From an activist's perspective, the 80's was very encouraging,-- alternatives gained acceptance, legitimacy and credibility within the toxicology, corporate and regulatory communities. And The Center for Alternatives to Animal Testing (CAAT) at Johns Hopkins University played a significant role in making this acceptance possible.
But these new technologies have yet to realize their full potential; the focus to date has been on development more than on bringing these technologies into routine usage. And the public, …

The purpose of this independent study is to define presently existing theories linking cigarette smoking and low back pain. It also provides a detailed description of nicotine and its physiological and psychological effects.
There are four theories outlined in this study which could serve as explanations for the correlation of cigarette smoking and low back pain. The first is the chronic cough associated with smoking. This cough increases mechanical stress including higher disc pressures and poor union and healing following spinal fusion surgery. The second theory deals with the vascular changes caused by nicotine which decreases vertebral blood flow and …

Formalized functional assessment, in the form of data that is uniform and complete, is desirable for objective and explicit documentation of functional improvement. A variety of functional assessment instruments are currently being utilized in the field of medical rehabilitation. However, there is not professional clarity as to which functional assessment instrument is optimal.
The concept of functional assessment is described. An overview of existing functional assessment instruments is presented. Criteria for evaluating the adequacy of functional assessment instruments is discussed.
There is no one best approach to assessing physical function. No instrument is perfect for all patients or all situations. …

A mysterious respiratory illness with high mortality was recently reported in the southwestern United States. Serologic studies implicated the hantaviruses, rodent-borne RNA viruses usually associated elsewhere in the world with hemorrhagic fever with renal syndrome. A genetic detection assay amplified hantavirus-specific DNA fragments from RNA extracted from the tissues of patients and deer mice (Peromyscus maniculatus) caught at or near patient residences. Nucleotide sequence analysis revealed the associated virus to be a new hantavirus and provided a direct genetic link between infection in patients and rodents.

The development of comprehensive theories regarding the determinants of
vulnerability toward drinking problems depends in part on longitudinal evidence
linking psychosocial precursors to clinically-relevant problem consequences. In
an investigation of some of the more promising psychosocial precursors of
problem vulnerability, we evaluated the long-term predictive effects of adolescent
cognitive motivations for alcohol use and sensation seeking on a wide variety of
adult drinking-problem consequences including driving while intoxicated (DWI).
Results indicated that the Cognitive Motivation factor was a significant, independent, nine-year predictor of a factor of Drinking-Problem Consequences. Over this same period, certain cognitive motivation and sensation seeking indicators independently …

Rapamycin-Induced Inhibition Of P34(Cdc2) Kinase Activation Is Associated With G1/S-Phase Growth Arrest In T Lymphocytes, W. G. Morice, Gregory J. Brunn, Gregory Wiederrecht, John Siekierka, R. T. Abraham

The macrolide rapamycin (RAP) is a potent inhibitor of interleukin-2 (IL- 2)-induced T-cell proliferation. Current models suggest that RAP, when complexed to its intracellular receptor, FK506-binding protein, interferes with an IL-2 receptor-coupled signaling pathway required for cell-cycle progression from G1- to S-phase. Here we show that RAP treatment inhibits the growth of an IL-2-dependent cytotoxic T-cell line, CTLL-2, in late G1- phase, just prior to entry of the cells into S-phase. In contrast, RAP- treated CTLL-2 cells retained the ability to respond to IL-2 with enhanced cytolytic activity, indicating that RAP was not a general suppressant of cellular responsiveness to …

Exploiting Structural Differences Among Heteroduplex Molecules To Simplify Genotyping The Dqa1 And Dqb1 Alleles In Human Lymphocyte Typing, Peter A. Zimmerman, Mary Carrington, Thomas B. Nutman

A novel approach to DNA probe hybridization and heteroduplex analysis, termed directed heteroduplex analysis (DHDA) is presented here to illustrate its utility in simplification of human lymphocyte antigen (HLA)- typing. By strategic labeling of single-stranded probe sequences, DHDA allows the identification of specific heteroduplex structures that contribute to the differentiation of DQAI and DQB1 alleles. Because of the high degree of polymorphism among major histocompatibility complex class 11 second exon sequences, this analysis of 50 different heteroduplex molecules provides evidence of the importance of unpaired bases and mismatched base pairs and their effect on heteroduplex electrophoretic-mobility differences. A critical review of current epidemiological trends and social science research demonstrates that there is an urgent need for expanding the range of female-controlled HIV prevention methods. Existing efforts to control the spread of HIV infection primarily through the encouragement of a reduction in the number of sexual partners, widespread condom promotion, and the control of other sexually transmitted infections are inadequate for many of the world's women. The barriers to women's participation as subjects in biomedical research are currently being challenged as a matter of legislative policy, medicine, and law. This Article catalogs the ways in which women have been disadvantaged by their exclusion and recent developments to redress them, and goes on to dissect the underlying rationales for excluding women from clinical trials. The author reveals the 'fundamental misconception' behind exclusionary rationales, and argues that research sponsors in fact have more to fear in the way of potential liability from the exclusion of women, even pregnant women and women of child-bearing capacity, than from their inclusion. …

Research was conducted to determine if there was a correlation between the ability of persons with multiple sclerosis (MS) to cope with their disease and several other variables which could be related to coping skills. More than two-thirds of the 98 subjects in the sample regularly attended MS support groups . The ability to cope was measured by a scale developed by Brooks & Matson (1982). The independent variables were : self-esteem factors, vulnerability to stress, depression, severity of disability, general health factors, length of illness and age. Stroke is the number one source of adult disability in the United States, with approximately 400,000 new stroke survivors each year. The cost of care and the loss of earnings as a result of stroke is considerable. Stroke rehabilitation is a health service provided to try to decrease the disability and the socioeconomic costs. Low back pain has an enormous socioeconomic impact in this country. Even with advancement in diagnostic technology, the incidence and severity of low back pain continues to increase. The intervertebral disc plays an important causative role in the production of low back pain. The intervertebral disc may cause direct discogenic pain by mechanical and/or chemical irritation of the nociceptor receptors found within the outer one-third of the annulus fibrosis, or cause back pain by an indirect method. This thesis examines the compulsory smallpox vaccination controversy in Utah, 1899-1901. It looks at the two smallpox epidemics during 1899-1901 and follows the boards of health attempts to eradicate smallpox primarily by compelling the vaccination of school children.
Dr. Theodore B. Beatty, secretary of the State Board of Health, championed the effort to vaccinate all Utahns; however, the opposition led by Charles W. Penrose, editor of the Deseret Evening News, produced anti-compulsion and vaccination information which influenced Utahns to generally oppose vaccination. Consequently, the legislature passed an anti-compulsory vaccination statute over the governor's veto to annul the courts decision …

Photodynamic Therapy (PDT) has emerged as a useful cancer treatment modality which utilizes a tumor localizing dye and activating light to selectively destroy neoplastic tissue. In an effort to understand the newly synthesized photosensitizers, we are studying them in a mouse tumor model grown on the dorsal side of the foot by in vivo magnetic resonance techniques. We have synthesized several photosensitizers which are specifically labeled with fluorine. Several coils appropriate for the tumor study by 19F NMR were designed and constructed for this project. Matching alcoholics to appropriate intervention methods increases the likelihood of successful treatment. In order to better accomplish this, researchers have sought to identify distinct types of alcoholism. One promising approach is the Type I/Type II model of alcoholism. Type I alcoholism is characterized by a later onset and less severe social complications. It seems to be inherited from either natural parent and can affect children of either sex; it seldom occurs without the presence of a stressful environment. Type II alcoholism is distinguished by early onset and greater severity of consequences. It appears to be inherited only from father to …
