Cancer is a notable disease due to its prevalence historically and geographically as well as its complexity. Cancer is driven by oncogenes, genes that cause normal cells to turn cancerous. Src is a notable oncogene in that it is present in several common cancers; colon, breast, and prostate cancer. Dr. Sauedo’s lab has demonstrated an interesting relationship between reactive oxygen species (ROS), reactive chemical compounds produced via cellular energy production, and Src. Phosphatase of Regenerating Liver (PRL) proteins regulate a number of important cellular processes, including cell growth and division. Humans have three PRL proteins: PRL-1, PRL-2, and PRL-3. An accumulation of evidence has shown that elevated levels of PRLs are strongly correlated with uncontrollable growth and metastasis of tumors. However, contradictory findings have arisen indicating that PRLs instead function to halt cell division thereby preventing uncontrollable tumor growth. In light of these results, the underlying mechanisms regarding how PRLs function within cellular processes remains unclear. The present study investigated the effects of increasing concentrations of progesterone on cell death by monitoring the expression of mRNAs specific for genes in the apoptosis and autophagy cell death pathway in Danio rerio ovary tissue using quantitative (q)PCR. Progesterone is used to regulate oocyte maturation in vivo; however, synthetic progesterone in oral contraceptives is found in the environment. Primers for Bcl-2 and Pi3K-III were used to investigate gene activity via mRNA produced. The results of the present study revealed that gene activity was variable as revealed by qPCR analysis. Exploration Of The Active Site Specificity Of Mala, A Glucosidase From The Predatory Bacterium Bdellovibrio Bacteriovorus, Christine Isabella

Sequencing of the HD100 genome of Bdellovibrio Bacteriovorus in 2005 revealed a gene for the putative maltase, MalA. However, given the bacterium’s observed disuse of prey carbohydrates as an energy source, this enzyme is seemingly out of place. In this study, the specificity and activity of MalA were explored through various enzymatic assays. Using p-nitrophenol-α-D-glucopyranoside (p-NPG) as a colorimetric substrate to allow for rapid and accurate detection of enzymatic activity through spectrophotometry, enzyme stability inhibition of p-NPG cleavage were explored. While numerous alpha-linked disaccharides were shown to inhibit MalA, only maltose was shown to be cleaved into glucose.
