Turkish Journal of Biology
The main purpose of this study was to investigate the effects of nutrient and macronutrient stress on complex variations in the phenolic profile and antioxidant status of Bellis perennis L. callus cultures. To obtain stress conditions, callus cultures were grown under insufficient nutrient conditions (different strengths of MS medium) or under conditions of limiting calcium (-Ca), magnesium (-Mg), or both (-Ca & -Mg). Phenolic content of callus cultures obtained from different stress treatments was detected quantitatively by LC-ESI-MS/MS analysis. The major compound was chlorogenic acid in all treatments. The accumulation of hesperidin, rutin hydrate, and chlorogenic acid was significantly induced …

The aim of this present study was to analyze global gene expression from a developing inflorescence of oil palm under complete defoliation stress. We used the Illumina platform to sequence six cDNA libraries of defoliated and nondefoliated tissues. The frequency of differentially expressed genes (DEGs) decreased with increasing fold change at P ≤ 0.05. Gene ontology analysis revealed that about 15% of all DEGs were responsible for stress response while 13.9% of genes were responsible for cell organization and developmental processes. DAVID functional analysis clustering of DEGs identified a higher enrichment score (ES) for cellular response to stress (ES = …

To create new varieties resistant to biotic and abiotic stress factors that possess high productivity and improved fruit quality, breeders require modern breeding methods and techniques. Double haploid (DH) lines are an invaluable breeding element due to their guaranteed, completely homozygous nature. One of the fastest methods for obtaining DH lines is anther culture. Microspore embryogenesis provides considerable opportunities in the areas of breeding and biotechnology and represents a model system for fundamental biological research. The effectiveness of pepper anther culture is still low, which affects the application of DH lines in breeding programs. A detailed study of the anther …

Sirtuins are NAD+-dependent deacetylases and ADP ribosyltransferases that are activated under stress conditions such as calorie restriction and starvation. Boron, which is mostly found in the form of boric acid (BA) or sodium borate (NaB), is known to bind NAD+. In this study, the effect of NaB on hepatoma cell line HEP3B was investigated by analyzing the proteins harvested from NaB-treated and serum-starved HEP3B cells. The NaB treatment (15 μg/mL) led to a decrease in the overall proteome acetylation and particularly in mitochondria, the synthesis rate of respiratory complexes, the amount of cellular reactive oxygen species, and the proliferation rate …

Glycation of proteins is a nonenzymatic process in which proteins react with reducing sugar molecules. This process takes place at ?-amino (?-NH+) groups of lysine or hydroxylysine residues as well as α-amino groups of N-terminal residues. In the present study glycation of ?-NH+ groups of lysines was computationally analyzed for 26 proteins based on their 3D structures. We determined the spatial relationship with acidic or basic residues and correlated them with the glycation prediction algorithm Netglycate 1.0 software, which employed primary structure exclusively for glycation site prediction. Of the lysines from 19 of the 20 proteins employed to build the …

Plant-sap sucking insects with deficient nutritional diets contain symbiotic microorganisms in a specific organ in the abdomen (bacteriome). Such symbionts were discovered to belong to various bacterial lineages. Ongoing genomic research concluded that these symbiotic organisms provide the essential nutrients that are absent in the insect’s diets such as amino acids and vitamins. While the identities and functions of several sap-sucking lineages have been studied, some lineages such as Fulgoroidea are not understood because recent results failed to confirm the presence of the symbiotic microbes even though older microscopy studies identified microbes in the bacteriomes. The objective of this research …

Amyotrophic Lateral Sclerosis (ALS) is a neurodegenerative disorder that occurs due to the death of motor neurons and leads to paralysis and death within three to five years after symptoms present (Byrne et al., 2013). Superoxide Dismutase 1 (SOD1) was first identified to be associated with ALS in 1993. The objective of this study is to determine which proteins interact with wild type and mutant SOD1 and find any similarities or differences between them. ALS is attributed to a gain of toxicity, therefore abnormal protein interactions in mutant SOD1 are important. The results of this study will provide insight on …

Insects with complex life cycles provide an ideal system for investigating the relationship between sex-skewing bacterial symbionts and the secondary loss of sexual reproduction. This research utilized the insect family Adelgidae, a group notorious for having peculiar and complex life cycles. In this family, some species have lost their sexual generation entirely, trapping them in asexual reproduction. Additionally, these insects are known to harbor multiple symbionts, both obligate and facultative. The array of facultative symbionts that have been detected includes the infamous sex-skewing symbiont Wolbachia. This symbiont is known to induce male killing, cytoplasmic incompatibility, and parthenogenesis in insects. Additionally, …

Many insects live on nutrient-poor diets; to compensate, they have evolved obligate associations with microorganisms that are transmitted directly between generations. The bacterial associates of some Hemiptera insects have been identified and understood, but several remain unknown. Our objective is to identify the diversity of bacterial endosymbionts in Issidae using modern microscopy and molecular methods, and compare their identities to known symbionts from related insects. To do so, we will extract DNA from insect samples, amplify bacterial symbiont 16S genes, sub-clone PCR products, sequence the products, and determine their phylogenetic relationships to other insect symbionts and free-living bacteria. Our study …

Aquatic prey subsidies entering terrestrial habitats are well documented, but little is known about the degree to which these resources provide fitness benefits to riparian consumers. Riparian species take advantage of seasonal pulses of both terrestrial and aquatic prey, although aquatic resources are often over-looked in studies of how diet influences the reproductive ecology of these organisms. Ideally, the timing of resource pulses should occur at the time of highest reproductive demand. This study investigates the availability of aquatic(mayfly) and terrestrial (caterpillar) prey resources as well as the nestling diet of the prothonotary warbler (Protonotaria citrea) at two sites along …

Neurodegenerative diseases like Alzheimer's disease (AD), Parkinson's disease (PD), and amyotrophic lateral sclerosis (ALS), include the progressive loss of structure and function of neurons leading to neuronal death. All of these diseases are fatal, as there is no cure for them. The causes of these diseases are unknown; however, there are many proposed mechanisms that lead to neurodegenerative diseases. Oxidative stress is the leading cause of cell death in neurodegenerative diseases, in addition to other mechanisms including endoplasmic reticulum stress, proteasome inhibition, nitrosative stress, inflammation and excitotoxicity. More understanding of the death mechanisms at work in neurodegeneration is necessary to …

Resting Superimposed on inherently high basal metabolic demands, the additional energetic requirements of reproduction can push female sea otters beyond physiological limits. Indeed, the resulting energy imbalance contributes to disproportionately high rates of mortality at the end of lactation in this species. To examine and quantify metabolic changes associated with reproduction, we measured the resting metabolic rate (RMR) of a female sea otter across gestation, lactation and non-reproductive periods. Concurrently, measurements were made on a non-breeding control female. Our results suggest that RMR declines during gestation. Conversely, RMR increases during lactation, reaches a peak at 3–4 months postpartum, and remains …

Evolutionarily conserved molecular processes involved in construction of the germline and embryonic development are essential for the procreation of many species. Infertility affects 15% of couples in the world and can be caused by dysfunctions during egg and sperm development, anatomic defects, as well as faulty embryonic development. Although there are some infertility disorders that are genetically defined, such as Turner and Klinefelter syndromes, many clinical infertility cases are diagnosed as idiopathic due to the lack of understanding of basic fertility mechanisms. Schmidtea mediterranea is a freshwater planarian species that has the ability to regenerate complete organisms, including germ cells …

Natural plant populations are often adapted to their local climate and environmental conditions, and populations of forest trees offer some of the best examples of this pattern. However, little empirical work has focused on the relative contribution of single-locus versus multilocus effects to the genetic architecture of local adaptation in plants/forest trees. Here, we employ eastern white pine (Pinus strobus) to test the hypothesis that it is the inter-genic effects that primarily drive climate-induced local adaptation. The genetic structure of 29 range-wide natural populations of eastern white pine was determined in relation to local climatic factors using both …

Hibernation is characterized by prolonged periods of inactivity with concomitantly low nutrient intake, conditions that would typically result in muscle atrophy combined with a loss of oxidative fibers. Yet, hibernators consistently emerge from winter with very little atrophy, frequently accompanied by a slight shift in fiber ratios to more oxidative fiber types. Preservation of muscle morphology is combined with down-regulation of glycolytic pathways and increased reliance on lipid metabolism instead. Furthermore, while rates of protein synthesis are reduced during hibernation, balance is maintained by correspondingly low rates of protein degradation. Proposed mechanisms include a number of signaling pathways and transcription …

Soil salinity is one of the most important limiting factors of agricultural productivity in the world. The Triticum aestivum salt tolerance-related gene (TaSTRG) possesses a functional response to salt and drought stress conditions. A variety of stress factors, such as salt, drought, abscisic acid, and cold, may induce the expression of TaSTRG in wheat. In this study, the TaSTRG gene was transferred to tobacco via Agrobacterium-mediated transformation. Overexpression of TaSTRG in transgenic tobacco plants indicated higher salt tolerance and mediated more vigorous growth than in wild-type plants. Under salt stress conditions, the transgenic tobacco plants had higher germination and survival …

Many molecular pathways have been proposed to mediate the beneficial effects of CR, however, the mechanism is still unknown. Circadian clock which is an internal time keeping system is regulated by feeding. Thus our aim was to study the effect of CR on the circadian clock. Here we show that CR significantly affects the expression of circadian clock genes in mice at the mRNA and protein levels, suggesting that CR reprograms the clocks at the transcriptional and …

Variation in the availability and distribution of food resources is a strong selective pressure on wild primates. We explored variation in Tibetan macaque gut microbiota composition during winter and spring seasons. Our results showed that gut microbial composition and diversity varied by season. In winter, the genus Succinivibrio, which promotes the digestion of cellulose and hemicellulose, was significantly increased. In spring, the abundance of the genus Prevotella, which is associated with digestion of carbohydrates and simple sugars, was significantly increased. PICRUSt analysis revealed that the predicted metagenomes related to the glycan biosynthesis and metabolic pathway was significantly increased in winter …
