Dysregulated ribosome biogenesis and p53 mutations are known to play oncogenic roles in various cancers, including pancreatic cancer. In this study, we demonstrated the therapeutic potential of BMH-21, a pharmacologic inhibitor of RNA polymerase I, against pancreatic cancer by uncovering a novel molecular mechanism involving RPA194-mediated ubiquitination of mutant p53 without affecting the ubiquitination of wild-type p53. Our key findings are that (i) BMH-21 selectively induces apoptosis and cell growth inhibition of pancreatic cancer cells with no effect on normal human pancreatic ductal epithelial cells; (ii) BMH-21 degrades RPA194; (iii) BMH-21 inhibits recruitment of both RPA194 and RPA135 on rDNA …

Foundation models have emerged as powerful feature extractors in computational pathology. However, they typically omit mechanisms for leveraging the global spatial structure of tissues and the local contextual relationships among diagnostically relevant regions-key elements for understanding the tumor microenvironment. Multiple instance learning (MIL) remains an essential next step following the foundation model, designing a framework to aggregate patch-level features into slide-level predictions. We present EAGLE-Net, a structure-preserving, attention-guided MIL architecture designed to augment prediction and interpretability. EAGLE-Net integrates multi-scale absolute spatial encoding to capture global tissue architecture, a top-K neighborhood-aware loss to focus attention on local microenvironments, and background suppression …

Radiation therapy is integral to the treatment regimens of over 50% of cancer patients. However, it is not biologically targeted to just the tumor, leading to adverse effects in peritumoral normal tissue. Selective uptake of gold nanoparticles (AuNPs) by tumors realizes tumor-specific radiosensitization via increased secondary electron release from high atomic number gold atoms. Here, we review AuNP-mediated radiosensitization and outline strategies to optimize tumor-targeted AuNP delivery. Modifying the physicochemical characteristics of AuNPs, including size, shape, charge, and surface chemistry, can increase their ability to evade the reticuloendothelial system (RES) and penetrate the dense tumor stromal architecture, thereby promoting tumor …

Drug repurposing is the process of reusing existing pharmaceuticals for novel clinical purposes, which offers advantages such as streamlined clinical trial access and reduced drug development costs. Clarithromycin (CAM), a member of the macrolide antibiotics family, is a promising candidate for repurposing in cancer therapy due to its known preclinical and clinical immunomodulatory and anticancer properties. In the current study, we investigated whether CAM could be repurposed as a preventive treatment for KRAS-mutant lung cancer, a subtype of lung adenocarcinoma that is strongly associated with heavy smoking. In this article, I critique the dominance of internalism in sport philosophy and outline an alternative theoretical framework that integrates internal and external elements while striving to transcend the dichotomous language used to conceptualize sport. The analysis begins by claiming that internalism conflates three fundamental teleological aspects of sport: goals, point, and purposes. I argue that this conflation limits internalism’s ability to explain the complexity of sporting practices. By carefully distinguishing these elements, I illustrate their distinct roles in shaping sport and explore how they interact. Grass-legume cover crop mixtures are promoted to balance ecosystem services, but predicting their decomposition and nutrient release remains a challenge. A two-year litterbag study investigated the decomposition of cereal rye, crimson clover, their field mixture, and a gradient of biomass proportions in a no-till corn system. Monocultures exhibited a strong functional dichotomy where rye decomposed slowly with a large recalcitrant fraction and clover decomposed rapidly. The field mixture yielded high biomass similar to rye and exhibited intermediate decomposition kinetics. Crucially, analysis of non-additive effects revealed a fundamental decoupling as the residue mass loss showed context-dependent synergistic acceleration but nitrogen release …

The recent spillover of highly pathogenic influenza A/H5N1 (HPAI-H5N1) viruses to cattle, other mammals, and humans poses a major risk to animal and human health. Virus adaptation to new species highlights the need for effective vaccines for animals and humans. We recently developed a rabies virus-based H5 vaccine encoding the HPAI-H5 antigen and presenting it on the surface of the rabies virus particle. To test the immunogenicity and efficacy of the vaccine in eliciting systemic and mucosal immune response, we vaccinated mice intramuscularly or intranasally with either live or inactivated and adjuvanted vaccine. Aortic thrombosis is a rare but life-threatening condition. It typically presents with abdominal or lower-limb ischemia. Although acute kidney injury (AKI) can accompany aortic thrombosis, AKI as the sole clinical manifestation has not been previously reported. A 69-year-old man with extensive atherosclerotic disease and chronic kidney disease presented with severe oliguria. Initial evaluation, including non-contrast computed tomography (CT), did not reveal any aortic abnormalities. Renal Doppler ultrasonography findings and markedly elevated D-dimer levels prompted contrast-enhanced CT, which demonstrated extensive thrombus formation extending from the descending thoracic aorta to the bilateral iliac arteries, with complete occlusion of both renal arteries and …

Ischemic limb disease continues to pose major clinical challenges. The effect of this disease causes restricted blood flow, which is followed by a limited ability to regenerate in the vascular tissues. The purpose of this study is to address the current limitations in producing large-scale generation of Smooth Muscle Cells (SMCs) derived from human induced pluripotent stem cells (hiPSCs) and then assessing their potential therapeutic efficiency individually and in combination with endothelial cells (ECs) for vascular repair. Purpose: Sexual and gender minority people (SGM) experience multiple cancer-related disparities, including higher rates of cancer risk factors, lower rates of cancer screening, higher lifetime risk of cancer, and unmet needs throughout cancer survivorship. Although many national organizations, including the National Cancer Institute (NCI) and the American Cancer Society (ACS), have emphasized the need for more cancer research among SGM communities, currently no consensus exists about the highest research priorities, promising research models, or mechanisms to support collaboration between geographically dispersed scientific teams.

Transect walks were used to collect data to determine species richness, indices of abundance, species diversity, and flight periods of butterfly species in Big Spring Hollow in the South Fork of Provo Canyon, Utah Co., Utah. Advantages and cautions are discussed for the use of this sampling method. A total of 26,493 butterflies were recorded from 232 transect walks during the 10 years of this study (2001–2005 and 2019–2023), with a mean annual total number of 2649.3 butterflies. A total of 86 butterfly species were identified along the Big Spring Hollow transect belonging to 5 families: 18 Hesperiidae, 19 Lycaenidae, …

Background: Alcohol, a known carcinogen since 1988, increases risk of cancers of the mouth, pharynx, larynx, esophagus, liver, breast, and colorectum. More than one half of US adults consume alcohol whereas about two thirds are unsure whether or unaware that alcohol intake is linked to cancer risk.
Methods: Our objective was to survey awareness, beliefs, intentions, policies, practices, and goals/perceived responsibilities related to establishing/promoting actions to reduce alcohol-related cancer risks from leaders of 72 NCI-Designated Cancer Centers.
Results: Sixty-one percent responded. Forty-three percent either believed in or were uncertain about alcohol's purported health benefits. Nevertheless, 86% considered it part of …

Background: Alcohol use disorder (AUD) represents a highly prevalent, costly, and often untreated condition in the United States. Post-traumatic stress disorder (PTSD) represents a common comorbidity with AUD which worsens outcomes and decreases functional outcomes. Suvorexant (SUV) shows clear promise as a novel therapeutic candidate to treat AUD and PTSD.
Methods: This study features a promising compound (i.e., suvorexant), the application of a well-established human laboratory paradigm (i.e., alcohol cue reactivity), and a novel early efficacy laboratory model (i.e., practice quit attempt) to provide a cost/time-efficient evaluation of safety and initial efficacy of suvorexant for AUD with comorbid PTSD. Background: Stroke is a sexually dimorphic disease, with different risk factors, incidence, outcomes, and treatment responses in men and women. While sex differences have been documented in preclinical studies, these findings often come from single-site studies with small sample sizes and require validation across diverse research settings.
Methods: We used data from the SPAN (Stroke Preclinical Assessment Network), a randomized, placebo-controlled, blinded, multilaboratory trial, to determine if sex differences in neurological outcomes are present in preclinical stroke models. Natural and working lands are often touted as natural climate solutions due to their ability to take up and store carbon, and land managers are increasingly tasked with considering how management actions impact carbon outcomes alongside other management goals. As a result, there is a need to assess how management actions may drive trade-offs and co-benefits between multiple management goals. However, quantitative analysis of trade-offs is not feasible for many local-scale management projects. Conjugated thiophene-based diboronate esters, 1–4, were synthesized to investigate how substituents on catechol alter the Lewis acidity and accessibility to boron, and influence binding interactions with low molecular weight Lewis basic amines. Crystallographic analysis indicated that both boron centres bind analyte and that Lewis base coordination induces deplanarization of the conjugated system. Spectroscopic studies demonstrate that amine binding disrupts conjugation, producing distinct and reproducible optical responses that depend on analyte basicity and structure, with association constants in organic solvent ranging from 103 to 106 M−1. The emergence of effective immunotherapies has drastically revolutionized clinical management of many cancer types. Among them, chimeric antigen receptor (CAR)-T cell therapy (CTT), as a groundbreaking approach, has been considered as a “living drug,” displaying unprecedented clinical outcomes with hematological malignancies, including B cell leukemia and lymphomas, and multiple myeloma. Despite the high remission rates and improved survival achieved with hematological cancers, the effectiveness of CTT in solid tumors remains largely unsatisfactory. The role of the tumor immune microenvironment (TIME) in modulating responses to antiestrogen therapy in hormone receptor-positive (HR+) breast cancers remains unclear. We analyzed pre- and on-treatment biopsies from patients with HR+ breast cancer treated with letrozole to induce estrogen deprivation (ED). Stromal tumor-infiltrating lymphocytes, assessed by H&E staining, and immune-related gene sets, including IFN-γ signaling genes, measured by RNA-Seq, were increased in ED-resistant tumors. Cyclic immunofluorescence and spatial transcriptomics revealed an abundance of CD8+ T cells and enhanced antigen processing and immune gene signatures in ED-resistant tumors. Importance: While home-based self-sampling for cervical cancer screening is an evidence-based strategy proven to increase screening access and uptake, it is not currently recommended in the US despite recent Food and Drug Administration approval of the first at-home self-sampling device. Little nationally representative research has examined preference for and drivers of home-based self-sampling over clinic-based testing (the standard of care).

EGFR-mutant lung adenocarcinomas (LUADs) that are vulnerable to the EGFR antagonist osimertinib (Osi) eventually relapse, owing in part to the emergence of drug-tolerant persister (DTP) cells that arise through epigenetic mechanisms. Intratumoral DTP cells can herald a worse clinical outcome, but the way in which DTP cells influence LUAD progression remains unclear. Osi-resistant (OR) cells exhibit typical DTP cell features, including a propensity to undergo senescence and epithelial-mesenchymal transition (EMT), which can activate heightened secretory states. The development of structurally distinct and modular photocatalysts for direct hydrogen atom transfer (HAT) has emerged as a key frontier in radical-mediated C-H functionalization of strong carbon-hydrogen bonds. Despite the largely untapped potential across the fields of organic synthesis, medicinal chemistry, and materials science, few classes of direct HAT photocatalysts are currently known, and the approaches to enhancing their photocatalytic activity remain underdeveloped. Background: Retroperitoneal leiomyosarcomas are aggressive malignancies. Complete surgical resection with negative margins is crucial to decrease the risk of recurrence but can be risky due to vascular involvement. The aim of our study was to evaluate the different approaches to IVC and renal vein management and their impact on postoperative complications.
Methods: We performed a retrospective review of patients who underwent surgery for retroperitoneal leiomyosarcoma with IVC and/or renal vein involvement at our institution from 2016 to 2024. This paper presents a novel numerical approach for solving the partial differential equations (PDEs), focusing on the Diffusion equation. The method combines a collocation approach with wavelet techniques to achieve high accuracy in approximating solutions. A detailed framework for the proposed method, explaining the discretization process at multiple collocation points and the formulation of the resulting system of linear equations is provided. An implementation is conducted to demonstrate the method's effectiveness in capturing the complex behaviors typical of the model studied. Omental tuberculosis (TB) is a rare form of abdominal TB, often presenting with nonspecific symptoms that mimic malignancies. We report a case of a 16-year-old female from India who presented with abdominal distension, vomiting, weight loss, and amenorrhea. Imaging revealed an abdominal mass, and a diagnosis of omental TB was confirmed via histopathology. This case highlights the diagnostic challenge posed by such atypical presentations, especially in TB-endemic regions. The patient also developed rifampicin-induced jaundice, which resolved upon drug cessation and supportive care. Anti-TB therapy was cautiously reintroduced at a modified dose. Latine older adults face disproportionate risk of chronic nutrition-related conditions and food insecurity, which can exacerbate or contribute to mental health-related concerns. Cultural stigma toward mental illness can result in Latine clients, especially older adults, somatizing symptoms and focusing on physical health goals in counseling. Our understanding of the impacts of climate change, sea-level rise (SLR), and freshwater management on the magnitude and variability of carbon fluxes in tidal forested wetlands remains limited. In this study, we applied a process-driven wetland biogeochemistry model, Wetland Carbon Assessment Tool—DeNitrification-DeComposition (WCAT-DNDC) model to explore responses of carbon fluxes in tidal swamp forests to climate change-induced alterations in hydrologic conditions and to predict impacts of planned reintroduction of river flows. We selected twelve sites in three habitats (throughput, relict, degraded) inside the Lake Maurepas swamp forests (Louisiana, USA) to represent various hydrological and salinity regimes.
