Retrospective Theses and Dissertations

Dialysed activated sludge was used as a substrate to test for enzymes which can hydrolyse activated sludge floc. Two hundred and fifty aerobic and anaerobic microorganisms were tested against activated sludge for the presence of hydrolytic enzymes. These included known genera and organisms obtained by various enrichment procedures. Anaerobic digester mixed liquor was contacted with activated sludge agar under anaerobic conditions. None of the microorganisms present in the digester liquor hydrolysed the floc. In order to further elucidate the hypothesis that the telencephalon of ectothermic vertebrates functions primarily as a non-specific arousal mechanism, facilitating lower brain areas, (the Arousal Hypothesis) the South African clawed frog, Xenoous laevis, was studied in four behavioral situations before and after telencephalon ablation, olfactory bulb ablation or sham surgery. Particular attention was paid to measurements of the temporal aspects of the behaviors studied.

The behaviors studied were:

1. Habituation to an acoustic-vibratory stimulus

2. The optomotor response

3. Escape from shallow water

4. Feeding behavior

Live Trap Preference Among Grassland Mammals, Robert K. Rose, Norman A. Slade, James H. Honacki

In two independent studies, small mammals of grassland communities in eastern Kansas, when given a choice, preferred Fitch to Sherman live traps. Except for the harvest mice in the demographic study, the type of preferred trap and magnitude of selection was remarkably comparable, with microtines showing slight, and cricetines strong, preferences overall. The merits of each type of trap are compared, and related to season, weather, prior experience, and trap-associated mortality.

Microscopic examination of the tracheal dimensions of normal Florida mice (Peromyscus floridanus) and cotton mice (P. gossypinus) showed no significant differences between the two species, but external examination showed the tracheal length of the Florida mouse to be longer than that of the cotton mouse. Microscopic examination of the intrapulmonary apparatus (bronchioles, alveolar ducts, atria, and alveoli) of normal Florida and cotton mice showed no significant differences in measurements between the two species. Cotton mice were exposed to exhaust gases produced by the burning of solid rocket motor (SRM) fuel. Mice exposed once for a duration of 10 min demonstrated …

Studies on the helminth parasites of salamanders from California have been very limited.

Between September 1975 and April 1977, 340 salamander belonging to seven species, Aneides flavipunctatus (7), A. lugubris (7), Batrachoceps attenuatus attenuatus (250), Ensatina eschscholtzii eschscholtzii (52), E. e. sierrae (17), Taricha torosa (6), and Dicamptodon ensatus (1), were examined for helminths and found to harbor 10 species, four of which are new. The parasites found are: Cestoda: Distoichometra ensatinae sp. nov. from Ensatina e. eschscholtzii and E. e. sierrae; and Baerietta diana (Helfer, 1948) Douglas, 1958 from Batrachoceps a. attenuatus. Myosin and myosin light-chain kinase have been isolated and characterized from small quantities of normal and SV40-transformed, murine astrocytic neuroglial cells in culture and from intact normal mouse brain. Sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis of the astrocyte myosins revealed a heavy chain of 200,000 daltons and two light chains of 20,000 and 15,000 daltons. These myosins are similar to other cytoplasmic myosins. The astrocyte 20,000-dalton light chain can be phosphorylated by an endogenous myosin light- chain kinase which has properties similar to those of the myosin light-chain kinase found in human platelets. Nemerteans are common invertebrates along coastlines in both temperate and arctic climates throughout the world. Amphiphorus imparispinosus is a carnivorous member of this phylum that is found abundantly in the intertidal areas of the Oregon coast. As in many other invertebrate phyla, endocrine integration within the nemerteans is thought to be accomplished by neurosecretory mechanisms. In Amphiphorus the presence of these neurosecretory systems has not been previously shown. The dispersal and dispersion patterns of Hydra fusca were examined. Hydra were placed in petri dishes at various densities. The water in the dishes was swirled forcing the animals to the center. The location of each animal was marked at time intervals thereafter. Analyses of the dispersal rates and dispersion patterns were made. Hydra dispersed from a central release point at a non-random rate. There was rapid movement from the center followed by a minimal daily movement. Somatic cell hybrids were constructed between BALB/c-RAG mouse cells and feline lymphoma cells by the hypoxanthine-aminopterin-thymidine selection scheme. RAG cells spontaneously produce an endogenous B-tropic type C virus. Cat-mouse hybrids preferentially segregate feline chromosomes and retain murine chromosomes,demonstrable by karyotypic and isozyme analyses. Despite the presence of the complete mouse genome, including the viral genome, virus production was diminished to 1-5% of the levels observed in RAG parents based upon particle-associated RNA-dependent DNA polymerase (reverse transcriptase) activity in the culture fluid. With the expanding use of plutonium-239 (239Pu) as a nuclear reactor fuel and increased recycling of spent reactor fuel to recover 239Pu, the possibility of an accidental human exposure exists. Little information is available on the effects of inhaled 239Pu on immune mechanisms. This study was performed in order to examine the effects of 239Pu dioxide (239Pu02) inhalation on the ability of mice to be immunized against a bac­terial respiratory infection in which cell-mediated immunity is the primary mechanism of host resistance to infection. Eighty-three Didelphis virginiana from the Portland metropolitan area were collected and studied from December 1974 through May 1976. Males averaged 3394 g, females 2318 g. An annual sex ratio of 68.7:31.3 was recorded. Data indicate the possibility of a year-round breeding season and suggest the occurrence of three peaks of reproductive activity: January-February, April-May and October-November. The six primary foods consumed annually are: leaf litter, mammals, gastropods, earthworms, grasses and pet food. Seasonal composition and diversity in the diet is marked. The scorpion Paruroctonus aquilonalis (Stahnke) was studied to better understand the influence that six potentially important factors (size, sex, temperature, nutrition, season, and humidity) have on its respiratory metabolism. Respiratory rate: temperature relations of oxygen consumption from 5°C to 36°C, and standard metabolism estimates at 20°C and 25°C were employed. Respirometry was performed on both freshly collected mature and immature scorpions and on laboratory-maintained animals.

Inverse weight: metabolism relationships were noted for males and females above 24°C, but below 24°C live weight did not influence metabolic rate among females. For mature males positive weight: metabolism patterns were found below 24 …

Thermal ecology and movement of the Leopard Lizard, G. wistizeniwere studied at Frog Spring, adjacent to the Alvord Desert, and at various locations throughout the Alvord Basin. Seasonal activity is known to occur from early May to mid-October, but abundance. gradually decreases after late July. During favorable late spring, and summer weather lizards exhibit a bimodal pattern of diel activity with some activity occurring at all hours of the day between 0600 and 1800 H. The nutritional conditions which permit maximum yield of trans-2,3- epoxysuccinic acid by Paecilomyces varioti NPRL 1123 were investigated. Copper and iron ions were added to decationized fermentation media to determine the optimum concentration and ratio of the ions for the accumulation of trans-2,3-epoxysuccinic arid. P-vitamin and amino acid studies were done by single omission of the growth factors in culture media. Organic acids in the cultures were recovered by ether extraction and identified by gas chromatography.

Repeated exposure to ultraviolet radiation brings about an increase in the DOPA-positive melanocyte population of the human. However, there is an age-dependent decline in the melanocyte population regardless of UV irradiation. In the rhesus monkey, a drop in the DOPA-positive epidermal melanocyte population has been observed after prolonged UV irradiation. The population of DOPApositive epidermal melanocytes in PET/Wmr mice reaches a peak during the first post-natal week after which the number of cells declines, fading almost completely within 3 weeks. The effects of constant photoperiod (14 hours of light) and decreasing photoperiod (13-8 hours of light) on the regressive phase of the ovarian cycle of the lizard Anolis carolinensis were investigated. Changes in body, liver, ovary and oviduct weights were measured and microscopic examinations of several ovarian parameters were made. In both light regimens body and liver weights decreased over the first four weeks of the experiment but both increased at the sixth week. The effects of porcine follicle stimulating hormone, 17B-estradiol, and a combination of the two on the process of vitellogenesis were studied in Anolis carolinensis maintained at 15C with no light for one week, and then at 15C or 32C with 14 hours of light for two additional weeks.

At both temperatures, the control animals subjected to 14 hours of light possessed greater numbers of previtel­logenic follicles and lower liver weights than the control animals sacrificed after one week in the dark. The relationship between lactic acid concentration and twitch tension was reevaluated in electrically stimulated frog sartorius muscle. If muscles stimulated under anaerobic conditions at a rate of 30 stimuli/min, contractile force decreased to 36% of the initial value in 15 min. Concomitantly lactate increased from 3.3 to 18.7 μmol/g of muscle. The correlation between the increase in lactate and the decrease in contractile force was significant (r = -0.99, P < 0.000001). Recovery occurred in two phases. A rapid increase in contractile force, which represented 20% of the total recovery, took place during the first 15 s and occurred concomitantly with an increase in ATP from 3.9 to 4.6 μmol/g. Lactate concentration did not change significantly during this period. The second phase of recovery of contractile force was complete in 50 min. Lactate concentration and contractile …

It has been thought for many years that the alveolar macrophage is the primary agent responsible for prevention or bacterial invasion or the lung. Numerous studies with Staphylococcus aureua, Staphylococcus epidermidis, Escherichia coli. Klebsiella, and Proteus have been cited to substantiate this statement. Together these organisms cause less than ten per cent of human bacterial pneumonias. Both in vivo and in vitro studies have shown that the alveolar macrophage is capable of bacterial clearance with time. Sadly lacking ss evidence that alveolar macrophages readily phagocytose Streptococcus pneumoniae which are responsible tor greater than ninety percent of all bacterial pneumonias. The …
