Aging is the main risk factor for cognitive decline. The hippocampus, a brain region critical for learning and memory formation, is especially vulnerable to normal and pathological age-related cognitive decline. Dysregulation of both insulin and intracellular Ca2+ signaling appear to coexist and their compromised actions may synergistically contribute to neuronal dysfunction with aging. This dissertation focused on the interaction between insulin, Ca2+ dysregulation, and cognition in hippocampal neurons by examining the contributions of insulin to Ca2+ signaling events that influence memory formation. I tested the hypothesis that insulin would increase cognition in aged animals by altering Ca2+-dependent physiological mechanisms involved …

Seasonally breeding animals reproduce during certain times of the year and, subsequently, behaviors, steroid hormone levels, and brain morphology change. The green anole lizard (Anolis carolinensis) is an excellent model to study the regulation of steroid hormone production because they have distinct hormonal and behavioral differences between sexes and seasons. As in other vertebrates, steroidogenesis in anoles is under the control of the hypothalamus-pituitary-gonadal (HPG) axis. We tested the hypothesis that natural variations in steroid hormone levels between sexes and seasons are mediated within the brain and gonad by examining four genes involved in steroidogenesis: StAR, Cyp17α1, HSD17β3, and Cyp19α1. …

Background Dietary nitrate has been shown to increase nitrate/nitrite levels in multiple populations, with potential blood pressure lowering effects. However, there are few reports among hypertensives. Aims We aimed to assess the effect of daily nitrate in subject with controlled hypertension vs. uncontrolled hypertension. Methods On day 0, hypertensives wore an ambulatory BP monitor (ABPM) for 24h and fasting blood was taken. Subjects then consumed concentrated beetroot juice (12.9mmol nitrate) for 14 consecutive days. On day 14 subjects consumed their last nitrate dose after fasting blood was drawn and again had an ABPM for 24h. Results According to baseline ABPM, …

Background: This study explored the possible role of FGF23 in pediatric hypercalciuria.
Methods: Plasma FGF23 was measured in 29 controls and 58 children and adolescents with hypercalciuria: 24 before treatment (Pre-Treated) and 34 after 6 months of treatment (Treated). Hypercalciuric patients also measured serum PTH hormone, 25(OH)vitD, phosphate, calcium, creatinine, and 24 h urine calcium, phosphate, and creatinine.
Results: There were no differences in age, gender, ethnicity, or body mass index either between controls and patients, or between Pre-Treated and Treated patients. Median plasma FGF23 in controls was 72 compared with all patients, 58 RU/mL (p = 0.0019). However, …

Whilst all military cohorts have been seen to experience both physiological and psychological stressors during occupational duties, the naval service experience different stressors due the prolonged periods of time spent at sea. Research has analysed some of these stressors however many them have focused on how individuals are affected because of time spent in combat zones. Background/aim: Micronutrients are indispensable in the prevention of diseases and maintenance of good health. Their deficiencies have been implicated in several diseases associated with oxidative stress (OS). This study aimed at investigating the levels of some vitamins and minerals in association with OS markers in diabetic foot ulcer (DFU) patients in Ibadan, Oyo State, Nigeria. Background/aim: We calculated the homeostatic model assessment (HOMA) for estimating insulin sensitivity and beta-cell function in normal, healthy nondiabetics with infections (malaria, influenza, HIV, Helicobacter pylori, Chlamydia pneumoniae, and hepatitis C virus), type 2 diabetic black patients, and healthy controls from Kinshasa, DR Congo. Materials and methods: A case-control study was carried out between 2006 and 2007 for black Central African participants managed for HOMA.Results: In total, 219 patients and 110 healthy controls were matched for sex and age. Not all women infected with chlamydiae develop upper genital tract disease, but the reason(s) for this remains undefined. Host genetics and hormonal changes associated with the menstrual cycle are possible explanations for variable infection outcomes. It is also possible that disease severity depends on the virulence of the chlamydial inoculum. It is likely that the inoculum contains multiple genetic variants, differing in virulence. If the virulent variants dominate, then the individual is more likely to develop severe disease. In December 2016, a panel of experts in microbiology, nutrition and clinical research was convened by the International Scientific Association for Probiotics and Prebiotics to review the definition and scope of prebiotics. Consistent with the original embodiment of prebiotics, but aware of the latest scientific and clinical developments, the panel updated the definition of a prebiotic: a substrate that is selectively utilized by host microorganisms conferring a health benefit. This definition expands the concept of prebiotics to possibly include non-carbohydrate substances, applications to body sites other than the gastrointestinal tract, and diverse categories other than food. Fifteen New Zealand rabbits were randomly assigned to one of 3 dietary treatment groups of 5 animals each and fed pelleted, extruded, or muesli diets in a completely randomized design experiment. Rabbits were placed in individual cages with ad libitum access to water and food for 45 days acclimation followed by 30 days experimental period. Feed intake of rabbits fed pelleted and extruded diets was greater (p < 0.05) than rabbits fed the muesli diet (125.6 and 130.4 vs. 91.9 g/d), but weight change and feed efficiency were not affected by treatment. Diet digestibility among the treatments was inconsistent when comparing results obtained from total fecal collection and AIA (please define) as an internal marker. Rabbits fed extruded and pelleted diets had lower (p < 0.05) cecal pH (6.42 and 6.38 vs. 7.02, respectively), and higher (p < 0.05) production of SCFA (18.5 and 19.0 vs. 11.7 mM, respectively) than those fed muesli. The fermentation products from rabbits fed pelleted and extruded diets had a greater proportion of butyrate and less propionate than rabbits fed muesli. The results of this study indicate that the basal dietary composition had a greater impact on diet utilization and cecal fermentation than food form.

PURPOSE: The purpose of this study is to evaluate the effects of enhanced ordering privileges among registered dieticians (RD) on oral nutritional supplement (ONS) ordering frequency and prevalence of oral nutritional supplement use among elderly patients admitted with heart failure (HF).
METHODS: This study was a single facility, retrospective pre-post implementation evaluation of the impact of expanded ONS ordering privileges among elderly patients (age ≥ 65 years old) hospitalized with heart failure at Frankfort Regional Medical Center (FRMC). The sample consisted of 105 patients for the pre-evaluation period and 259 for the post-evaluation period.

Type 1 Diabetes suppression in diabetes prone mice following immunization with Group A Streptococcus correlates with the B lymphocyte response to N-acetyl-D-glucosamine (GlcNAc) present in the Group A Carbohydrate cell wall polysaccharide. GlcNAc-specific B cells recognize cryptic GlcNAc-epitopes in beta cells. Following exposure of GlcNAc-modified antigens on beta cells during cellular apoptosis, GlcNAc-specific IgM antibodies opsonize these epitopes and mediate classical complement pathway activation. In vitro, opsonization of apoptotic beta cells with GlcNAc-specific IgM increases their uptake by dendritic cells, while suppressing the subsequent presentation of beta cell-derived antigens to diabetogenic T lymphocytes. Individuals with chronic poststroke hemiparesis (PS) exhibit increased variability in spatiotemporal kinematic variables related to endpoint control during walking. For nonimpaired individuals, walking faster or slower than a preferred walking speed, results in less consistent stepping behavior. In three studies I investigated variability in swing phase foot trajectory area on a step-by-step basis (SBS-FTA) in nonimpaired individuals and people with PS. Human cytosolic sulfotransferase 1C4 (hSULT1C4) is a dimeric Phase II drug-metabolizing enzyme primarily expressed in the developing fetus. SULTs facilitate the transfer of a hydrophilic sulfonate moiety from 3’-phosphoadenosine-5’-phosphosulfate (PAPS) onto an acceptor substrate altering the substrate’s biological activity and increasing the compound’s water solubility. While several of the hSULTs’ endogenous and xenobiotic substrates have been identified, the physiological function of hSULT1C4 remains unknown. Rodents have been the model of choice for decades in biomedical research due to their relatively high physiological similarity to humans and amenability to genomic modification. BACKGROUND: Asthma is a common childhood respiratory disease, affecting around 20% of Irish children. In other populations, vitamin D receptor (VDR) polymorphisms have been associated with asthma risk. We aimed to investigate the association between 2 VDR polymorphisms and uncontrolled paediatric asthma. METHODS: 44 asthmatic children and 57 healthy volunteers were studied. The VDR TaqI gene variant in exon 9 (T/C) (rs731236) and ApaI (rs7975232) in intron 8 (C/T) were determined, using TaqMan®Assays. Study Objectives: Our group and others have reported a high rate of vitamin D deficiency in obstructive sleep apnea (OSA), where vitamin D levels (25(OH) D) correlate negatively with OSA severity and some of its associated metabolic alterations. Klotho is an anti-aging protein that when upregulated, extends lifespan and enhances cognition. Klotho knockout mice age prematurely and die by ~8 weeks of life after developing cognitive impairment. Klotho function outside of the brain is well established but the protein’s action within the brain remains unknown. As klotho regulates peripheral stem cell populations, I sought to determine whether klotho contributes to cognition through a role in the maintenance of the adult hippocampal neurogenic niche. To investigate the effects of genistein on estrogen deficiency with high-fat high-fructose (HFHF) diet-induced NASH rats. Female Sprague-Dawley rats (n=48) were randomly divided into ovariectomized (OVX) and non-ovariectomized (non-OVX) group. ความสำคัญและที่มาของปัญหาการวิจัย: ผู้ป่วยฟอกเลือดนั้นมีภาวะเนือยนิ่งหรือการมีระดับกิจกรรมทางกายที่ต่ำ, กล้ามเนื้อลดลง (sarcopenia) และมีการคั่งของของเสียจากภาวะไตวาย (uremic toxins) ซึ่งสัมพันธ์กับอัตราตายจากโรคหัวใจเพิ่มสูงขึ้น การออกกำลังกายขณะฟอกเลือด (intradialytic exercise) อาจเพิ่มระดับกิจกรรมทางกายของผู้ป่วยฟอกเลือดได้ วัตถุประสงค์ของการวิจัย: เพื่อศึกษาว่าการออกกำลังกายขณะฟอกเลือดสามารถเพิ่มระดับกิจกรรมทางกายของผู้ป่วยฟอกเลือดได้หรือไม่ ระเบียบวิธีวิจัย: การศึกษานี้เป็นการศึกษาวิจัยแบบสุ่มที่มีกลุ่มควบคุมแบบเปิด (open-labelled randomized controlled trial) ในผู้ป่วยฟอกเลือดโดยวิธี online HDF โดยผู้ป่วยจะถูกสุ่มเป็น 2 กลุ่ม ได้แก่ กลุ่มที่ไม่ต้องออกกำลังกาย และ กลุ่มที่ออกกำลังกายขณะฟอกเลือด (intradialytic exercise, IDX) โดยใช้จักรยาน (cycle ergometer) และวัดระดับกิจกรรมทางกายในชีวิตประจำวันโดยใช้เครื่องสวมใส่บริเวณข้อมือ (wrist-worn wearable triaxial accelerometer (Fitbit flex2®)) วัดผลเลือด มวลกล้ามเนื้อ และ สมรรถภาพของร่างกาย ตอนเริ่มต้นและที่ 6 เดือน ผลการศึกษา: มีผู้ป่วย 12 ราย อายุเฉลี่ย 53.1±14.5 ปี และมีค่า body mass index (BMI) 23.23 ± 5.5 กก/ม2 ระดับกิจกรรมทางกายโดยการวัดจำนวณก้าวจากเครื่องเพิ่มขึ้นเมื่อเทียบกับกลุ่มควบคุม (+1956.2 ± 2164.18 Vs -1302.92 +/- 1956.03 ก้าว, P = 0.021) ระดับฮีโมโกลบินและอัลบูมินเพิ่มขึ้นในกลุ่มที่ออกกำลังกายขณะฟอกเลือดเมื่อเปรียบเทียบกับกลุ่มควบคุม [+1.0 (-0.1,2.7) vs. -1.4 (-3.4,0.2) กรัม/ดล., P= 0.025 และ +0.30(0.08, 0.50) vs. -0.15(-0.28, 0.05) กรัม/ดล., P=0.012] และยังมีการลดลงของระดับฟอสเฟตในเลือด -1.95 (-2.8, -0.63) vs. …
