of the stereochemically constrained Dpg analog was examined in the crystalline state by x-ray diffraction and in solution using NMR, IR, and CD methods. The secretagogue activity of the peptide on human neutrophils was determined and compared with that of a stereochemically constrained, folded type II β-turn analog incorporating 1-aminocyclohexanecarboxylic acid (Ac6c) at position 2 (f-Met- Ac6c -Phe-OMe), the …

Economic efficiency of beef production to weaning and to slaughter was estimated in three groups, different in milk available (low, medium, and high) to the calves but the same in growth potential. Efficiency was the ratio of income to expenses. Income was derived from cull cows and calves at weaning or carcasses of calves fed to slaughter. Feed and non-feed expenses for the cowherd and for calves to weaning or to slaughter were included. Efficiencies were estimated assuming observed reproductive rates and energy requirements for maintenance, as well as for equal reproductive rates and equal requirements for maintenance in the …
