Within this thesis the source, presence, and the estimated ecotoxicological effects of cardiovascular medicines, specifically statins and beta-blockers are described within the Buckhannon, Tygart Valley, and West Fork Rivers of West Virginia. Included within this thesis is a general introduction providing key background information and relevant research related to the source, presence, and known toxicity of statins and beta-blockers. Harmful algal blooms (HABs) result from outbreaks of any of several different species of toxin-producing phytoplankton and that can have major detrimental effects on marine ecosystems and pose severe health and economic threats to human communities. Of particular concern along the United States West Coast are HABs of pennate diatom genus Pseudo-nitzschia that produce the potent neurotoxin domoic acid (DA). The coastal ocean between Cape Mendocino, CA, and Cape Blanco, OR is a hotspot for Pseudo-nitzschia spp. HABs. The Keweenaw area continues to be influenced by the century of copper mining that ended nearly 50 years ago. This project is focused on Torch Lake, an aquatic ecosystem that has been heavily impacted by mining waste disposal. The watershed has been impaired by mine discharge and tailings, smelter and smokestack plumes, and poor waste disposal practices. The lake is listed as a Great Lakes Area of Concern with beneficial use impairments of restrictions on fish consumption and a degraded benthic community. Microcystin-LR is a hepatotoxin produced by cyanobacteria. Aeromonas hydrophila and Edwardsiella piscicida infections are leading causes of losses in market-sized channel catfish (Ictalurus punctatus). These older fish should have natural immunity in place and a predisposing factor is likely a prerequisite for these disease outbreaks. While microcystin-LR rarely causes mortality in warm-water aquaculture, we believe it may be a predisposing factor that leads to bacterial disease outbreaks during the summer months due to its ability to damage the liver. Fish gills are sensitive to a variety of environmental factors because they regulate the exchange of oxygen and ions between the fish and the external environment. This study examined the effects of polyacrylic acid (PAA) coated titanium dioxide nanoparticle injections (TiO2 NPs) on goldfish (Carassius auratus) over 14 days. Following the experimental period, we examined gill histological samples and looked for signs of an inflammatory response through blood glucose levels and relevant gene expression. TiO2 injected fish developed a more robust ILCM (interlamellar cell mass) compared to the non-injected controls, which remained elevated over 14 days post injection. …

Persistent organic pollutants (POPs) are lipophilic semi-volatile organic chemicals that present a range of challenges to marine biota, specifically marine mammals that often occupy a high trophic position in the food web. POPs have become a global problem since they have been shown to cause immunologic, teratogenic, carcinogenic, neurological, and reproductive complications in living organisms due to their resistance to biodegradation and their lipophilic nature. Marine mammals can accumulate these toxic substances through direct ingestion, trophic transfer, adsorption, and maternal offloading. Heavy metal pollution can have numerous negative impacts on stream fishes, including both lethal and sublethal effects. Because of the sensitivity of fishes to toxins, they are excellent environmental indicators of stream and watershed health. The Tri-State Mining District is a Superfund site located in parts of Missouri, Kansas and Oklahoma that offers a good opportunity to study sublethal effects of heavy metal pollutants on fish behavior. Cyanobacterial harmful algal blooms (cyanoHABs) are frequently observed in water bodies used for recreation and drinking water production and can be detrimental to humans, animals, and general water quality. CyanoHABs are natural occurrences, but human activities such as agriculture, land use change, and runoff from urban and rural landscapes can promote and accelerate their expansion. The lampricide 3-trifluoromethyl-4-nitrophenol (TFM) has been used for decades to control invasive sea lamprey (Petromyzon marinus) populations in the Great Lakes, normally with little harm to other fish populations. However, due to the nature of lampricide treatments and the chemical properties of TFM, adverse effects to non-target fishes and non-target mortality occasionally occur. Polycyclic aromatic hydrocarbons (PAHs), constituents of crude oil, are implicated as a potent source of adverse toxicological effects on living organisms. To model the effects of PAHs in response to environmental oil spill disasters a species of killifish (Fundulus heteroclitus) was captured and exposed to crude oil in a laboratory-controlled setting. Over a period of 7 days, fish were dosed with crude oil by gavage, culled, and organs were harvested for analysis. Exposure of teleost fishes to these contaminants can negatively impact sex differentiation and reproductive output. This research attempts to connect patterns in growth and migration of an anadromous species. The goal of this research was to understand habitat movements and growth of juvenile alewives (Alosa pseudoharengus) in the Penobscot Estuary and Bay through the use of otolith microchemistry, otolith growth increments, and a laboratory stable isotope turnover study. Understanding the connection between growth and movement of juvenile alewives may lead to more accurate and sophisticated conservation and restoration methods for anadromous species.

The lateral giant neuron in the crayfish is a neuron that controls the reflexive escape response; the context and experience-dependent tuning of this response is critical for survival. Serotonin modulates synaptic responses in this neuron, and social experiences (stress) changes the modulatory role of serotonin. I investigated the mechanisms of induced synaptic response changes and serotonergic modulatory changes after silver nanoparticle exposure to ask if 1) contaminant induced stress changes serotonergic modulation in a manner consistent with social stress and 2) shed light on potential neurotoxic effects of widely used, but poorly understood nanomaterials. Contaminated sediments in marine environments have been shown to be good indicators of ecological risk and a means to assess anthropogenic impacts on marine habitats and the animals that inhabit them (Long et al. 1995, Rattner 2009). Although biomarkers are frequently used to assess sublethal effects of contaminants, a lack of mechanistic linkages to higher-level effects limits the predictive power of biomarkers. Bioenergetics has been proposed as a framework for linking cellular effects to whole-animal effects. We investigated sublethal effects of exposure to wastewater treatment facility effluent in freshwater mussels in situ, thereby capturing ecologically relevant exposure conditions. Our study focused on the energetic biomarker AMP-activated protein kinase (AMPK), while also considering more traditional biomarkers like heat shock proteins (HSP70), and antioxidant enzymes (i.e., superoxide dismutase (SOD), glutathione-S-transferase (GST)). The DeepWater Horizon (DWH) oil spill contaminated a large area in the Gulf of Mexico in summer 2010. It is likely that many aquatic species in the Gulf were affected by the spill. Atrazine is among the most heavily applied pesticides worldwide, and recent evidence suggests that it may be unsafe at environmental levels. It is a known endocrine disruptor and a suggested neurotoxin. The US EPA Maximum Contaminant Level (MCL) is 3 μg/L for human ingestion and 200 μg/L for limited human exposure. Several environmentally persistent degradates have been identified, including deethylatrazine (DEA), deisopropylatrazine (DIA) and hydroxyatrazine (HA). No MCLs have been established for these degradates, although some are suggested toxins. The pesticide, 3-trifluoromethyl-4-nitrophenol (TFM), has been highly successful in the control of sea lamprey (Petromyzon marinus) populations in the Great Lakes. Treatments with TFM involve applying it to streams, where it targets larval sea lamprey which live burrowed in the stream substrate. While the toxic mechanism of TFM has been elucidated, and its effects on sea lamprey described, its effects on non-target fish species such as rainbow trout (Oncorhynchus mykiss) and lake sturgeon (Acipenser fulvescens) are not as well understood. Azaspiracids (AZAs), polyether marine toxins, are produced by marine microalgae and accumulate in filter-feeding bivalve molluscs, such as mussels, clams, oysters and scallops, while grazing on phytoplankton, from which they derive nutrition. The Azaspiracids are a class of polyether marine biotoxins that present a significant health risk to consumers of shellfish. Since their discovery in the late 1990s several human intoxications have been reported, and traced to Irish shellfish farms resulting in closures or harvest rejections. As such incidents involving azaspiracid shellfish poisoning have a major socio-economic impact on communities that rely on shellfish farming revenues.

Due to increasing pressure from Ohio River bass anglers with regard to decreasing catch rates in 1981,1990, and 1997, the West Virginia Division of Natural Resources (WVDNR) initiated a study of black bass populations. The Ohio River Valley Water Sanitation Commission (ORSANCO), in conjunction with state and Federal agencies, has collected population data on Ohio River fishes at lock chambers and has measured various water quality parameters since 1957. An okadaic acid (OA) etiology has been demonstrated for most outbreaks of diarrhetic shellfish poisoning (DSP) in Europe, An isomer of OA, dinophysistoxin-2 (DTX-2), has recently been found in Irish waters and, in 1991 and 1994, was the major toxin during prolonged infestations in cultivated mussels (Mytilus edulis) along the south-west coast of Ireland.

The OA class of DSP toxins have been analysed by HPLC using modified versions of three established fluorescent derivatisation methods. A ‘dual tag’ method for the confirmation of specific toxins has also been developed.

Exposure to trace heavy metals is being extensively studied in relation to their adverse and toxic effects on organisms. Processes affecting compartmentalization, biogeochemical cycling, and transport of trace metals in ecological systems must be elucidated if the impact of trace metal releases on biota are to be understood. Because elemental cycles are often closely linked to hydrologic processes, freshwater and marine systems serve as both vectors and sinks for trace metals in the environment. The Effect Of Thermal Increases On The Acute Toxicity Of Copper And Hexavalent Chromium To Channel Catfish (Ictalurus Punctatus), Alan Kirk Millis

48-hr, 96-hr, and incipient median tolerance limits of channel catfish (Ictalurus punctatus Rafinesque) to copper and hexavalent chromium were determined at temperatures of 50, 60, 70, and 80 F. Standard static bioassay procedures were used. The incipient TLm for copper ranged from 0.56 ppm to 2.15 ppm and from 22.5 to 39.5 ppm for hexavalent chromium. The 48-hr and 96-hr results were somewhat higher. The highest resistance for both metals occurred at 70 F. At 80 F, however, there was a sharp decrease in tolerance.
