Gc×Gc-Ms And Parallel 2dlc-Ms Based Untargeted Metabolomics., Md Aminul Islam Prodhan

Electronic Theses and Dissertations

Untargeted metabolomics aims to analyze as many metabolites as possible in a biological system. Due to the vast complexity of the metabolome, bioanalytical platforms have been developed for untargeted metabolomics. While comprehensive two-dimensional gas chromatography mass spectrometry (GC×GC-MS) has been used in different scientific fields, its use in untargeted metabolomics is limited due to several challenges, such as complicated data analysis and limited accuracy of metabolite identification. Two-dimensional liquid chromatography mass spectrometry (2DLC-MS) has also been employed in untargeted metabolomics, but with limited metabolite coverage. This dissertation describes the efforts in developing new data analysis algorithms to improve the accuracy …

Synthesis And Evaluation Of Novel Silica Hydride-Based Stationary Phases For Bioanalytical Applications Sep 2019

Synthesis And Evaluation Of Novel Silica Hydride-Based Stationary Phases For Bioanalytical Applications

Seiichiro Watanabe

Most HPLC columns are packed with silanol (Si-OH)-rich type-B silica that often
participates in undesired electrostatic interactions with sample solutes and produces a poor analyte peak shape. These silanols are also known to facilitate the surface hydration required for HILIC mode to retain polar analytes. However, the hydrated surface composition can easily fluctuate, which results in poor reproducibility and requires a lengthy equilibration step. In this study, four novel stationary phases have been developed by using the TYPE-CTM silica material, in which up to 95% of the surface silanols have been replaced with Si-H group. One of the advantages of …

Nmr Metabolomics Protocols For Drug Discovery, Fatema Bhinderwala, Robert Powers Jan 2019

Nmr Metabolomics Protocols For Drug Discovery, Fatema Bhinderwala, Robert Powers

Chemistry Department: Faculty Publications

Drug discovery is an extremely difficult and challenging endeavor with a very high failure rate. The task of identifying a drug that is safe, selective and effective is a daunting proposition because disease biology is complex and highly variable across patients. Metabolomics enables the discovery of disease biomarkers, which provides insights into the molecular and metabolic basis of disease and may be used to assess treatment prognosis and outcome. In this regard, metabolomics has evolved to become an important component of the drug discovery process to resolve efficacy and toxicity issues, and as a tool for precision medicine. A detailed …
