Transition metal centers possessing an octahedral geometry are useful for understanding the natural role of metal ions in biology. Complexes based on such metals might improve upon the widely used square planar Pt(II) anticancer drug cisplatin, cis-[Pt(NH3)2Cl2]. The octahedral triaquatricarbonylrhenium(I) cation, fac-[Re(CO)3(H2O)3]+, has been shown to possess anticancer activity. Furthermore, Re(I) complexes containing unstable rhenium isotopes have found utility as radiodiagnostic and radiotherapeutic agents. In this dissertation, novel dendronized cellulose derivatives with biodegradable backbones were created by using sodium carboxymethyl cellulose (CMC) as the starting material. CMC was first transformed into its corresponding tetrabutyl ammonium salt, which reacted with 2-chloro-N-methylpyridinium iodide (CMPI) to produce an activated ester of [2-(N-methyl pyridium) carboxymethyl cellulose] iodide (MPCMCI). The reaction of MPCMCPI and di-tert-butyl 4-[2-(ter-butoxycarbonyl)ethyl]-4-aminoheptanedicarboxylate (BA) produced a new derivative with BA dendron elaborated on cellulose backbone (CMCBA). Xanthene dyes are one of the oldest synthetic dyes. Fluorescein was first synthesized by von Baeyer in 1871. Since its discovery, it has been extensively studied. Fluorescein can exist as four different ionic forms (cationic form, neutral form, monoanionic form and dianionic form). Under physiological conditions (pH 7.4), fluorescein mainly exists as the dianionic form which grants it large quantum yield and excellent solubility in water. This renders fluorescein and its derivatives useful for studies in biological media. Simple water soluble lanthanum and europium complexes are effective at detecting neutral sugars as well as glyco- and phospholipids. In solutions at physiologically relevant pH the fluorescent lanthanum complex binds neutral sugars with apparent binding constants comparable to those of arylboronic acids. Interference from commonly occurring anions is minimal. The europium complex detects sialic acid-containing gangliosides at pH 7.0 over an asialoganglioside. This selectivity is attributed, in large part, to the cooperative complexation of the oligosaccharide and sialic acid residues to the metal center, based on analogous prior studies. My graduate work has focused on the development of new fluorophore materials towards addressing several interdisciplinary research problems. Selective detection of sialic acid via a boron-amide interaction. Concentration changes of sialic acid can be monitored ratiometrically in the visible region using a boronic acid based chemosensor. The technique shows great promise for the detection of sialic acid, a critical cell surface residue involved in cancer, graft rejection and virulence in certain bacteria. Cα,α-disubstituted amino acids (ααAAs) are widely utilized to conformationally constrain peptides. Several pentapeptides containing dipropylglycine (Dpg) at alternating positions and their α-amino acid counterpart L-norvaline (Nva) analogues were synthesized to fully investigate the impact of Dpg on peptide backbone structure in aqueous solution. CD, VCD and NMR spectral analysis suggest that Dpg containing peptides adopt more ordered structures relative to their Nva containing analogues. Studies of grafting polymer on surfaces with chemical bonds started in the 1970’s, but few projects have focused on grafting polypeptide on spherical particles. In this research, polypeptides poly(å-carbobenzyloxy-L-lysine) (PCBL) or poly(ã-benzyl-L-glutamate) (PBLG) were grafted on silica sphere surfaces by using the “grafting from” method. The polypeptide-grafted silica spheres are called SiPCPs. The growth of polypeptide chains as a function of monomer amount and reaction time was explored. The properties of a mixture of PBLG rod-like polymer and PBLG SiPCPs were investigated. At high concentrations of these two components, the phase behavior was studied. Chapter 1 presents a concise introduction to porphyrins, especially on the synthesis of meso-tetraphenylporphyrins, which were synthesized during the course of my research and were used for further purposes in the following chapters. Chapter 2 describes the synthesis of a series of cobaltacarborane-porphyrin conjugates for the boron neutron capture therapy (BNCT) of tumors. Using an efficient and high yield ring-opening reaction, N-substituted porphyrins were also synthesized. The spectroscopic and structural properties of these compounds are presented. Membrane-bound factor Xa alone catalyzes prothrombin activation following initial cleavage at Arg(271) and prethrombin 2 formation (pre2 pathway). Factor Va directs prothrombin activation by factor Xa through the meizothrombin pathway, characterized by initial cleavage at Arg(320) (meizo pathway). We have shown previously that a pentapeptide encompassing amino acid sequence 695-699 from the COOH terminus of the heavy chain of factor Va (Asp-Tyr-Asp-Tyr-Gln, DYDYQ) inhibits prothrombin activation by prothrombinase in a competitive manner with respect to substrate. Construction of biomotors is an exciting area of scientific research that holds great promise for the development of new technologies with broad potential applications in areas such as the energy industry and medicine. Herein, we demonstrate the fabrication of prealigned microarrays of motile Escherichia coli bacterial cells on SiOx substrates. To prepare these arrays, holed surfaces with a gold layer on the bottom of the holes were utilized. The attachment of bacteria to the holes was achieved via nonspecific interactions using poly-l-lysine hydrobromide (PLL). This research focuses on superacid perfluoroalkyl(aryl) sulfonimides and their derivatives as a source of stable zwitterionic salts. The strong resonance stabilization of sulfonimides imparts unexpected stability to the zwitterions.
Two types of zwitterions were investigated. The first are symmetrical diaryliodonium zwitterions (DZs) as potential photo acid generators (PAGs) for microlithography. Driven by the demand for smaller feature sizes in the microelectronics industry, microlithography using shorter wavelength light has evolved and new PAGs are of interest. Photovoltaic cells and fluorescence sensing are two important areas of research in chemistry. The combination of photon-activated electron donors with electron acceptors provides a strong platform for the study of optical devices. A series of four oligothiophene-ruthenium complexes has been synthesized. Variation in oligothiophene length and bipyridine substitution allowed comparison of these variables on electronic properties. The longer oligothiophenes display lower energy absorption and emission compared to the shorter ones. Aromatic conjugation appears more complete with para-, rather than meta-, substitution. Oligothiophenes and Ru(bpy)32+ are highly fluorescent individually, but fluorescence is quenched when connected. Bonds of carbon to fluorine are …

Several novel polymer monoliths for the analysis of peptides and proteins were synthesized using polyethylene glycol diacrylate (PEGDA) as crosslinker. Photo-initiated copolymerization of polyethylene glycol methyl ether acrylate and PEGDA yielded an inert monolith that could be used for size exclusion liquid chromatography of peptides and proteins. This macroscopically uniform monolith did not shrink or swell in either water or tetrahydrofuran. More importantly, it was found to resist adsorption of both acidic and basic proteins in aqueous buffer without any organic solvent additives. New RNA interaction interfaces are reported for designing RNA modules for directional supramolecular self-assembly. The new interfaces are generated from existing ones by inserting C-loops between the interaction motifs that mediate supramolecular assembly. C-Loops are new modular motifs recently identified in crystal structures that increase the helical twist of RNA helices in which they are inserted and thus reduce the distance between pairs of loop or loop-receptor motifs from 11 to 9 base-stacking layers while maintaining correct orientation for binding to cognate interaction interfaces. The structural protein flagellin, FlaA derived from hyperthermophilic Aquifex pyrophilus, a rod shaped eubacterial species that grows near hydrothermal vents at an optimum growth temperature of 85 °C forms elongated thermostable flagella nanotubes. FlaA was successfully over-expressed for the first time using pET28c plasmid in E. coli BL21 cells at 37 °C. The protein was isolated from inclusion bodies that were solubilized by alkaline pH, refolded by dialysis and purified by chromatographic techniques. The alpha-helical coiled-coil motif serves as a robust scaffold for incorporating electron-transfer (ET) functionality into synthetic metalloproteins. These structures consist of a supercoiling of two or more R helices that are formed by the self-assembly of individual polypeptide chains whose sequences contain a repeating pattern of hydrophobic and hydrophilic residues. Early work from our group attached abiotic Ru-based redox sites to the most surface-exposed positions of two stranded coiled-coils and used electron-pulse radiolysis to study both intra- and intermolecular ET reactions in these systems. Characterization of phosphoproteins-including detection, identification of phosphoproteins and identification of phosphorylation sites-is mostly done with radiolabeling and proteomic techniques. Three main topics related to phosphoprotein characterization are included in this dissertation. First, large-scale characterization of the CHO (Chinese hamster ovary) cell phosphoproteome was done using two dimensional gel electrophoresis (2DE) separation, visualization of phosphoproteins by radiolabeling or a phosphoprotein specific dye, followed by MALDI-TOF identification. Methionine aminopeptidase (MAP) is a ubiquitous, essential enzyme involved in protein N-terminal methionine excision. According to the generally accepted cleavage rules for MAP, this enzyme cleaves all proteins with small side chains on the residue in the second position (P1′), but many exceptions are known. The substrate specificity of Escherichia coli MAP1 was studied in vitro with a large (>120) coherent array of peptides mimicking the natural substrates and kinetically analyzed in detail. Peptides with Val or Thr at P1′ were much less efficiently cleaved than those with Ala, Cys, Gly, Pro, or Ser in this position. Certain residues …

Nanoparticles of gold derivatized with monolayers of thiocarboxylic acid ligands, core/shell nanoparticle composites of gold and silica, gold-glutathioneclusters, and titania nanoparticles were synthesized and their surface properties (extent of ligand coverage, net negative charge, and zeta potential) characterized by capillary electrophoresis as a function of pH, ionic strength, and dielectric constant for the first time. Treatment of pulping wastewater by using a novel electrochemical reactor,i.e.the three phase fluidized bed electrode,reactor were invesigated.The effects of air flow rate,applied voltage,pH value and FeSO4 concentration on electrolysis efficiency were investigated.It was clearly shown that the 20 V of voltage,0.3 m3·h-1 of air flow rate,1000 mg L-1 of FeSO4 concentration and pH≈2.5 were its best conditions.COD removal could be 70.1% under these conditions.

The lithium iron phosphate cathode materials were synthesized by solid-state reaction combined high-rate ball milling under the temperature ranging from 400 ℃ to 700 ℃.The crystalline structure, morphology of particles, and electrochemical performance of the sample were investigated by X-ray diffraction, scanning electron microscopy and charge-discharge test. The results showed that the sintering temperature had great influences on the crystal structure, morphology and electrochemical performances of LiFePO4. The sample synthesized under 600℃ showed the best charge-discharge performances with the first specific discharge capacity of(128.8) mAh/g and the 15th specific discharge capacity of 129.1 mAh/g at 0.1C rate, …

(Chemical Equation Presented) The relative gas-phase energetics of several low-lying isomers of 1,7-dioxaspiro[5.5]undecane and 1,7,9-trioxadispiro[5.1.5. 3]hexadecane have been calculated with second-order Møller-Plesset perturbation theory and basis sets as large as aug-cc-pVQZ. Relative energies in THF, dichloromethane, acetone, and DMSO have been estimated with corrections from polarized continuum model calculations at the B3LYP/6-311+G(d) level. The analysis of proteins in biological fluids by capillary electrophoresis (CE) is of interest in clinical chemistry. However, due to low analyte concentrations and poor concentration limits of detection (CLOD), protein analysis by this technique is frequently challenging. Coupling preconcentration techniques with CE greatly improves the CLOD. An on-line preconcentration-CE method that can selectively preconcentrate any protein for which an antibody is available would be very useful for the analysis of low abundance proteins and would establish CE as a major tool in biomarker discovery. While thioacetate-terminated oligo(phenylene vinylene)s (OPVs) have been synthesized and employed in applications involving the formation of metal–molecule–metal junctions, the synthesis and application of potentially more versatile α,ω-dithiol OPVs have not previously been described. Here, a thiomethyl-precursor route to the synthesis of α,ω-dithiol OPVs is reported and their ability to form well-ordered self-assembled monolayers (SAMs) without the addition of exogenous deprotection reagents is described. α,ω-Dithiol OPV monolayers exhibit thicknesses consistent with molecular length and are nearly defect-free, as assayed by electrochemical measurements.
