Cryptococcus neoformans is an encapsulated yeast responsible for severe meningoencephalitis. The importance of epidemiological studies on cryptococcosis has increased since the beginning of the AIDS epidemic. C. neoformans exists in two varieties containing four serotypes, C. neoformans var. neoformans (serotypes A and D) and C. neoformans var. gattii (serotypes B and C). Locally C. neoformans var. neoformans has been associated with pigeon feces during those months having an average temperature of 64.2°F j(17.8°C) and above. Clinical and environmental isolates of C. neoformans obtained from regional hospitals and environmental samplings, respectively, have been grouped into their variety status utilizing canavanine-glycine-bromthymol blue …

Interleukin 4 (IL-4), insulin, and insulin-like growth factor I (IGF-I) efficiently induced DNA synthesis in the IL-3-dependent murine myeloid cell lines FDC-P1 and FDC-P2. Although these factors could not individually sustain long-term growth of these lines, a combination of IL-4 with either insulin or IGF-I did support continuous growth. The principal tyrosine-phosphorylated substrate observed in FDC cells stimulated with IL-4, previously designated 4PS, was of the same size (170 kDa) as the major substrate phosphorylated in response to insulin or IGF-I. These substrates had phosphopeptides of the same size when analyzed by digestion with Staphylococcus aureus V8 protease, and each …

The purpose of this study was to collect normal flexion and extension trunk strength values for a population of individuals between the ages of 50 and 70 years of age. Twelve female and eight male volunteers in generally good health were tested in a seated position using a Biodex isokinetic dynamometer. Each performed five reciprocal maximal efforts of concentric flexion-extension cycles at the three isokinetic speeds of 60, 120 and 180{dollar}\sp\circ{dollar}/s. Raw and body weight adjusted data for peak torque and total work were analyzed. Results suggest strength decreases with age and that extension strength is greater than flexion strength. …

Researchers have begun to employ social-skills training in efforts to change the sexual behavior of adolescents. However, despite the promise of social-skills training, little is known about how social skills are related to the sexual practices of adolescents. The present article reviews the current literature and proposes a conceptual framework for understanding the relations between social skills and sexual behavior of adolescents by (a)examining the relationship between sexual activity and social skills, (b) examining the development of sexual-interaction skills and deficits, (c) summarizing what is currently known about the relations between social skills and sexual behavior, (d) discussing current issues …

The core-binding factor (CBF) binds the conserved core motif in mammalian type C retrovirus enhancers. We analyzed the phosphate contacts made by CBF on the Moloney murine leukemia virus enhancer by ethylation interference assay. The phosphate contacts span 9 bp centered around the consensus core site. To examine the sequence preferences for CBF binding, we employed the technique of selected and amplified binding sequence footprinting (T. K. Blackwell and H. Weintraub, Science 250:1104-1110, 1990). The consensus binding site for CBF defined by selected and amplified binding sequence footprinting is PyGPyG GTPy.

Often referred to as "cot death" or "crib death," sudden infant death syndrome (SIDS) is the single leading cause of infant death in infants aged 28 days to 1 year of life. Although the exact etiology of SIDS remains unknown, epidemiological studies have identified risk factors associated with higher rates of SIDS. In this study, the relationship between sudden infant death syndrome and maternal education among black women who gave birth in the United States during 1983 was examined.
The population for this study consisted of black infants who were born in 1983 and died before their first birthday. Among …

Mammalian type C retrovirus enhancer factor 1 (MCREF-1) is a nuclear protein that binds several directly repeated sequences (CNGGN6CNGG) in the Moloney and Friend murine leukemia virus (MLV) enhancers (N. R. Manley, M. O'Connell, W. Sun, N. A. Speck, and N. Hopkins, J. Virol. 67:1967-1975, 1993). In this paper, we describe the partial purification of MCREF-1 from calf thymus nuclei and further characterize the binding properties of MCREF-1. MCREF-1 binds four sites in the Moloney MLV enhancer and three sites in the Friend MLV enhancer. Clonal expansion of antigen-reactive T lymphocytes is driven by the lymphokine interleukin 2 (IL-2). To further elucidate the mechanisms of IL-2 action, we have utilized a differential hybridization procedure to clone IL-2-induced immediate-early genes from an IL-2-stimulated human T-cell cDNA library. To increase the frequency of IL-2-induced transcripts represented in the library, the protein synthesis inhibitor cycloheximide was included during the 2-hr IL-2 stimulation to superinduce gene expression, and the uridine analogue 4-thiouridine was utilized to enable selective purification of newly synthesized transcripts. From the enriched library, we have isolated eight IL-2-induced genes, six of which represent previously unrecognized human …

Transtracheal jet ventilation (TTJV) using a percutaneously inserted intravenous (IV) catheter for the patient who cannot be ventilated or tracheally intubated or, using a jet stylet for changing endotracheal tubes (ETT) in patients for whom subsequent ventilation and/or tracheal reintubation may be difficult, are extremely valuable therapeutic options. The jet ventilation system must have a sufficiently high pressure-oxygen source to drive oxygen through noncompliant tubing and through relatively small IV catheters and/or jet stylets in order to achieve adequate ventilation and oxygenation. The purpose of this study was to determine the personal and professional characteristics of successful individuals who have had a positive influence on the development of public policy concerning the environment. Additionally, which characteristics are most likely to enhance success and determine which of these characteristics might contribute to efforts toward developing public environmental policy in the future were investigated. The discovery of a base-exchange modification in the first position of the anticodon of four transfer RNA (tRNA) isoacceptor families has been shown to be correlated with a decrease in the rate of cell division in several tumorigenic tissues. This fact led to the hypothesis that wobbling in the anticodon might be a control point wich mediates the rate of translation. A suitable method for separation of different tRNA isoacceptor families had to be created in order to generate a means for quantitatively measuring degrees of this base modification in tRNA. Most studies on the biological activity of selenium have focused on its role at the active site in the ubiquitous enzyme, glutathione peroxidase (GSH-Px) which catalyzes the removal of hydrogen peroxide and organic hydroperoxides (1). A connection between selenium deficiency and a wide range of diseases in animals and humans has been reviewed (2). Development of various forms of human cancer (3), failure of the immune system (4), as well as some cardiovascular diseases (5) have been reported or suspected to correlate with selenium deficiency. Organophosphorus compounds (OPs) comprise one of the major classes of pesticides in use today because they are highly effective and less toxic. Because of their extensive use, there is a need for monitoring OPs in water and vegetables. Gas chromatography (GC) is the usual method for OPs screening, but it is time-consuming and has the high cost of instrumentation. The goal of this project was to find an easy method which was sensitive and had less instrumentation required to test OPs.

Mice carrying the Tight skin (Tsk) mutation harbor a genomic duplication within the fibrillin-1 (Fbn 1) gene that results in a larger than normal in-frame Fbn 1 transcript. In this study, the consequences of the Tsk mutation for fibrillin-containing microfibrils have been examined. Dermal fibroblasts from Tsk/+ mice synthesized and secreted both normal fibrillin (approximately 330 kD) and the mutant oversized Tsk fibrillin-1 (approximately 450 kD) in comparable amounts, and Tsk fibrillin-1 was stably incorporated into cell layers. Immunohistochemical and ultrastructural analyses of normal and Tsk/+ mouse skin highlighted differences in the gross organization and distribution of microfibrillar arrays. Rotary …

Extending Continuous Versus Discontinuous Conditioned Stimuli Before Versus After Unconditioned Stimuli, Melody Albert, Sean Ricker, Rick A. Bevins, John J.B. Ayres

Conditioned suppression was used with rats to study the effects of extending conditioned stimuli (CSs) before versus after the delivery of unconditioned stimuli. These extensions are termed B and A extensions, respectively. Within-group designs were used to compare the effects of extending CSs when 2-min parts of those CSs were separated by temporal gaps of 6 min versus a separation of no gap. The results were as follows: (a) B extensions weakened conditioning more than did A extensions, with or without gaps; (b) under some conditions, this asymmetrical effect persisted with extended training; (c) gaps between 2-min parts of a …

Examination of the role of base-off cobalamin species (where the 5,6-dimethylbenzimidazole ligand coordinated to cobalt is detached by protonation of the imidazole nitrogen) in differentiation between homolytic and heterolytic cobalt-carbon bond cleavage mechanisms is a primary step in better understanding B12-dependent enzyme catalysis. X-ray absorption edge spectroscopy provides the first direct structural evidence of five-coordination in base-off adenosyl- and base-off methylcobalamin complexes at room temperature. Integration of 1s-3d pre-edge transitions of the base-off species reveals the dependence of coordination number on temperature. Gradual increases in 1s-3d transition intensities, as the temperature is increased from 180 K to 298 K, reflect …

The fusion of influenza virus (A/PR/8/34 strain) with PC-12 cells was monitored by a fluorescence assay, and the results were analyzed with a mass-action model which could explain and predict the kinetics of fusion. The model accounted explicitly for the reduction in the fusion rate constant upon exposure of the virus to low pH, either for the virus alone in suspension or for the virus bound to the cells. When the pH was lowered without previous viral attachment to cells, an optimal fusion activity was detected at pH 5.2. When the virus was prebound to the cells, however, reduction of …

This article reviews the literature on the long-term consequences of childhood physical abuse. Empirical research is discussed within 7 topic areas: aggressive and violent behavior, nonviolent criminal behavior, substance abuse, self-injurious and suicidal behavior, emotional problems, interpersonal problems, and academic and vocational difficulties. The studies reviewed involve primarily adult populations, although pertinent findings from literature on children and adolescents are briefly summarized. Some variables that affect the relation between physical abuse and long-term consequences are examined. These moderator variables include maltreatment characteristics, individual factors, family factors, and environmental factors. The article ends with suggestions for future research on long-term consequences …

The practice of medicine is inextricably entwined with the management of health care information system (HCIS). Major problems with HCIS are that it has not evolved from the needs of end users, and lacks of an organizational base^ to manage the HCIS in the health care environment. The object of this paper is to examine a direction to effectively support and enhance the health care activities through managing HCIS. An integrated focus is provided through a framework for analyzing the structure and infrastructure of HCIS in hospitals.
