The most desirable way of treating endocrine deficiencies would be by the successful transplantation of functional endocrine tissue into the deficient animal. However, unless the donor and recipient are identical twins or from an inbred strain of animals, an immune response is usually elicited in the recipient by the foreign tissues leading to rejection of the transplant. Thyroid gland cells which have been enzymatically dispersed from thyroid tissue and cultured in vitro are capable of reforming functional thyroid tissue when implanted into animals of the same strain. Therapeutic lung lavage is a procedure by which the interior of the lung of a living animal is “washed” with a suitable solution. Broncho­pulmonary lavage, a special type of lung lavage in which the bronchioles and alveoli are washed, is the major concern of this thesis. Broncho­pulmonary lavage was investigated in 1920 as a possible treatment for lungs damaged by irritant gases and in the late 1950‘s as a method of hemodialysis. Currently, it is effectively used for the treatment of patients with obstructive lung disease and for the removal of radioactive materials from the mammalian lung. For our investigation, an established line (E line) of baby hamster kidney cells which was transformed by the incorporation of Simian Virus 40 into the cellular genom was used. The induction of this virion by irradiating these cells with ultraviolet light was studied. The purpose of this thesis was to determine the wavelength dependency (at four selected ultraviolet mercury emission lines) of the induction of the SV40 genome from these cells. The viral titres of induction were measured by the agar overlay plaque assay method and by the fluorescent antibody microassay technique.

Studies reporting a high concentration of WR-2721 in mouse salivary glands led to our studies of possible radioprotection of these glands by this drug from ionizing radiation. Oral effects of radiation in the presence of WR-2721 were studied in mice and dogs. Histological evaluation of mouse salivary glands irradiated with 1000 rads of 60Co showed essentially no difference between control and experimental animals. Almost full regeneration of the serous salivary components occurred by 6 months in both groups and neither group had changes in the mucous glands. The effect of temperature on calcification in Bossiena orbigniana orbigniana (Decaisne, 1842) [Manza, 1940; Silva, 1957] (Corallinaceae) was studied using calcium-45 labeling of terminal segments under laboratory conditions. B. orbigniana has optimum calcium uptake for the temperatures 17-21°C within a temperature range of 3-37°C. These results suggest that optimum calcification in B. orbigniana should take place from late spring to early autumn. Q10 values for 3-21°C averaged 3.39 indicating an enzyme-governed reaction. It is proposed that thallus growth rate and calcification are not directly related. It is believed that the optimum temperatures (17-21°C) affect the enzymes towards an increase …

Prairie rattlesnakes, of the species Crotalus viridis virldis, were orally, infected with tetrathyridia of Mesocestoides corti to determine the host response to the parasite. Snakes were maintained at constant temperatures of 25, 30, and 35 C for 4--24 weeks. Blood chemistry evaluations of the host were made by standard hematological methods. Eosinophil counts, total proteins, and serum gamma globulins increased while packed-cell volumes and hemoglobins decreased. Autopsy studies revealed the first documented asexual multiplication of Mesocestoides corti in a laboratory infected ectothemic host, with the small intestine being the preferred organ. The preference for a rocky situation was analyzed under laboratory conditions. In three-compartmented choice boxes, Peromyscus californicus and P. eremicus chose a compartment with rocks and a sandy floor over a compartment with just a sandy floor but P. maniculatus did not show a preference. In a similar experiment, in continual total darkness, the three species chose the compartment with the rocks. UM-key did not prefer the compartment with rocks when the rocks were encased in plastic and were under a normal light regime. A study was undertaken to investigate the application of tissue culture and histochemical banding techniques normally used in mammalian studies to fish cells and to examine the mechanisms of chromosome evolution and speciation in salmonids by comparing the chromosomes of the hybrid to those of the parental types. The application of banding techniques was generally unsuccessful in revealing bands, though quinacrine staining did reveal the presence of two fluorescent spots in the vicinity of the centromere. These were thought to be heterochromatic blocks, possibly related to the high incidence of centric fusion or fission in salmonids. Results of chromosome analysis …

The Relationship Between Gill Na+ + K+ -Activated Adenosine Triphosphatase Activity And Sodium Regulation In Acclimation Of The Blue Crab, John Lewis Harris

Evidence from this study suggests that Na++K+-activated ATPase is involved in the ionic regulation of sodium in the hemo-lymph of the blue crab in low salinity environments. The time course of changes in gill Na++K+-ATPase specific activity follow- ing transfer of crabs from JO o/oo to 5 o/oo salinity indicates that a major increase in activity occurred within 2.5-3.0 hr. Apparently, a 8.8-11.1% decrease in hemolymph sodium ion concen-tration to about J55-J60 meq/1 in approximately 2-J hr was sufficient to activate the ATPase cation transport system in the gills of Callinectes sapidus. The effect of bilateral eyestalk ligation or ablation on the blood Na+ and ca++ concentrations of the euryhaline blue crab, Callinectes sapidus, was measured in 10 o/oo and 5 o/oo sea water. The ablations caused a significant decrease in the blood Na+ and ca++ over a 24 hour period. The serum Na+ level increased significantly 6 hours following injections of an eyestalk extract whereas the blood total ca++ level did not change significantly. Eyestalk ligation, however, did not cause significant blood Na+ and ca++ changes. The blood Na+ level of intact crabs increased significantly in acclimating to colder water when …

Nitrogen cycling was studied in the hypersaline northern arm of the Great Salt Lake (332,480 mg/l total dissolved solids) using a sediment-water microcosm simulation of that system. The study demonstrated that nitrate, ammonia, and urea were not stimulating to the halophilic bacteria in the microcosm. The bacteria were simulated only indirectly simulated bacterial growth. In addition, no nitrification could be demonstrated during the study.

Weekly analyses of water samples from the north arm of the lake itself showed that nitrate and nitrite were absent, leaving ammonia as the only significant form of inorganic nitrogen in the northern lake basin. Other …

This study compares insect and cattle consumption of crested wheatgrass (Agropycon spp.) on a site in Sterling Ranch, Utah County, Utah. The hypothesis tested is that insect consumption in general, and specifically consumption by Labops hesperius Uhler, significantly reduces total available cattle forage.

Labops and grasshoppers were probably the major cause of secondary damage at the study site, while the impact of thrips is unknown. A detrimental impact on range was suggested by the data, as insects consumed forage equivalent to 2.8 animal units per month, while cattle consumed forage equivalent to 2.1 animal units per month at the …

The effects of hibernation on learning ability and retention were studied in Myotis velifer, a small cave bat. Bats were maze trained using illumination intensity as visual discrimination and food as positive reinforcement. In experiment one, bats placed in a hibernaculum at 7 C for intervals of 4 or 10 days after learning had memory retention similar to control animals maintained at 31 C. In experiment two the effect of placing animals daily in a 7 C hibernaculum for 5-7 hr immediately after learning was studied. The androgenic glands of the Pacific crayfish, Pacifastacus leniusculus trowbridgii Stimpson, 1859, were studied experimentally and by light and electron microscopy. The androgenic glands proliferate in the spring of the year concurrently with an increase in mitotic activity in the testes. Degeneration of a major portion of the glands occurs in the fall, when spermatogenic activity in the testes is near completion. Cellular development appears to progress from small undifferentiated cells through stages of vacuolization and hypertrophy and terminates with degeneration or lysis. Large differences in certain morphological traits between populations of brown trout (Salmo trutfa L.) in the lakes of northern Sweden led us to initiate an electrophoretic examination in an effort to understand the genetic structure of these populations. A total of 549 brown trout from four lakes were examined at 10 electrophoretically detected loci. Genetic variation was found at only three loci, coding for lactate dehydrogenase (LDH), alpha-glycerophosphate dehydrogenase (AGP), and creatine phosphokinase (CPK). A total absence of heterozygotes at the LDH-I locus in Lake Bunnersjoarna, coupled with significant correlations between LDH-I type and both CPK-I allele frequencies and …

The genetic control of the supernatant form of aspartate aminotransferase (AAT) was studied in the cutthroat trout (Salmo clarki) through a series of experimental matings. 509 individuals of eight families were examined to determine (1) the number of loci, (2) the mode of inheritance (i.e. disomic or tetrasomic), and (3) the linkage relationship of the loci involved. The variation observed is controlled by a duplicated locus resulting from a presumed tetraploid event of an ancestral salmonid. Fertile White Leghorn (Gallus gallus) and Bobwhite Quail (Colinus virginianus) were subjected to 15 min exposures produced by burning solid rocket motor (SRM) fuel. Comparative mortality data were collected. Chicken eggs were further used to study the effects of exposure on water relations and blood gas parameters. Chicken embryos exposed once on day 19 or incubation demonstrated and LD50 of 204 ppm; the LD50 for quail embryos was 175 ppm. When mortality was regressed on the final exposure concentration, chicken and quail embryos exposed on days 12 and 19 showed LD50's of 127 and 86 ppm respectively, and embryos exposed …

Four eastern Kansas populations of the prairie vole, Microtus ochrogaster were live trapped from 1970—73 to gain insight into the population regulation of this species. All four populations exhibited a 2—yr cycle in numbers with peak densities generally occurring in June 1972. Peak densities were followed by a decline in numbers, a recovery, and a population crash in spring 1973. Reproductive parameters changed dramatically as density rose and fell. The summer breeding season in the crash year of 1973 was shortened by at least 3 mo. A reduction in breeding activity occurred during the summer of every year of the …

Coyote food habits were ascertained by identifying the undigested material in 1143 scats collected at monthly intervals from specific road transects on the USERDA Hanford Reservation in Washington from 1974 to 1976. Tracks at artificially established scent posts along the principal transect provided evidence that the scats were left by coyotes and not bobcats or badgers. On the basin plain (150m elevation) where the Aptemisia tpidentata/Poa sandbepgii Association predominates, the average monthly percent occurrences in the 491 scats collected in 1975 2 were pocket mouse 60, leporid 39, pocket gopher 12, grasshopper 12, fruit 10, cricetine mice 7, darkling beetle …

This study of mammals of the Bull Run Planning Unit has a dual character. First, mammals of special scientific or natural history interest such as threatened or endangered species were sought. This was in conjunction with a Mt. Hood Bull Run Planning Unit. Second, a zoogeographical study of mammals of the Bull Run was performed. Abundance and distributional data was recorded for each species observed. This data was related to availability to moisture as indicated by plant associations trapped. An examination of 24 genera of aquatic plants having floating leaves and leaf-like thalli has been made. Functional stomata occur on the adaxial sides of floating leaves of angiosperms and some ferns, and in some floating leaves there are stomata on the abaxial surface as well. Most floating leaves have prominently chambered mesophyll, and in some instances the chambers are locally enlarged and form buoys.

The boundaries of the project encompass sections of North Las Vegas, Las Vegas, East Las Vegas and areas along Las Vegas Wash extending to its drainage point in Las Vegas Bay in Lake Mead. The incorporation of phosphoproteins and phenol-bound proteins into the cuticle of Periplaneta americana was analyzed. Radioactive monosodium phosphate was injected into newly ecdysed cockroaches. Incorporation into haemolymph proteins as well as tanned cuticle was observed. Phosphoproteins were observed to be combined with the cuticle to a greater extent than monosodium phosphate. This suggests that phosphate is being incorporated into the cuticle more readily than monosodium phosphate. The relevance of these observations is discussed in light of phosphate being a possible crosslinking agent between chitin and cuticle protein.

The previous cytological and electrophysiological work suggests that the optic tectum matures during metamorphic climax. It was the aim of the present work to attempt to compare pre metamorphic (Stage XII) larval and post metamorphic juvenile, Rana catesbeiana, using a photostimulated evoked electroencephalographic (E.E.G.) response in the optic tectum and correlate that with tissue studies to verify neurophysiological maturation of the optic tectum.
