Allergens are non-infectious environmental proteins that can trigger T helper 2 (Th2) cell-mediated inflammation in atopic individuals. Allergens are structurally diverse and typically lack pathogen-associated molecules patterns (PAMPs), which are recognized by pathogen recognition receptors (PRRs). PRRs are the canonical sensors of the innate immune system that typically detect PAMPs in microbes, initiating signaling pathways that drive pathogen clearance. Because allergens generally do not activate PRRs in the same way as infectious agents, alternative detection mechanisms are likely responsible for their recognition and the subsequent initiation of Th2 cell-mediated inflammation. Distinct epithelial cell states arise during differentiation, but mechanisms generating transcriptomic diversity among them remain poorly defined. The human ureter urothelium contains basal progenitor, intermediate cells, and terminally differentiated umbrella cells. Prior single-cell RNA sequencing revealed similar global gene expression profiles across these states, raising the question of how distinct identities emerge. Here, we show that alternative cleavage and polyadenylation (APA) introduces a major layer of transcriptomic diversity during urothelial differentiation, largely independent of changes in mRNA levels. Analysis of 13,544 urothelial cells identified hundreds of differentiation-associated APA events. Single-cell imaging revealed spatially specific APA patterns, and reporter assays demonstrated …

Increasing plastic pollution has become a critical environmental issue, leading to  challenges in management. Specifically, microplastics (MPs) are of particular concern  because they can persist in aquatic ecosystems and adversely affect marine  organisms. The objective of this study was to assess the occurrence of MPs  in the soft tissues of Patella caerulea (Linnaeus, 1758) collected from the Mersin  Viranşehir coastal area. Eyelids play a crucial role in protecting the eyes of vertebrates; however, certain  reptile species, such as Ophisops elegans, completely lack movable eyelids. In this  study, we present the first video-documented evidence of a blink-like reflex in O.  elegans, captured through endoscopic field observations and corroborated by laboratory video analysis. The observed movement consisted of a brief convex  deformation of the ocular surface, lasting approximately 0.3 s and resembling a blink  despite the presence of a fixed transparent brille. Hydroxyapatite (HAp) is an effective inorganic gene delivery carrier due to its ability to transport genetic cargo across cell membranes, protect it from proteolysis, and enable escape from late endosomes via pH-controlled dissolution. However, its transfection efficiency remains lower than that of viral agents, prompting studies of hybrids with cationic molecules or phases to enhance the gene delivery performance. This study reports on the synthesis of HAp in regular and reverse micellar regions of a ternary microemulsion system composed of cetyltrimethylammonium bromide (CTAB), 1-hexanol and water. Fusobacterium nucleatum is a Gram-negative anaerobe associated with periodontitis and colorectal cancer. It secretes putrescine, a polyamine that promotes biofilm formation by oral co-colonizers and enhances the proliferation of cancer cells. However, the physiological importance of putrescine for F. nucleatum itself remains unexplored. Here, we show that putrescine biosynthesis, mediated by the ornithine decarboxylase gene oda, is essential for F. nucleatum viability. Deletion of oda was only possible when a functional copy was provided in trans, and CRISPR interference of oda expression resulted in complete growth defects. The family Arenaviridae encompasses zoonotic, rodent-borne pathogens (e.g., Lassa, Machupo, and Junín viruses) that cause severe viral hemorrhagic fevers with high case fatality rates. The current therapeutic landscape is severely limited, underscoring the urgent need for novel antiviral strategies. A promising approach involves combining directly acting antivirals with host-targeted antivirals. A compelling host-targeted antiviral target is the aryl hydrocarbon receptor (AHR). This ubiquitous ligand-activated transcription factor is a recognized pro-viral host factor across multiple viral families. Safety is crucial in the oil and gas refinery industry for production and distribution. Companies must ensure a workplace free from common injuries and protected against serious incidents. This paper evaluates employees’ knowledge, attitudes, practices, and workplace environment at North Refineries Company (Baiji). A descriptive cross-sectional study was conducted from September 10, 2024, to September 20, 2025, involving 480 employees (378 workers and 102 managers). Data were collected through face-to-face interviews and analysed using SPSS version 24. North Refineries Company (Baiji). workers scored 72.49 % on general knowledge and 96.83% on attitudes, but only 53.97% on safe behaviour. Division managers …

Shallow bathymetry data constitute the core foundational information underpinning the ecological, resource, and strategic values of island and coastal zones. Satellite-derived bathymetry has evolved into a key technical means for large-scale acquisition of shallow bathymetry information. Focusing on satellite-based lidar bathymetry, optical imaging bathymetry, and synthetic aperture radar (SAR) bathymetry, this study systematically summarizes the core principles, applicable scenarios, technical strengths, and inherent limitations of each technology. It is shown that satellite-derived bathymetry has undergone an evolutionary shift from theoretical construction to practical application, and from single-method, single-data-source approaches to multi-technology, multi-source data fusion frameworks. Credit Card Fraud Detection (CCFD) has become a critical challenge to financial security due to increasingly sophisticated fraudulent activities. This study investigates the effectiveness of Meta-Heuristic (MHT) optimization techniques in improving fraud detection (FD) through feature selection (FS) and model optimization. To address class imbalance, Random Under-Sampling (RUS) was applied. Patients with poorly differentiated thyroid cancer (PDTC) and anaplastic thyroid cancer (ATC) face a much poorer prognosis than those with differentiated thyroid cancers. Around 25% of PDTCs and 35% of ATCs carry the BRAFV600E mutation, which constitutively activates the MAPK pathway, a key driver of cell growth. Although combining BRAF and MEK inhibitors can shrink tumors, resistance often develops. The exact cause of this resistance remains unclear. [Objective ] This study aims to screen the strains of lactic acid bacteria (LAB ) suitable for Suancai fermentation.[Method ] Traditional fermented Suancai from the Dai ethnic region of Yunnan was used as the research samples for the isolation and purification of LAB.The purified isolates were evaluated for fermentation characteristics,including nitrite degradation,acid production,acid resistance,and salt tolerance.Additionally,the antimicrobial activity,antibiotic susceptibility,hemolytic activity,and amino acid decarboxylase activity were assessed to identify dominant strains.[Result ] A total of 208 LAB strains were isolated from 40 Suancai samples.Preliminary screening based on nitrite degradation identified 20 strains with high efficiency,and 16S rDNA sequencing identified 15 strains …

Hypoxic eye diseases represent a pivotal yet often underappreciated contributor to the onset and progression of many retinal disorders. When hypoxia persists or exceeds the tissue's compensatory capacity, it triggers pathological retinal neovascularization, blood-retinal barrier disruption, and neuronal apoptosis, ultimately resulting in irreversible visual impairment. Connexins (Cxs) form gap junction channels and hemichannels and regulate retinal cell proliferation, differentiation, and survival, thereby playing a central regulatory role in the pathogenesis of hypoxic ocular diseases. In addition to gap junctions, Cx hemichannels promote transmission of molecules between intra- and extracellular environments, further influencing retinal homeostasis under hypoxic stress. Epigenetic modifications such as DNA methylation play a fundamental role in oncogenesis and the progression of neoplasms neoplasias. DNA methyltransferase inhibitors (DNMTi) constitute a family of therapeutic agents that impede the methylation at the 5-position on cytosine nucleotides, thereby modulating the epigenetic regulation of tumor suppressor genes, oncogenes, and other key regulatory genes. The first-generation DNMTi azacitidine and decitabine have demonstrated substantial efficacy in the treatment of medically non-fit, older patients with acute myeloid leukemia (AML) or myelodysplastic syndrome (MDS) ineligible for intensive chemotherapy (IC), by virtue of their favorable safety profile. However, the specific associations between various online screen sedentary behaviors and depressive symptoms remain unclear. Therefore, this study conducted a two-year nationwide cross-sectional survey to examine how different types of online screen behaviors affect depressive symptoms in children and adolescents. Here, we report that a dietary long-chain fatty acid enhances the yeast Candida albicans colonization of the murine gut partly by eliciting modifications to the fungal cell surface. In recent years, hybrid organic–inorganic metal halides have been at the forefront of materials research. Typically, the functional (e.g., optoelectronic) properties of hybrid halides are derived from the inorganic structural part, whereas the organic structural units can add extra advantages in terms of stability, rigidity, and processability. Here, we report the design, synthesis, and characterization of two new hybrid materials in which the outstanding photophysical properties originate from the organic structural part. The new compounds, (C15H16N)2CdCl4 and((Br)C15H15N)2CdCl4, have 2D layered Ruddlesden–Popper-type perovskite structures. These hybrids are blue-white …

Capsaicin, a naturally occurring polyphenol, is known to affect energy expenditure and muscle fatigue and modulate contractions in skeletal muscle. The L-type Ca2+ channels are known to be an important ion channel involved in the various muscle functions and the effect of capsaicin on the skeletal L-type Ca2+ channels is currently unknown. Effective and safe treatments for neglected tropical diseases caused by parasites, such as Chagas disease and sleeping sickness, remain lacking, posing a significant challenge for researchers worldwide. The rational design of dimeric compounds inspired solely by the pharmacophoric core of benznidazole (2-nitroimidazole) has proven to be a promising strategy for antiparasitic development. Thus, in the present work, it was increased the linker between the active units (2-nitroimidazole) to improve the interaction with TcNTR, facilitating the bioactivation of the longest dimers. Post-translational modifications are a critical tool bacteria employ to survive the challenges of microbial life. Acetylation continues to gain more recognition as a modification of unique importance because the sources of acetylation – acetyl-phosphate and acetyl-coenzyme-A – are critical metabolites associated with central carbon metabolism, acetate overflow metabolism and acetate assimilation. This suggests that acetylation, indelibly coupled to metabolic flux, integrates metabolic sensing with any cellular process or pathway where an acetylation alters the function of a protein. Adverse fire regimes threaten biodiversity, potentially leading to population declines and increased extinction risk. Understanding how varying fire regimes affect threatened species is essential for effective ecosystem management, including in Western Australia where diverse ecosystems are exposed to wildfires, Indigenous burning, and prescribed fire. We compiled and synthesised data on the threat of adverse fire regimes to threatened animal taxa in Western Australia. Using a threat classification scheme, we ranked adverse fire regimes as having either no, low, medium, high, or unknown impact on each taxon. ESR1 mutations are the leading cause of endocrine therapy resistance and progression in ER-positive metastatic breast cancer. ESR1 mutations are detected in ~50% of metastatic breast cancer patients, and identification of effective targeted therapeutics are critically needed. Here, we identified enrichment of dysregulated replication stress and DNA damage responses in multiple ESR1 mutant models. Targeting the replication stress response utilizing checkpoint inhibition in combination with PARP inhibition synergistically suppressed growth, induced cell cycle arrest, and attenuated DNA replication. Background: Cardiorespiratory fitness is an integrative measure of cardiometabolic health and predictor of survival, yet little is known about its molecular underpinnings. Small molecule metabolites and lipids are increasingly recognized as exercise-stimulated signaling molecules and candidate molecular transducers of cardiorespiratory fitness.

Methods: We performed nontargeted liquid chromatography mass spectrometry-based plasma metabolomics in 654 participants (mean age, 35 years; 55% women) from the HERITAGE Family Study (Health, Risk Factors, Exercise Training, and Genetics) who had cardiorespiratory fitness (maximal oxygen uptake [VO2max]) measured by cardiopulmonary exercise testing and underwent 20 weeks of supervised endurance training. Mesenchymal stem cells (MSCs) are widely used in regenerative medicine due to their self-renewal capacity, multilineage differentiation potential, and paracrine activity. Limitations associated with the sourcing and scalability of human MSCs have increased interest in xenogeneic alternatives, particularly sheep umbilical cord–derived MSCs (sUC-MSCs). These cells display high proliferative capacity, stable karyotypes, and a typical MSC immunophenotype (CD44, CD73, CD90, CD105), with low basal immunogenicity. sUC-MSCs exhibit osteogenic, chondrogenic, and adipogenic differentiation potential and secrete angiogenic and immunomodulatory factors, including VEGF, HGF, IGF-1, TGF-β, and IL-10.
