Factors Affecting Apothecia Production And Primary Infection By Monilinia Vaccinii-Corymbosi On Vaccinium Angustifolium, Ian Leonard

Electronic Theses and Dissertations

Mummy berry, caused by Monilinia vaccinii-corymbosi (MVC), is a prolific disease of Vaccinium angustifolium (wild blueberry) leading to decreased yield in wild blueberry fields throughout the Downeast (DE) and Midcoast (MC) regions of Maine (ME). This study aimed to identify factors affecting primary inoculum production and infection by MVC on wild blueberry, and what bud stages of wild blueberry are most susceptible to infection. Through common garden (CGE), field and incubation experiments conducted in 2021 and 2022, factors affecting carpogenic germination of MVC pseudosclerotia and relationships between susceptible wild blueberry buds and environmental factors were analyzed. The CGE conducted in …

Extraction And Characterization Of Antifungal Compounds Produced By Lowbush Blueberry Plants In Response To Monilinia Vaccinii-Corymbosi Infection, Sophia Suriano

Honors College

Maine’s wild blueberries are a vital economic and ecological resource for growers, consumers, and researchers alike. Fungal diseases like Monilinia vacciniicorymbosi (MVC) reduce the yield of berries from infected plants by killing plant tissues and damaging fruit. Understanding what blueberry plants use to defend themselves against fungal pathogens can give a greater insight into increasing plant immunity as a whole. This project aims to better understand the wild blueberry antifungal defense response. I extracted bioactive compounds from the healthy leaves of low severity and high severity disease-affected plants and separated the molecules with thin layer chromatography (TLC). Aspergillus sp. acted …

Genetic Sources Of Resistance To Potato Blackleg Soft Rot Caused By Dickeya Dianthicola, Lucas K. Heroux

Electronic Theses and Dissertations

Solanum tuberosum L. is the world's most important non-cereal food crop, capable of producing more food per land unit on less water than any other crop. Only rice, wheat, and maize are produced in larger quantities than potato. The potato tuber, a modified stem turned storage organ is nutrient dense and a staple in diets across the world. The potato crop is expected to grow and contribute significantly to the global food supply. However, potato production has increasingly been threatened by unfavorable environmental conditions, and susceptibility to pest and disease. Perhaps the most famous of all the Irish Potato Famine …

Determining Effects Of Management Practices On Potato Early Dying And Soil Microbiome And Assessing Risk Of Fungicide Resistance In Verticillium Dahliae, Kedi Li

Electronic Theses and Dissertations

Potato early dying (PED) is a yield-constraining soilborne disease of potato, caused by Verticillium spp. with V. dahliae being the predominant causal agent. Since the pathogen inhabits soil for long periods, PED management aims to reduce the population of V. dahliae in soil. Benzovindiflupyr and azoxystrobin are effective chemicals and frequently used in the control of V. dahliae. In this study, field trials were conducted at Aroostook Farm, Presque Isle, ME in 2019 and 2020. Chemical and biological products have been studied for PED control, and fungicide resistance was also examined. To evaluate fungicide resistance, benzovindiflupyr was characterized on …

Characterization Of Dickeya Dianthicola And Pectobacterium Parmentieri Causing Blackleg And Soft Rot On Potato, Tongling Ge

Electronic Theses and Dissertations

Potato blackleg and soft rot (PBSR), which can be caused by Dickeya spp. and Pectobacterium spp., is a serious problem worldwide. The recent outbreak of PBSR in the Northeastern USA, caused primarily by D. dianthicola, has resulted in significant economic losses since 2015. This seedborne disease is highly associated with and therefore spread by seed tuber distribution. To understand how the outbreak occurred and where the pathogen originated, a total of 1204 potato samples were collected from 11 northeastern states from 2015 to 2020. All the samples were processed for bacterial isolation and DNA extraction. Dickeya dianthicola and P. …

Evaluating The Current Weed Community In Wild Blueberry Fields And Ipm Strategies For Spreading Dogbane (Apocynum Androsaemifolium), Anthony G. Ayers

Electronic Theses and Dissertations

Lowbush blueberry (Vaccinium angustifolium) is Maine’s third largest crop (USDA 2020 a). From 2017 – 2019 the three seasons yield average was 27,200 tons were harvested from 19,500 acres for a value of $22,468,000 (USDA 2020 c). Lowbush blueberries are managed on a two-year cycle. Every other year, lowbush blueberry fields are pruned to the ground either through the use of a tractor mounted flail mower or a prescribed burn (Yarborough 2009). Pruning is a necessary part of managing lowbush blueberries as the second-year growth produces the highest yield but steadily declines in subsequent years (Drummond et al. …

Population Structure And Reproductive Biology Of Monilinia Vaccinii-Corymbosi (Reade) Honey In Lowbush Blueberry In Maine, Katherine A. Ashley

Electronic Theses and Dissertations

Lowbush blueberry, Vaccinium angustifolium Aiton, is both an economically and culturally important crop in Maine, being one of the few endemic crops to North America. The fungus Monilinia vaccinii-corymbosi (Mvc) causes mummy berry disease and is a significant pathogen of both highbush (Vaccinium corymbosum) and lowbush blueberries. While impacts of this disease are not regularly documented, it is estimated that 30-50% of the yield in an unmanaged field can be lost because of Mvc. This disease is typically managed with fungicides or burning of the field during years when the field is pruned, however, the impacts to the …

The Potential For Dickeya Dianthicola To Be Vectored By Two Common Insect Pests Of Potatoes, Jonas K. Insinga Dec 2019

The Potential For Dickeya Dianthicola To Be Vectored By Two Common Insect Pests Of Potatoes, Jonas K. Insinga

Electronic Theses and Dissertations

Dickeya dianthicola (Samson) causing blackleg and soft rot was first detected in potatoes grown in Maine in 2014. Previous work has suggested that insects, particularly aphids, may be able to vector bacteria in this genus between plants, but no conclusive work has been done to confirm this theory. In order to determine whether insect-mediated transmission is likely to occur in potato fields, two model potato pests common in Maine were used: the Colorado potato beetle (Leptinotarsa decimlineata Say) and the green peach aphids (Myzus persicae Sulzer). Olfactometry and recruitment experiments evaluated if either insect discriminates between infected and …

Factors Affecting Survival And Carpogenic Germination Of Pseudosclerotia Of Monilinia Vaccinii-Corymbosi, The Causal Agent Of Mummy Berry, On Vaccinium Angustifolium In Maine, Tyler L. Case

Electronic Theses and Dissertations

The primary focus of this research project was to identify factors in the field affecting survival and the environmental conditions affecting carpogenic germination of pseudosclerotia of Monilinia vaccinii-corymbosi (MVC) which is a common fungal pathogen of lowbush blueberry (Vaccinium angustifolium) in Maine. The interactions between biological oranisms, such as fungi and insects with pseudosclerotia prior to and during the overwintering process were investigated in the research outlined below. Fungi from the surfaces of pseudosclerotia were screened as potential antagonists in a co-plate experiment. Most of the potentially antagonistic fungi tested reduced the width of the MVC reference cultures. These potential …

Chemical And Non-Chemical Control Of Potato Pink Rot, Xuemei (Missi) Zhang Dec 2016

Chemical And Non-Chemical Control Of Potato Pink Rot, Xuemei (Missi) Zhang

Electronic Theses and Dissertations

Pink rot of potato (Solanum tuberosum) is a widespread soilborne disease that causes significant losses in the field and storage. It is caused by Phytophthora erythroseptica (Pethybr.), an oomycete pathogen that produces sexual spores that can survive in soil for years. The management of pink rot mainly relies on chemical control. However, the most effective chemical in pink rot control, mefenoxam, is losing its efficacy owing to the development of mefenoxam resistance in P. erythroseptica. To evaluate alternative fungicides (including chemical and biological fungicides) to mefenoxam in pink rot control, two greenhouse experiments and three field trials …

Reverse Genetic Analysis Of A Cysteine Protease-Encoding Gene (Rd19a) Of Arabidopsis Thaliana In Relation To The Mechanism Of Resistance To The Piercing/Sucking Insect Myzus Persicae, Siobhan A. Cusack

Honors College

A recent study in Solanum bulbocastanum (a wild relative of the cultivated potato) aiming to identify potential genes involved in aphid and pathogen resistance mechanisms found that a homolog of the Arabidopsis thaliana cysteine protease gene RD19a is upregulated during aphid infestation. RD19a is upregulated in response to abiotic stresses such as drought and high salinity, and rd19a mutants show increased susceptibility to bacterial infection. In this study, Arabidopsis rd19a mutants and wild-type plants were subjected to aphid feeding to observe and compare the molecular, physiological and phenotypic responses. The aim was to further establish the proof of concept regarding …
