Reaction rates of catalytic cycles over supported metal catalysts are normalized by the exposed metal atoms on the catalyst surface, reported as site time yields which provide a rigorous standard to compare distinct metal surfaces. Defined as the fraction of exposed metal surface atoms to the total number of metal atoms, it is important to measure the dispersion of supported metal catalysts to report standardized rates for kinetic investigations. Contamination of groundwater, initiated from years of human negligence, continues to persist despite major advancements in water remediation technologies. The generated groundwater plumes can continuously replenish contaminants such as dense nonaqueous phase liquids (DNAPLs) through the aid of slow-moving water. Thus, long-term treatment of this flow system has proven to be problematic. Surface enhanced Raman spectroscopy (SERS) has been widely applied for rapid and sensitive detection of various chemical and biological targets. Here, we incorporated a syringe filter system into the SERS method to detect pesticides, protein toxins and bacteria in water. For the detection of chemical and protein targets, silver nanoparticles (Ag NPs) were aggregated by sodium chloride (NaCl) to form nanoclusters that could be trapped in the pores of the filter membrane to from the SERS-active membrane. Researchers at Idaho National Labs have noted unexpectedly high Cd content in empty cladding hulls after processing in the Mark-IV ER. It has been theorized that Cd metal is transporting from the LCC pool through the eutectic LiCl-KCl salt bath to the anode baskets containing the empty hull where it is retained as a Zr-Cd intermetallic. This study sought to replicate the Cd contamination in a dry Ar glovebox using small-scale analogue of the Mark-IV ER salt-Cd metal system.

This thesis reports the development of optical sensors to analyze alcohols in fuel samples. One optical sensor enables the analysis of phenol in aviation fuels and cellulosic biofuels. It is critical to monitor the concentration of phenols in fuels because they increase the presence of solid oxidative deposits and lower the thermal stability of fuels. Preliminary studies of another optical sensor have been conducted to detect ethanol in E10 gasoline samples. "Single particle (SP)-ICP-MS methods were developed to characterize and quantify Ti-containing, titanium dioxide, silver, and gold NP concentration, size, size distribution and dissolved metal element concentration in surface water and treated drinking water. The effectiveness of conventional drinking water treatments (including lime softening, alum coagulation, filtration, and disinfection) to remove NPs from surface water was evaluated using six-gang stirrer jar test simulations. Six-gang stirrers were used to simulate drinking water treatments including lime softening, alum coagulation, powdered activated carbon sorption, filtration, and disinfection by free chlorine. This study evaluates four methods for dissolving complex glassy debris resulting from nuclear detonations. The samples of interest simulate the glassy debris generated from a nuclear detonation’s fireball coming in contact with solid masses. Each method attempts to achieve dissolution through different approaches involving either acid digestion, alkaline digestion, or molten salt fusion. Two of the four methods were modified to retain all elements of the debris or surrogate debris. A major cause of concern for estrogens found in the environment is that even at very low concentrations (10-100 ng/L) they can have a negative effect on aquatic ecosystems by disrupting endocrine systems. Due to the increasing importance of monitoring the levels of estrogens in the environment, a detection method was developed to identify and quantify three natural estrogens (estrone, 17β-estradiol and estriol) using gas chromatography with flame ionization detection (GC-FID). Eastern hemlock hosts the hemlock woolly adelgid, an introduced sap-feeding insect that causes rapid deterioration of the host. Like most conifers, eastern hemlock produces a variety of constitutive and induced defenses, primarily terpenoids. To explore the relationship of terpenoid defenses with adelgid infestations, we artificially infested hemlocks at a forest site and a plantation site, and compared their terpenoid concentrations to those in control trees. Infested trees showed lower terpenoid concentrations than control trees, suggesting that eastern hemlock not only fails to induce production of terpenoids in response to adelgid infestation, but becomes less able to produce carbon-based defenses due …

Gold nanoparticles produced using the Turkevich method were able to have their hydrophobicity modified using octanethiol in a novel method for SERS application. Both amphiliphic GNPs and hydrophobic GNPs were produced and differentiated by Raman signals. The amphiliphic GNPs were able to enhance the SERS signals of the protein emulsifier in the emulsion in situ and the hydrophobic GNPs were able to enhance the SERS signals from canola oil. Further purification of the hydrophobic GNPs proved to have higher enhancement and sensitivity, but still poor consistency which is typical of SERS. The “chemical nose” sensing method using supramolecular complexes composed of cationic monolayer functionalized gold nanoparticles (AuNPs) and fluorescent proteins (FPs) can successfully distinguish serum proteins, mammalian cells, tissue lysates, and chemotherapeutic drug mechanisms. Metabolomics aims to identify and quantify all of the small molecules within a cell. The relative concentrations of metabolites within cells grown under different experimental conditions, or different species, can be compared using pool size data. Exposing cellular cultures to isotopically labeled compounds allows the progress of metabolites through metabolic pathways to be tracked, known as flux. Multiple Reaction Monitoring (MRM) is a powerful tandem mass spectrometry (MS/MS) tool frequently implemented in proteomic studies to provide targeted analysis of proteins and peptides. The selectivity that MRM delivers is so strong that it provides the quadrupole mass spectrometers (QQQ), on which it is commonly employed, with pertinence to proteomic studies that they would otherwise lack for their relatively low resolution. Two biological systems were studied using LC/ESI/MS/MS on a triple quadrupole operated in SRM (selected reaction monitoring) scan mode. The first bacterium system is aquatic and microscopic in size known as Roseobacter. The second mammalian system is terrestrial and large in size relative to humans known as Holstein cows.

Roseobacter is a clade of marine bacteria abundant in the ocean. Roseophages are viruses that infect Roseobacter and cause viral lysis. Bacterial spores have remarkable resistance to a variety of harsh conditions, causing spoilage in food industry and becoming the primary bacterial agent in biowarfare and bioterrorism. In this study, inactivation mechanisms of Bacillus amyloliquefaciens (BA) spores by non-thermal plasma (NTP) were investigated by using Fourier-transform infrared spectroscopy (FTIR) as a major tool to exam spores after NTP treatment. Pretreated silica gels with distinct silica surfaces containing either isolated silanols, vicinal silanols, siloxanes, or some combinations of the three, were reacted with several organometallic compounds to determine how each compound reacted with the different silica surface species. The organometallic compounds used were trimethylaluminum, triethylaluminum, diethylaluminum chloride, ethylaluminum dichloride, methylaluminoxane, dibutylmagnesium, and bis-cyclopentadienyl dimethyl zirconium.

These reacted silica gels were analyzed using diffuse reflectance infrared Fourier transform spectroscopy and ICP-AES elemental analysis. High concentrations of dissolved iron are present in downgradient groundwater samples adjacent to the western border of the West KL Landfill. Analysis by Rudder (1988) of groundwater samples indicates oversaturation of selected iron oxides, iron sulfides and siderite in monitoring wells TW-4 and M3.

Previous research suggests that pyrogallol, gallic acid, tannic acid and similar organic compounds may retard the oxidation of ferrous iron in the shallow leachate plume of the West KL landfill (Rudder, 1988). The lipopolysaccharides (LPSs) of the Rhizobium phaseoli CE3 and its mutants CE109, CE309 and the transconjugate Rhizobium leguminosarum LB2/109.11 were isolated and analyzed by gas chromatography (GC) and 1H nuclear magnetic resonance (NMR) spectroscopy. CE3 is the wild type strain which is nodulation-proficient (Nod+). Strain CE109 and CE309 have a transposon, Tn5, inserted into Region I and Region III of their respetive chromosomes. The reaction between N-(2-pyridylmethyl)iminodiacetatonickel(II) and copper(II) was studied spectrophotometrically between pH 2.30 and 3.30 over a 10-fold variation in copper concentration at an ionic strength of 1.25M, and a temperature of 25.0+0.1oC. The reaction is first order in both NiPyIDA and copper(II). Terms involving proton attack and a copper dependent dissociation of the nickel complex were resolved from the data. Evidence is given for a stepwise unwrapping mechanism PyIDA from the nickel complex, followed by copper attack to give a dinuclear intermediate. A general mechanism is presented which is consistent with the kinetic data.

A micellar effect upon the rate of hydrolysis of hydroxamic acids in aqueous perfluorooctanoic acid has been demonstrated for hydroxamic acids with chain lengths of six , eight, and ten carbons. In this system the counterion, hydrogenion, is a catalyst. Typical rate constant-surfactant concentration profiles for micellar catalysis were obtained. The results for these hydrolyses are consistent with the pseudo-phase model for micellar effects.

In addition the Arrhenius activation energies were determined for the reaction of octanohydroxamic acid at surfactant concentrations above and below the critical micelle concentration of the surfactant. The activation energy is lower for reaction in the …
