The increase in blood levels of GLP-1 with dietary resistant starch is thought to be associated with the activities of hypothalamic NPY/AgRP neurons and the gene expression of glucokinase in the ARC of the hypothalamus. Exendin-4 has been shown to be associated with fatty acid synthesis and oxidation. In this project, it was proposed that 1) food intake will be decreased in GLP-1R KO mice, the glucokinase (GK) mRNA expression in the liver and in the hypothalamus, the hypothalamic NPY mRNA expression will be up-regulated by exendin-4 treatment; and 2) exendin-4 will decrease fatty acid synthesis and increase fatty acid …

Angiogenesis is the process of developing and elongating blood vessels. In obesity, angiogenesis controls the development of the adipose tissue, allowing it to expand as energy stores increase. When angiogenesis is blocked in rodent models, adipose tissue not only stops expanding, but regresses, proving a possible mechanism for weight loss. Nt, a Chinese herbal decoction, and gallic acid, an active antiangiogenic ingredient in Nt, were tested in clinical trials in combination for possible use as a supplement for weight loss. Diets are typically poorer and risk of chronic disease is greatest in low-income populations. A relationship has been established in the literature between food costs and diet quality, where lower cost diets are generally those of the poorest quality. Food group intake, energy/nutrient intake, and diet cost were assessed in 64 female food stamp recipients in Southeast Louisiana. From one 24-hour dietary recall collected at the beginning of the monthly resource cycle (Day 1) and one at the end (Day 2), nutrient intakes and diet costs were able to be analyzed between different time frames. It has been proposed that the glucose sensing mechanism in the hypothalamus and hindbrain is similar to pancreatic â cells, and brain glucose sensing may be involved in the regulation of food intake. For the first part of the dissertation, it is proposed that molecules involved in â-cell glucose sensing, including glucokinase (GK), glucose transporter GLUT2, sulfonylurea receptor-1 (SUR1), glucagon-like peptide-1 receptor (GLP-1R), and the feeding-related neuropeptide Y (NPY), are colocalized in specific areas in the hypothalamus and hindbrain.
