Dietary Fish Oil Sensitizes A549 Lung Xenografts To Doxorubicin Chemotherapy, W. Elaine Hardman, Mary Pat Moyer, Ivan L. Cameron

A549 xenografts were allowed to grow in nude mice to about 5 mm in diameter, then diets were changed to modified AIN-76 diets containing 19% wt/wt. fish oil (FO) or 20% wt./wt. com oil (CO). Ten days later dietary ferric citrate (0.3% wt./dry wt.) was added and doxoribicin (DOX) treatment (3.6 mg/kg i.v. each of the 5 days for 18 days) commenced. Treatment with DOX halted the growth of tumors in the CO fed mice. However, in those mice, which consumed FO or FO with ferric citrate, treatment with DOX caused significant tumor regression.

Resistance to antimicrobial agents used to treat severe Campylobacter spp. gastroenteritis is increasing worldwide. We assessed the antimicrobial resistance patterns of Campylobacter spp. isolates of human and animal origin. More than half (n = 32) were resistant to sulphonamide, a feature known to be associated with the presence of integrons. Analysis of these integrons will further our understanding of Campylobacter spp. epidemiology.

Under free running conditions, FREQUENCY (FRQ) protein, a central component of the Neurospora circadian clock, is progressively phosphorylated, becoming highly phosphorylated before its degradation late in the circadian day. To understand the biological function of FRQ phosphorylation, kinase inhibitors were used to block FRQ phosphorylation in vivo and the effects on FRQ and the clock observed. 6-dimethylaminopurine (a general kinase inhibitor) is able to block FRQ phosphorylation in vivo, reducing the rate of phosphorylation and the degradation of FRQ and lengthening the period of the clock in a dose-dependent manner. Galactokinase deficiency is an autosomal-recessive inborn error of galactose metabolism whose major clinical manifestation is the development of cataracts during the first months of life. This metabolic disorder is caused by defects in the first enzyme of the Leloir pathway, galactokinase, encoded by the gene GALK1 on chromosome 17q24. Despite the identification of a number of conserved domains in GALK1, understanding of the functional significance of these regions and the molecular basis of the disorder is limited. This is largely due to the rarity of the disease and the fact that the small number of GALK1 mutations identified to-date are …

The transition from a planktonic (free-swimming) existence to growth attached to a surface in a biofilm occurs in response to environmental factors, including the availability of nutrients. We show that the catabolite repression control (Crc) protein, which plays a role in the regulation of carbon metabolism, is necessary for biofilm formation in Pseudomonas aeruginosa. Using phase-contrast microscopy, we found that a crc mutant only makes a dispersed monolayer of cells on a plastic surface but does not develop the dense monolayer punctuated by microcolonies typical of the wild-type strain. Flavonols are polyphenols, secondary plant metabolites commonly found in plants and foods of plant origin. They have widespread biological properties in the human body. The recent discovery of their potential antioxidant activities has prompted extensive research. Flavonols particularly quercetin are potent antioxidants with higher antioxidant properties than the well known antioxidant vitamins C and E. Several epidemiology studies have demonstrated a strong inverse association between flavonoid intake and risk of coronary heart disease. The association with cancer is less defined with only some studies showing an inverse association and others not. In view of their potential to act as antioxidants …

At present, toxoplasmosis is one of the most common opportunistic infections in immunocompromised patients. The need for a reliable experimental model is crucial not only for achieving a better understanding of the pathophysiology of this infection but also for developing a better method to evaluate new therapeutic regimens. An animal model for the study of articular cartilage regeneration in-vivo facilitated by stress-shielding is introduced. The object of the model is to test the hypothesis that some form of cartilaginous tissue will grow upon a large joint surface in vivo with the joint in normal motion. The model utilizes the known capability of immature cells to differentiate. The source of cells is bleeding subchondral bone. In addition, the model provides a mechanically shielded environment in which cell differentiation and maturation can occur. The neurogenetic prototypic disease on which we chose to test our gene therapy strategy is Canavan disease (CD). CD is an autosomal recessive leukodystrophy associated with spongiform degeneration of the brain. At present the disease is uniformly fatal in affected probands. CD is characterized by mutations in the aspartoacylase (ASPA) gene, resulting in loss of enzyme activity. In this review, recent evidence is summarized on the etiology and possible treatments for CD. In particular, we discuss two gene delivery systems representing recent advances in both viral and liposome technology: a novel cationic liposome-polymer-DNA (LPD) complex, DCChol/DOPE-protamine, as well as recombinant …

Transgenic mice express increased levels of neurotrophin-3 (NT-3) in epithelium when their ancestors are microinjected with the NT-3 gene. The NT-3 growth factor expressing transgenic mice are useful in the study of neurodegenerative disorders of the brain such as Parkinson's syndrome and Alzheimer's disease, of the spinal cord motor neurons such as amyotrophic lateral sclerosis, and for testing drug candidates for the treatment of these diseases.

Cardiovascular disease is the leading cause of death in the industrialized nations, and accounts for 1 million deaths in the United States each year (McArdle, Katch & Katch, 1991). One of the major risk factors for cardiovascular disease is physical inactivity, which is a behavioral modified risk factor. Physical inactivity plagues 59% of the people in the United States (ACSM Resource Manual, 1998). Physical inactivity often leads to obesity, diabetes, hypertension and hypercholemia, and places sedentary individuals at more risk for cardiovascular disease (CVD), or coronary heart disease (CHD). Eating disorders, in the form of anorexia nervosa and bulimia nervosa, have been recognized as significant mental-health issues for the last three decades, and the incidence is rising as we approach the new millennium. Currently, many women who come into a mental-health setting due to depression, anxiety, low self-esteem, relationship issues, sexual issues, etc., are also struggling with eating-disordered behaviors, thoughts, and feelings. These behaviors, thoughts, and feelings may remain well hidden from the counselor throughout the course of therapy or until they become severe and more difficult to treat. Conjugated bilirubin is an important but difficult metabolite to measure accurately, elevated levels of conjugated bilirubin in the serum are indicative of obstructive jaundice and therefore can be used to discriminate this disorder from other forms of unconjugated hyper bilirubinaemia.

Purpose: The aim of this study was to determine the risk factors in the development of total parenteral nutrition associated cholestasis which is a significant clinical problem for pediatric surgeons. Methods: The medical records of 54 neonates who had received total parenteral nutrition for more than two weeks were reviewed retros pectively. Prematurity, duration of parenteral nutrition, enteral starvation, composition of the parenteral nutrition solution, the number of septic episodes and the presence of jejunostomy were evaluated as risk factors. Brochopulmonary infection in cystic fibrosis (CF) patients is associated with chronic progressive lung disease. The role of host factors, such as neutrophil functions, in the progressive pathologic processes in the respiratory tract of these patients, is poorly defined. Neutrophil chemotaxis is one of the first mechanisms of host defense to become activated after bacterial invasion has occurred. This study was aimed to evaluate the role of neutrophil chemotaxis in CF and also to determine whether an acute bacterial infection and the nutritional status can affect neutrophil chemotaxis or not. Sporulation is a developmental process in which temporally distinct sets of genes regulate the processes of meiosis and ascospore formation. Sporulation-specific genes are classified into early, middle, and late genes based on their peak time of expression during sporulation. Regulation of two late genes, SPR1 and SPR2, was studied. The function of SPR2 is unknown. Its regulatory region contains two Mid Sporulation Elements (MSE) repeated in tandem that are sufficient to confer sporulation-specific regulation to a CYCl-lacZ reporter. These sequences have UAS (upstream activation sequence) properties, as each 14 base pair element is sufficient to confer sporulation-specific regulation …
