Developmental Expression Of Pdgf, Tgf-A, And Tgf-J Genes In Preimplantation Mouse Embryos, Daniel A. Rappolee, Carol A. Brenner, Richard Schultz, David Mark, Zena Werb

Control of growth and differentiation during mammalian embryogenesis may be regulated by growth factors from embryonic or maternal sources. With the use of single-cell messenger RNA phenotyping, the simultaneous expression of growth factor transcripts in single or small numbers of preimplantation mouse embryos was examined. Transcripts for platelet-derived growth factor A chain (PDGF-A), trans- forming growth factor (TGF)-a, and TGF- 1, but not for four other growth factors, were found in whole blastocysts. TGF-cx, TGF-(31, and PDGF antigens were detected in blastocysts by immunocytochemistry. Both PDGF-A and TGF-a were detected as maternal transcripts in the unferilized ovulated oocyte, and again …

Hearing and memory losses, cognitive decline, easily fractured bones, crankiness and depression are often considered to be inevitable accompaniments of the aging process. Yet many losses which have traditionally been thought of as age determined are, on more careful examination, turning out to be merely age-associated. Many of the declines we associate with being elderly can be explained in terms of lifestyle, habits, diet and other psychosocial factors which are not a necessary part of the aging process

Paget Bone Disease And Heredity: A Case Report, Donato Agnusdei, Roberto Civitelli, Angelo Camporeale, Carlo Gennari

Evidence on familial aggregation of Paget disease of bone shows that the trait is controlled by a single dominant gene. Due to the late onset of the disease, the primary biochemical abnormalities leading to the characteristic roentgenographic features are still unknown. We report the case of a 24-year-old woman who had an elevated serum alkaline phosphatase on routine analysis. Family history revealed that her father and paternal grandmother had Paget bone disease. This pattern is compatible with an autosomic dominant inheritance. Complete laboratory workup confirmed high heat labile alkaline phosphatase values, along with high serum osteocalcin and urinary hydroxyproline excretion. …

A deleterious effect of spaceflight on human health is the loss of bone associated with the prolonged lack of gravitational stress on the skeleton. A potential for bone loss is indicated by the hypercalciuria and decreased heel bone density determined in Skylab astronauts. Histological studies of juvenile laboratory rats in actual spaceflight and in simulation models document a suppression of bone formation with little alteration of bone resorption. The mechanism for net bone loss needs to be defined in order to develop measures for its prevention or correction.

Light touch and low-frequency vibration sense are often impaired in diabetic patients. Meissner corpuscles are distally located mechanoreceptors that respond to these stimuli. Peripheral sensory neuropathy in diabetes progresses from distal to proximal, therefore, Meissner corpuscles are ideally suited to study diabetic neuropathy in the C57BL/Ks (db/db) mouse.
Corpuscles from the forepaw digital pads of 9 diabetic and 9 nondiabetic littermates were studied by electron microscopy. These were divided into 3 age groups; 2.5 months, 10 months, and 17 months old.

The effects of systemic hypercapnia on brainstem metabolism were studied with the [6-14C] glucose autoradiography method. Neuronal pools which are metabolically active take up more [6-14C] glucose and maybe revealed pictorially as darker areas on autoradiograms. Since a high concentration of carbon dioxide stimulates central chemoreceptors and hence enhances respiratory drive, the darker regions on the autoradiograms may represent regions concentrated with respiratory-related neurons. Two discrete regions in the ventrolateral medulla (a column near nucleus ambiguus and a column in the lateral reticular nucleus) appeared dark and were analyzed with an image processing system, which converted …

To aid in the diagnosis of certain clinical types of Herpes virus infections, we evaluated and compared five serological methods for measuring Herpes virus antibody. The five tests we compared are complement fixation test (CF), indirect immunofluorescent antibody assay (IFA), anticomplementary immunofluorescent assay (ACIF), enzyme linked immunoassay test (ELISA), and the latex agglutination test (LA). The main objective of this study was to establish which of the currently available serologic tests was best suited for clinical laboratory use and which may then replace the complement fixation test. The association of obesity and hypertension has been clearly established. Despite evidence that weight loss reduces blood pressure, this aspect of treatment is often not stressed. We studied the effects of nutritional intervention in a group of inner-city, predominantly black, hypertensive patients to determine resultant blood pressure changes. Weight reduction was achieved by 57% of the patients, with a loss of more than 4.5 kg (10 lb) in 18%. Changes in weight were highly correlated with blood pressure changes. Correlations were greater for systolic than diastolic pressures, for pressure measured in the standing than the supine position, and for men …

Minnesota Multiphasic Personality Profiles Of The Morbidly Obese, Philip J. Lanzisera, Allan Dehorn, Carolyn Macdonald, Conni Sroka, J. David Fachnie

Minnesota Multiphasic Personality Inventory profiles of 132 morbidly obese individuals participating in a protein-sparing modified fast were examined via factor analytic methods yielding five interpretable psychometric factors. The results support the hypothesis that the morbidly obese are a psychologically heterogeneous population which can be subdivided conceptually. However, the findings fail to support the hypothesis that any particular psychopathology is intricately linked to the development of morbid obesity.
