The first mammalian neuropeptide to be characterized was substance P (sP) , and it is now recognized that sP is a member of a structurally related family of peptides, the tachykinins. Extensive studies have demonstrated. that sP and some related tachykinin peptides play key roles as neurotransmitters and neuromodulators. The synthesis of different members of the tachykinin family is in part due to the modifications' of three sP-encoding preprotachykinin (PPT) mRNA's that are derived from a single sP gene. Within the last five years, there has been increasing interest in the role of neuropeptides in the neuroregulation/modulation of both male and female sexual behavior. The impetus for this derives from several findings. Firstly, certain neuropeptides and their receptors are located in brain loci previously implicated in both male and female sexual behavior. Secondly, and most important, is the rapidly accumulating evidence that the concentrations of certain peptides within these behaviorally-relevant loci are regulated by gonadal steroids .

Substance P (sP), a neuroactive peptide, has now been implicated in a wide range of behaviors. One in particular is female rat sexual behavior. Recently, it has been shown that bilateral injections of sP into the dorsal midbrain central gray (dMCG) of estrogen-primed female rats facilitate sexual behavior (lordosis). Uncertainty remains, however, concerning the mode of action of synaptically released sP. Indeed, most of our understanding of sP activity has revealed that its full structural sequence is not needed for biological activity. In order to examine the role of the medial zona incerta in female sexual behavior, ovariectomized estrogen and progesterone treated female rats were tested for sexual receptivity following bilateral injections of the selective neurotoxin, ibotenic acid (3 ug/0.3 ul dissolved in phosphate-buffered saline) directly into the medial zona incerta. These injections produced a significant attenuation of lordosis behavior in highly receptive females when compared to saline injected controls (shams). This decrease in sexual receptivity was also reflected in a significant increase of rejections of male mount attempts. Behavioral experiments examined the role of morphiceptin in male rat copulatory behavior. Male copulatory behavior was recorded subsequent to bilateral injections of either 10, 500, or 1000 ng of morphiceptin into the medial preoptic area (MPOA) in sexually vigorous male rats. In the first experiment, all three doses of morphiceptin injected bilaterally into the MPOA produced a dramatic delay in the initiation of copulation. Both mount and intromission latencies significantly increased following injections of all three doses of morphiceptin when compared to saline injected controls. No other parameter was affected. Since the discovery of the law of effect, psychologists have been investigating a number of the parameters of reinforcing stimuli. The law of effect states that a response is learned, or not learned, depending upon the events (effects) that follow it. To evaluate the aspects of reinforcing stimuli. typically an organism is deprived of food or water and an increase in the probability or speed of a response is noted when small amounts of food or water are made to follow a response. Mixed and multiple aperiodic and ratio schedules of reinforcement were maintained by rats which had stainless steel electrodes surgically implanted in specific areas of their midbrains. The "reinforcing" stimulus used was a 100-cycle biphasic, square wave of a one-second duration. The voltage ranged from 1.0 to 2.0 volts and the current from .10 to .50 milliamperes. Four subjects (S's) were used in the study. Three of the S's received electrical brain stimulation (EBS) and the fourth received food pellets as reinforcement. Only the latter "food control" animal ,was run on a 36-hour food deprivation schedule. All other subjects were not …
