NADPH-cytochrome p450 oxidoreductase (CYPOR) is a membrane-bound protein in living cells. CYPOR delivers electrons to cytochrome p450 proteins (CYPs) to catalyze metabolism of drugs and synthesis of steroids. Extraction and solubilization of CYPOR from the membrane is typically done with the TritonX-100 detergent. The amount of the solubilized protein by this detergent, however, remains relatively low to structurally analyze CYPOR with NMR spectroscopy. The goal of the first project in this thesis was to optimize the amount of the extracted CYPOR from the E. coli membrane using various detergents and additives. Cytochrome P450 is a broad class of heme monooxygenase enzymes which catalyze various oxidative transformations. There are two main kinds of mammalian P450s: steroidogenic and drug metabolizing P450s. The first project involves a steroidogenic P450, CYP17A1, occupying a central role in the biosynthesis of steroid hormones. It catalyzes hydroxylation reaction on pregnenolone and progesterone, generating 17OH-pregnenolone and 17OH-progesterone, presumably utilizing a “Compound I” species. However, these hydroxylated products can be further processed in a second oxidative cycle to cleave the C17–C20 bond to form dehydroepiandrosterone or androstenedione, respectively, a crucial step in androgen production. The scientific community is interested in furthering the understanding of shock wave structures in water, given its implications in a wide range of applications; from researching how shock waves penetrate unwanted body tissues to studying how humans respond to blast waves. Shock wave research on water has existed for over five decades. Previous studies have investigated the shock response of water at pressures ranging from 1 to 70 GPa using flyer plate experiments. There is an interest in producing accurate and reliable computer simulations to predict the dynamic behavior of heterogeneous materials and to use these simulations to gain further insight into experimental results. In so doing, a more complete understanding of the multiple-length scales involved in heterogeneous material compaction can be obtained. Mesoscale computer simulations of dynamically shocked materials have proven to be a beneficial resource in unraveling data not observed in planar shock impact experiments, such as stress and temperature interactions between grains. The use of analytical and numerical techniques in solving the coupling of evanescent modes in a microwave waveguide through slots can be optimized to create a uniform magnetic field excitation on axis within a waveguide. This work has direct applications in Electron Paramagnetic Resonance (EPR) where a 100~kHz time-varying magnetic field is incident on a sample contained in a microwave cavity. Typical cavity designs do not take into consideration the uniformity of the 100~kHz field modulation and assume it to be uniform enough over the sample region from quasi-static principles.
