In this paper, we study the Poisson-gamma model for recruitment time in clinical trials. We proved several properties of this model that match our intuitions from a reliability perspective, did simulations on this model, and used different optimization methods to estimate the parameters. Although the behaviors of the optimization methods were unfavorable and unstable, we identified certain conditions and provided potential explanations for this phenomenon and further insights into the Poisson-gamma model.

The dissertation comprises two projects related to causal inference based on observational data. In healthcare research, where abundant observational data such as claims data and electronic records are available, researchers often aim to study the treatment effect and the pathway of that effect. However, estimating treatment effects in observational data presents challenges due to confounding factors. The first project focuses on estimating continuous treatment effects for survival outcomes, while the second concentrates on mediation analysis, allowing the exploration of the pathway of the causal effect. This dissertation is in the field of Nonparametric Derivative Estimation using
Penalized Splines. It is conducted in two parts. In the first part, we study the L2
convergence rates of estimating derivatives of mean regression functions using penalized splines. In 1982, Stone provided the optimal rates of convergence for estimating derivatives of mean regression functions using nonparametric methods. Using these rates, Zhou et. al. in their 2000 paper showed that the MSE of derivative estimators based on regression splines approach zero at the optimal rate of convergence. In the paper, the empirical process in informative model of random censorship from both sides is investigated. For it, the limit Gaussian process with mean zero is founded. Under investigating of empirical process, the characterization properties of the considered informative model is used. Life-testing and reliability experiments are commonly used reliability assessment methods to gain sound knowledge about product or system lifetime distributions. Usually, a sample of items of interest is subjected to stresses and environmental conditions that characterize the normal operating conditions. During the life-test, successive times to failure are recorded and lifetime data are collected. Rocket is a crash game developed by DraftKings, an American publicly traded online casino, sports betting and fantasy sports company. DraftKings Rocket is a game played with a rising rocket. In system reliability, practitioners may be interested in testing the homogeneity of the component lifetime distributions based on system lifetimes from multiple data sources for various reasons, such as identifying the component supplier that provides the most reliable components.

In the first part of the dissertation, we develop distribution-free hypothesis testing procedures for the homogeneity of the component lifetime distributions based on system lifetime data when the system structures are known. In this work, we provide an overview of the Cox proportional hazards model for time to event or survival analysis and the notion of propensity score matching to deal with confounding factors. A full analysis is reported in Chapter 2 concerning mortality for in-center dialysis patients with sickle cell disease to demonstrate the application of a general analysis strategy that has some logistical benefits over more traditional approaches to accounting for confounding variables. In this paper, a New Generalized Gamma-Weibull (NGGW) distribution is developed by compounding Weibull and generalized gamma distribution. Some mathematical properties such as moments, Rényi entropy and order statistics are derived and discussed. The maximum likelihood estimation (MLE) method is used to estimate the model parameters. The proposed model is applied to two real-life datasets to illustrate its performance and flexibility as compared to some other competing distributions. The results obtained show that the new distribution fits each of the data better than the other competing distributions.

In this paper, Weibull-Linear Exponential distribution (WLED) has been investigated whether being it is a well-fit distribution to a clinical real data. These data represent the duration of remission achieved by a certain drug used in the treatment of leukemia for a group of patients. The statistical inference approach is used to estimate the parameters of the WLED through the set of the fitted data. The estimated parameters are utilized to evaluate the survival and hazard functions and hence assessing the treatment method through forecasting the duration of remission times of patients. Inverse probability weighting is a popular technique to accommodate selection bias due to non-random sampling and missing data. In the first chapter, we develop an inverse probability weighted estimator and an augmented inverse probability weighted estimator of regression coefficients for a linear model with randomly censored covariates, when the censoring mechanism may be dependent on the outcome. We investigate the asymptotic properties of both estimators and evaluate their finite sample performance through extensive simulation studies. However, the use of antibiotics can contribute to the development of resistance. In the United States (U.S.), approximately 2.8 million people are infected with antibiotic-resistant bacteria each year, and more than 35,000 people die as a result.
