Pyk-2 is a protein tyrosine kinase, highly expressed in human hippocampus, dentate gyrus and olfactory bulb. In this study, polyclonal antibodies against the C-terminal of pyk-2, starting from Pro-759, were generated by injecting rabbits with the bacterially expressed GST fusion construct, generated by cloning the C-terminal domain of pyk-2 into pGEX-3X vector and affinity purification to increase the specificity of recognition. Anti-pyk-C was shown to recognize a 123 kD band in Western Blots of 293 cells transfected with pyk-2, as well as PC-12, COS and 3T3 cells and not to cross-react with FAK transfected Sf-21 lysates, in spite of high …

The effects of cyclosporin A (CsA) on rat pancreatic islet \beta and D cells and plasma glucose levels at doses of 50 mg/kg body weight applied for 20 days were investigated. Fifteen Swiss Albino rats were used as the study group and 5 were the control group. Rat pancreatic islet \beta and D cells were stained immunocytochemically by anti-insulin and anti-somatostation antibodies. \beta and D cells from 15 different islets were evaluated qualitatively and quantitatively. Endogeonus opioid peptides play an important role in the generation of the pre-ovulatory LH surge. We investigated the interrelationship between the oploid and indolaminergic systems in the regutalion of LH secretion, and the involvement of \mu-and \kappa-opioid subtypes in this process. Concsious female rats bearing cannula in the femoral artery were injected intraperitoneally with \mu-(diamorphine) and \kappa -(U-50488H) opioid agonists either alone or with their respective antagonists, naloxone and MR2266, on pro-oestrus. Malate Dehydrogenase (MDH) and Lactate Dehydrogenase (LDH) activities were demonstrated in the mitochondrial and cytosolic fractions of an intestinal cestode, Taenia saginata, and then the in vitro effects of three anthelminthics, albendazole, niclosamide, and piperazine on these enzymes were investigated. The Vmax and Km values of MDH were found to be 3.00 \mumol (min mg protein) -1 and 0.0166 mM respectively. The effect of primaquine on the enzymatic antioxidant defence mechanisms of erythrocytes were determined by measuring the activities of the below enzymes, before and after 30 minutes of incubation of hemolyzates with 1 mM primaquine at 37°C. Among the enzymes studied glucose-6- phosphate dehydrogenase seemed the most sensitive enzyme; 57 % and 78 % inhibition at zero (due to the binding of primaquine to NADP binding site) and 30 minutes of incubation (due to the reactive oxygen species), respectively. The consumption of coagulation inhibitors will augment coagulation triggered excessively in sepsis. The determination of levels of these inhibitors may open new substitution treatment schedules to overcome the plateau of the achieved improvement ratio with antimicrobial treatment. PCNA (proliferating cell nuclear antigen) immunoreactivity in the uterus of mature rats was investigated after long term administration of clomiphene citrate and ovariectomy. In the luminal epithelium and glandular epithelium and endometrial stroma regions, few PCNA-immunoreactive cells were observed. PCNA reactivity was estimated for the luminal and glandular epithelium and for the stromal cells. Treatment of clomiphene citrate decreased the proliferation of the luminal epithelial cell and stromal cells to the same extent as ovariectomy. Tympanic membrane perforations arise from a variety of causes. Some of them require myringoplasty. In this study, we used hyaluronic acid, a polysaccharide with high-molecular- weight which may be obtained commercially and affects modulations of cell function. An experimental myringoplasty was performed on rats in which a chronic dry tympanic membrane perforation was formed. While physiological saline was embedded in Gelfoam ® which was supported under and over the graft, in group 1, hyaluronic acid embedded gelfoam was used in group 2. Cardiac troponin T (TnT) is a regulatory contractile protein not normally found in blood. Minor elevations of TnT in patients with unstable angina have been associated with an increased risk of subsequent death or Acute Myocardial Infarction (AMI). We used a neewly developed enzyme immunoassay for TnT to determine whether its presence in the serum of patients with unstable angina was a prognostic indicator. It has been estimated that between 2% to 28% of the general adult population has radiographic evidence of elongated styloid process. In adults, the mean radiographic lenght of the styloid process is 20 to 30 mm and its tip is located between the external and internal carotid arteries, just lateral to the tonsillar fossa. Eagle's syndrome occurs when an elongated styloid process or ossified stylohyoid ligament causes recurrent throat pain or foreign body sensation, dysphagia, or facial pain. One of the factors affecting the accuracy of readings of blood pressure is the equipment used. Defects or inaccuracy of aneroid sphygmomanometers may be source of error in blood pressure measurement. We inspected 100 sphygmomanometers for physical defects and assessed their accuracy against a standard mercury manometer at four different pressure points. 46 of the 100 sphygmomanometers were determined to be intolerant (deviation from the mercury manometer by greater than±3 mm Hg at two or more of the test points). The vitamin D receptor (VDR), a member of the nuclear receptor superfamily, is an integral part of the body's calcium regulatory system. Mammalian and avian VDR genes were isolated, sequenced and characterized. In this study we cloned the vitamin D receptor from amphibians. We received a cDNA library prepared from amphibian (frog) tissues and prepared a cDNA probe based on the avian VDR. We screened the library and located the positive clones. The effects of diets containing 2% and 4% calcium (Ca) on deoxycorticosteron (DOC) salt hypertension were studied in 29 male rats weighting 210-260 g. Rats were categorized into four groups: Group I: control rats (6), group II: DOC hypertensive group (n=8), group III: DOC-low Ca diet (2% CaCl 2 ) (8) and group IV: DOC-high Ca diet (4% CaCl_2 ) (n=7). Diastolic dysfunction is characterized by increased resistance to ventricular filling, inappropriate upward shift of the diastolic pressure-volume relationship and exercise intolerance. Inappropriate tachycardia, decreased diastolic compliance and impaired systolic relaxation lead to diastolic dysfunction. Optimal therapy will depend on the type and pathophysiologic phase of disease. Doppler echocardiography is a valuable tool to diagnose diastolic dysfunction and therapeutic effects of drugs on diastolic function in different groups of patients. This experiment was conducted to investigate the dose-dependent effects of gentamicin (GM) on the neuroepithelial morphology of the posterior crista ampullares (PCA) of the adult chinchilla. The experiment focused on hair cell (HC) morphology to monitor GM's dose-dependent effects and also to define an ototoxically-efficient and safe dose what would subsequently be used in a time course study. There is no ideal surgical technique for cervical spondylosis. The choice of surgical procedure remains controversial. A series of 25 patients who had undergone subtotal vertebrectomy and iliac bone fusion between 1985 and 1995 were reviewed. There were 19 men and 6 women with an average age of 55.6 years. All of the patients had cervical myelopathy and /or radiculopathy due to multi-level spondylosis and 7 with associated ossification of the posterior longitudinal ligament. The protective effect of vitamins E and C on the gastric mucosal barrier in rats irradiated with X-rays was investigated. Thirty-two male Wistar Albino rats were used. The animals were divided into four groups and arranged of follows: the first group (n=8) was the sham group; the second group (n=8) was the control group which was irradiated with X-rays, the third group (n=8) to which vitamin C was administered and the fourth group (n=8) to which vitamin E was given.
