15 healthy sedentary men, 19-25 years old an did not have any programmed physical activity, were studied. The subjects were asked to run submaximal 15-20 min every day for 5 weeks. Erythrocyte lipid peroxidation, superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) activities and glucose and uric acid levels were determined in blood samples which were taken before and immediately after acute exercise and after the end of 5-week training program. The roles of serum on the initial attachment and adhesion strength of CHL cells, which is gained following initial attachment, spreading and growth, on tissue culture polystyrene dish were determined by using a convergent Microflow chamber. In the presence of 10% fetal calf serum 98%±2% of cells were attaching within 2 hours at 37°C. While only 74%±3% of cells were attaching during the same period of incubation in serum free medium. QSR1 is an essential Saccharomyces cerevisiae gene, which encodes a 60S ribosomal subunit protein required for joining of 40S and 60S subunits. Truncations of QSR1 predicted to encode C-terminally truncated forms of Qsr1p do not substitute for QSR1 but do act as dominant negative mutations, inhibiting the growth of yeast when expressed from an inducible promoter. The dominant negative mutants exhibit a polysome profile characterized by 'half-mer' polysomes, indicative of a subunit joining defect like that seen in other qsr1 mutants (D. P. Eisinger, F. A. QSR1 is a recently discovered, essential Saccharomyces cerevisiae gene, which encodes a 60S ribosomal subunit protein. Thirty-one unique temperature-sensitive alleles of QSR1 were generated by regional codon randomization within a conserved 20-amino-acid sequence of the QSR1-encoded protein. The temperature-sensitive mutants arrest as viable, large, unbudded cells 24 to 48 h after a shift to 37 degrees C. Polysome and ribosomal subunit analysis by velocity gradient centrifugation of lysates from temperature-sensitive qsr1 mutants and from cells in which Qsr1p was depleted by down regulation of an inducible promoter revealed the presence of half-mer polysomes and a large pool of free 60S …

The present study was conducted to determine if vitamin E and selenium could have a positive synergistic effect during experimental Chagas' disease. In the first phase of the study, 25 female C3HeB/FeJ mice were infected with a Brazil strain of Trypanosoma cruzi while receiving vitamin E supplementation of 0 IU/kg, 200 IU/kg, 400 IU/kg, 600 IU/kg, 800 IU/kg in their diets. In the second phase, 20 female C3HeB/FeJ mice were infected while receiving 400 IU/kg vitamin E plus selenium supplementation of 2 ppm, 4 ppm, 8 ppm, 16 ppm in their drinking water. The intracellular mechanism(s) underlying the upregulation of the hepatic Na+/taurocholate cotransporting polypeptide (ntcp) by prolactin (PRL) are unknown. In this report, we demonstrate a time-dependent increase in nuclear translocation of phosphorylated liver Stat5 (a member of the ignal ransducers and ctivators of ranscription family) that correlated with suckling-induced increases in serum PRL levels. In electrophoretic mobility gel shift assays, nuclear Stat5 exhibited specific DNA-binding ability towards IFN-gamma-activated sequence (GAS)-like elements (GLEs; 5'TTC/A-PyNPu-G/TAA-3') located in the -937 to -904 bp region of the ntcp promoter. Transient cotransfections in HepG2 cells revealed that PRL inducibility (2.5-3-fold) required coexpression of the long form …

Ozone is a major component of photochemical smog. High levels of this pollutant, sufficient to affect human health are found in many urban areas worldwide. Though limited studies in humans are supported by extensive findings from animal experiments, a difficulty in interpreting the results of these experiments has lead to an ambiguity on the biochemical mechanism of ozone toxicity. To elucidate the mechanism by which ozone causes cell damage and eventual cell death we conducted a comprehensive study using Escherichia coli K-12 as a model.

Serum chemistry and hematological values from 220 traumatic spinal cord injured patients (157 male and 63 female between the ages of 15-47, with greater than six months from injury) were compiled via chart review. Traumatic spinal cord injured patient’s reference ranges were determined and compared with the general population reference ranges. Reference ranges within the spinal cord population were compared by age, gender, level of injury and chronicity of injury. The reference ranges determined for all groups within this spinal cord population fall within the reference ranges of the general population. Migraine headaches are a common and debilitating syndrome causing untold suffering and loss of productivity. A review of the literature indicates that there are two primary factors involved in triggering these headaches: high levels of blood lipids and high levels of free fatty acids. Under these conditions platelet aggregability is increased, which is associated with decreases in serotonin and heightened prostaglandins levels. These conditions may produce vasodilation, the most immediate precursor to migraine headache. A high fat diet is one factor that may directly affect this process. Aim: To test the hypothesis that a strategy including cholesterol screening and dietary education is more effective than dietary education alone in changing dietary behavior and serum cholesterol levels.
Methods: Individuals at four worksites were enrolled in a randomized trial with a 'full intervention' condition in which subjects were told their serum cholesterol value and also received a dietary change kit (n=236), and a 'partial intervention' condition in which subjects received the same dietary change kit, but were not told their serum cholesterol value (n=284). The early recruitment and use of the chemoreceptive senses, olfaction and gustation, are important for early behavior development in fathead minnows and the ability of larvae to survive after hatching. Chemoreception is used in the location of food, avoidance of predators, and for intraspecific communication. Furthermore, some studies have shown the olfactory system to be adversely impacted by environmental contaminants and low pH. Since the fathead minnow is a commercially raised baitfish in the aquaculture industry, and serves as a standard test organism for bioassays and reaction studies of aquatic contamination by the United States Environmental Protection Agency, an understanding …

Purple membrane (PM, λ" role="presentation">λmax" role="presentation">max 570 nm) of H. halobium on treatment with sulphuric acid changes its colour to blue (λ" role="presentation">λmax" role="presentation">max 608 nm). The purple chromophore can be regenerated from the blue chromophore by exogeneous addition of anions such as CI−" role="presentation">− and HPO42−" role="presentation">2−4. Chloride ion is found to be more effective than the dibasic phosphate ion in regenerating the purple chromophore. Nevertheless, one thing common to the anion regeneration is that both CI−" role="presentation">− and HPO42−" role="presentation">2−4 show marked pH effect. At pH 1.0 the efficiency of …

The Neurospora crassa eas (ccg-2) gene, which encodes a fungal hydrophobin, is transcriptionally regulated by the circadian clock. In addition, eas (ccg-2) is positively regulated by light and transcripts accumulate during asexual development. To sort out the basis of this complex regulation, deletion analyses of the eas (ccg-2) promoter were carried out to localize the cis-acting elements mediating clock, light, and developmental control. The primary sequence determinants of a positive activating clock element (ACE) were found to reside in a 45-bp region, just upstream from the TATA box. Dietary supplementation with increased levels of vitamin E can enhance the immune system of C3HeB/FeJ mice to enable many of them to survive an otherwise lethal dose of Trypanosoma cruzi, the causative agent of Chagas' disease. To study the role of diet on the course of experimental Chagas' disease, C3HeB/FeJ mice were fed pelleted, synthetic food containing no vitamin E and selenium, 800 IU/kg vitamin E, 2 ppm selenium, or food containing 800 IU/kg vitamin E plus 2 ppm selenium. The purpose of this investigation was to determine if there is a difference in the type of microorganisms found in oral sites classified as gingival health, mild-moderate periodontitis, or severe periodontitis as measured by the DMDx* Test. A purposive sample of one hundred oral sites was selected from adult patients. Arm autotomy in asteroids, as tail autotomy in vertebrates, typically occurs when the animal is attacked and appears to facilitate escape. One assumes autotomy has a cost, but it rarely has been demonstrated in the field in vertebrates and never demonstrated in asteroids. The concentration of lipid was 40 % less and the amount of kilojoules 85 % less in the pyloric caeca of the asteroid Stichaster striatus Muller & Troschel (Echinodermata: Asteroidea) with autotomized arms than in those of individuals with intact arms collected in the field. as the pyloric caeca are used as nutrient reserves, individuals with autotomized …

This study investigated the site of ATP utilization in the signal transduction pathway of bacterial chemotaxis and localized the point of convergence of a methylation-independent system of chemotaxis with the methylation-dependent system. The identity of the signal originating from the phosphotransferase system was investigated by substituting the fructose-inducible HPr-like protein FPr for HPr in transport and chemotaxis. In addition, a novel chemoattractant, glycerol, was identified for Salmonella typhimurium. Histidine-auxotrophic S. typhimurium strains ST23 (hisF) and ST171 (hisF cheB) were depleted for ATP. Reuptake plays an important role in regulating synaptic and extracellular concentrations of glutamate. Three glutamate transporters expressed in human motor cortex, termed EAAT1, EAAT2, and EAAT3 (for excitatory amino acid transporter), have been characterized by their molecular cloning and functional expression. Each EAAT subtype mRNA was found in all human brain regions analyzed. The most prominent regional variation in message content was in cerebellum where EAAT1 expression predominated. EAAT1 and EAAT3 mRNAs were also expressed in various non- nervous tissues, whereas expression of EAAT2 was largely restricted to brain.
