The best-supported hypothesis for why zebras have stripes is that stripes repel biting flies. While this effect is well-established, the mechanism behind it remains elusive. Myriad hypotheses have been suggested, but few experiments have helped narrow the field of possible explanations. In addition, the complex visual features of real zebra pelage and the natural range of stripe widths have been largely left out of experimental designs. In paired-choice field experiments in a Kenyan savannah, we found that hungry Stomoxys flies released in an enclosure strongly preferred to land on uniform tan impala pelts over striped zebra pelts but exhibited no …

LGD-4033, a selective androgen receptor modulator, and MK-677, a growth hormone secretagogue, are becoming increasingly used amongst recreationally-active demographics. However, limited data exist describing their effects on health- and androgen-related biomarkers. The purpose of this case study is to report changes in body composition and biomarkers during and after continued co-administration of LGD-4033 and MK-677. We additionally aimed to examine muscular strength and intramuscular androgen-associated biomarkers relative to non-users. A 25-year-old male ingested LGD-4033 [10 mg] and MK-677 [15 mg] daily for 5 weeks. Blood and body composition metrics were obtained pre-, on-, and post-cycle. One repetition maximum (1RM) leg …

Cancer cachexia is a lethal metabolic syndrome featuring muscle wasting with preferential loss of fast-twitching muscle mass through an undefined mechanism. Here, we show that cancer induces muscle wasting by selectively degrading myosin heavy chain (MHC) subtypes IIb and IIx through E3 ligase UBR2-mediated ubiquitylation. Induction of MHC loss and atrophy in C2C12 myotubes and mouse tibialis anterior (TA) by murine cancer cells required UBR2 up-regulation by cancer. Genetic gain or loss of UBR2 function inversely altered MHC level and muscle mass in TA of tumor-free mice. BACKGROUND: Sarcopenia, generally described as "aging-related loss of skeletal muscle mass and function", can occur secondary to a systemic disease.
AIM: This project aimed to study the prevalence of sarcopenia in chronic ambulatory stroke survivors and its associated risk factors using the two most recent diagnostic criteria.
DESIGN: A cross-sectional observational study.
SETTING: A scientific laboratory.
POPULATION: Chronic stroke.
METHODS: Twenty-eight ambulatory chronic stroke survivors (12 females; mean age=57.8±11.8 years; time after stroke=76±45 months), hand-grip strength, gait speed, and appendicular skeletal muscle mass (ASM) were measured to define sarcopenia. BACKGROUND: Altered peripheral immune/inflammatory system and brain volumetric changes have been implicated in the pathophysiology of bipolar disorder (BD). This study aimed to evaluate how peripheral levels of cytokines are related to volumetric brain changes in euthymic patients with BD.
METHODS: Euthymic patients with BD (n = 21) and healthy controls (n = 22) were enrolled in this exploratory study. Multimorbidity in the elderly is a major public health issue with serious consequences. This study investigated the effects of health promotion programs on perceived self-efficacy and self-care practices among the elderly with multimorbidity using a quasi-experimental study design from February to July 2021. The study samples were the elderly from Doi Saket District, Chiang Mai Province, Thailand, selected by a multistage random sampling technique. The purpose of this study was to determine the efficacy of a 6-week suspension training exercise program on fitness components in older adults. Eleven participants (age = 80 ± 5 yrs) completed a 6-week suspension training exercise program. Pre- and post-fitness assessments comprised of body composition, handgrip dynamometer, functional reach, and balance. The 6-week suspension training intervention required individuals to perform suspension training exercises for fifty minutes, twice per week. A major challenge of genome-wide association studies (GWASs) is to translate phenotypic associations into biological insights. Here, we integrate a large GWAS on blood lipids involving 1.6 million individuals from five ancestries with a wide array of functional genomic datasets to discover regulatory mechanisms underlying lipid associations. We first prioritize lipid-associated genes with expression quantitative trait locus (eQTL) colocalizations and then add chromatin interaction data to narrow the search for functional genes. Polygenic enrichment analysis across 697 annotations from a host of tissues and cell types confirms the central role of the liver in lipid levels and highlights the selective …

Avian dinosaurs evolved skeletal adaptations resulting in numerous fused spinal structures, like the synsacrum, present in extant birds. The synsacrum is a highly fused region necessary for stiffening the spine, and it fuses the spine to the pelvic ilium. Conventional wisdom suggests that fusion occurred in ovo during embryonic development via the cartilage matrix that patterns the future vertebrae. We show that post hatch birds have intervertebral discs separating all vertebrae of the spine, leading us to re-examine this notion in chickens up to 10 weeks of age. AHCC is a beta-glucan derived from the shiitake mushroom. It is used as an immune supplement and is known to boost the immune system. The mechanisms involved are increased lymphocyte proliferation and reduction of inflammation. The effects have been well studied in T cells. However, factors produced by T cells that affect B cells have not been studied. In this study, we explored the gene products stimulated by the AHCC exposure in T lymphocytes which play a role in B cell function. Mitochondrial glutamate-oxaloacetate transaminase 2 (GOT2) is part of the malate-aspartate shuttle, a mechanism by which cells transfer reducing equivalents from the cytosol to the mitochondria. GOT2 is a key component of mutant KRAS (KRAS*)-mediated rewiring of glutamine metabolism in pancreatic ductal adenocarcinoma (PDA). Here, we demonstrate that the loss of GOT2 disturbs redox homeostasis and halts proliferation of PDA cells in vitro. GOT2 knockdown (KD) in PDA cell lines in vitro induced NADH accumulation, decreased Asp and α-ketoglutarate (αKG) production, stalled glycolysis, disrupted the TCA cycle, and impaired proliferation. Previous research has provided evidence that aerobic HIIT exercise can induce important physiological adaptations and elicit improvements in health and fitness parameters. However, most of the research has taken place in a laboratory setting with specialized equipment and monitoring devices. It begs the question, is HIIT accessible to the general aging population? The objective of the current research was to employ an age-predicted HRmax to prescribe a minimally invasive 4x4 cycle ergometer HIIT protocol. Objective:optimizing the extraction process of methylated catechin from Zijuan tea.Methods:Using acid ethanol as extraction solvent, based on three single factor experiments, the extraction time, extraction temperature and liquid-solid ratio were used to investigate the interaction of the above three factors on the extraction rate of methylated catechin from Zijuan tea.Results:The optimum extraction conditions were as follows: extraction time 100 min, extraction temperature 75 ℃, solid-liquid ratio(m Zijuan tea∶VSolvent)1∶74(g/mL); under these conditions, the mass concentration of methylated catechin in the extract could reach 26.7 mg/g, and the relative error with the verification value was 1.48%.Conclusion:HPLC and …

Diseases linked to high cholesterol have been well studied and several diseases-associated candidate genes have been suggested. Among these, the apolipoprotein E gene (APOE) gene associated with lipid metabolism and lipid-related traits was reported in the non-Hispanic population. There are three types of alleles APOE ε2, ε3, and ε4. The APOE ε4 allele has been identified as a biomarker for Alzheimer’s disease (AD) and cardiovascular disease (CVD). In recent years, there have been limited studies regarding the association between the APOE gene and hypercholesterolemia in the Mexican American population. Cooperative Extension’ National Framework for Health Equity and Well-Being (Framework) provides a roadmap by which Cooperative Extension can help ensure that all people have that opportunity. Mild Traumatic brain injury (mTBI) is a signature wound in military personnel, and repetitive mTBI has been linked to age-related neurogenerative disorders that affect white matter (WM) in the brain. However, findings of injury to specific WM tracts have been variable and inconsistent. This may be due to the heterogeneity of mechanisms, etiology, and comorbid disorders related to mTBI. Non-negative matrix factorization (NMF) is a data-driven approach that detects covarying patterns (components) within high-dimensional data. Listeriosis is a bacterial infection caused by the gram-positive pathogen Listeria monocytogenes. When compared to other foodborne illnesses, listeriosis has a higher death rate due to an increased incidence of complications such as meningitis, hydrocephalus, and sepsis in immunocompromised populations. Elderly individuals experience a condition known as immunosenescence, which is a gradual compromisation of the immune system brought on by natural aging. DNA methylation (DNAm) has been reported to be associated with many diseases and with mortality. We hypothesized that the integration of DNAm with clinical risk factors would improve mortality prediction. We performed an epigenome-wide association study of whole blood DNAm in relation to mortality in 15 cohorts (n = 15,013). During a mean follow-up of 10 years, there were 4314 deaths from all causes including 1235 cardiovascular disease (CVD) deaths and 868 cancer deaths. Ancestry-stratified meta-analysis of all-cause mortality identified 163 CpGs in European ancestry (EA) and 17 in African ancestry (AA) participants at p < 1 × 10

Cognitive dysfunction is an important consequence of ischemic stroke, which can progress in the first few years and is primarily determined by clinical factors. This study aimed to investigate the clinical determinants of cognitive dysfunction in stroke survivors in West Nusa Tenggara Province, Indonesia. This cross-sectional study assessed 255 ischemic stroke survivors with a mean age of 57.1±9.3 years old and 29–79 years old, recruited consecutively in three main hospitals in West Nusa Tenggara Province between March 2019 and October 2021. Benign prostatic hyperplasia (BPH) is a severe illness affecting middle-aged and geriatric male patients. This disease normally occurs at the age of 40 or above and is also associated with sexual dysfunction. Alpha-blockers and 5α-reductase inhibitors are the preferred drugs practiced to treat BPH. However, invasive surgical therapy remains the gold standard for managing the condition in the case of refractory and intricate BPH conditions. Cardiovascular disease is the leading cause of death in the United States. Cardiac function declines with age, and promotes a number of age-related disorders, including atrial fibrillation and heart failure. Mitochondrial oxidative stress has long been suspected as a primary driver of aging pathology in multiple tissues, including the heart and brain. We used several phenotypic assays to characterize chemically-induced accelerated cardiac aging, as well as two transgenic knockout models for heart and brain specific loss of superoxide dismutase (SOD2), a mitochondrial antioxidant protein.

Background: A single high-fat meal (HFM) is known to elevate blood lipid levels above what is normal, resulting in postprandial lipemia (PPL). Chronic hyperlipemia has been associated with increased risk for cardiovascular disease (CVD) and type-2 diabetes mellitus (T2DM) development. Current physical activity (PA) guidelines recommend at least one set of 8-12 repetitions of each exercise to attenuate this risk. Adults older than 50 (OF) years (y) are at an increased risk for cardiometabolic disease compared to younger adults (YA). Aerobic exercise (AE) performed in the pre-prandial period has been observed to attenuate PPL in both OF and YA. However, …

Insulin-like growth factor 1 (IGF-1) is a neuroendocrine signaling hormone that plays a vital role in growth and development, as well as learning and memory. Inhibition of this hormone results in cognitive impairments like those seen with age-related decline. While a majority of research has focused on the role of IGF-1 on neurons, the role of astrocytes still needs to be explored. Our research investigates how astrocytes and cognition are affected as a result of direct regulation of localized IGF-1 production in early development and after puberty.
