The ability of DNA tumor virus proteins to trigger apoptosis in mammalian cells is well established. For example, transgenic expression of a simian virus 40 (SV40) T-antigen N-terminal fragment (N-termTag) is known to induce apoptosis in choroid plexus epithelial cells. SV40 T-antigen-induced apoptosis has generally been considered to be a p53-dependent event because cell death in the brain is greatly diminished in a p53-/- background strain and is abrogated by expression of wild-type (p53-binding) SV40 T antigen. The intracellular fate of Chlamydia psittaci during infection of a monocytic cell line, THP1, was characterized. Cytochalasin D inhibited phagocytosis of latex beads but had no effect on infection by C. psittaci, and vacuoles expressed the transferrin receptor, suggesting accessibility to the endocytic pathway. Early Chlamydia-containing vacuoles expressed major histocompatibility complex (MHC) class I molecules, and most vacuoles fused with host cell lysosomes, since they expressed LAMP-1 and had acidic pHs. In cells prestimulated with gamma interferon, vacuoles also expressed MHC class II molecules, suggesting that the monocytes might effectively process Chlamydia-derived antigens for presentation by MHC class I and …

Ozone is a major component of photochemical smog. High levels of this pollutant, sufficient to affect human health are found in many urban areas worldwide. Though limited studies in humans are supported by extensive findings from animal experiments, a difficulty in interpreting the results of these experiments has lead to an ambiguity on the biochemical mechanism of ozone toxicity. Tat and Rev, small proteins encoded by the Human Immunodeficiency Virus type 1 (HIV-1), regulate its pattern of gene expression within infected cells. Tat increases transcription from the integrated pro virus by approximately a thousand-fold; Rev effects a shift in the pattern of splicing of the viral mRNA. Both are likely to interact with cellular proteins in executing their respective functions. Tat has additional activities (including neurotoxicity and inhibition of lymphocyte activation) which may be extraneous to its viral functions.

In order to identify cellular proteins interacting with Tat and Rev, the genes for both were cloned into plasmid vectors …

From 1990 to 1992 we surveyed for Flammulated Owls (Otus flammeolus) in 3 areas in Idaho: Salmon National Forest (SNF), Payette National Forest and adjacent Hells Canyon National Recreation Area (PNF-HCNRA), and Nez Perce National Forest (NPNF). We also collected and summarized information on all historic and modern records of Flammulated Owls in Idaho. Flammulated Owls were detected on 65% of 68 routes (2–16 km in length) surveyed at densities ranging from 0.04 to 1.25 singing males/40 ha. Owls were detected on survey routes as early as 10 May and as late as 23 July. Mean percent canopy …

Totals of 101 native Yellowstone cutthroat (Oncorhynchus clarki bouvieri), 27 introduced lake trout (Salvelinus namaycush), and 40 introduced longnose sucker (Catostomus catostomus) from Yellowstone Lake, Wyoming, USA, were examined for eye flukes. Metacercariae of the trematode fluke Diplostomum were in vitreous humor and/or lens of 94% of Yellowstone cutthroat trout, 92% of lake trout, and 78% of longnose sucker. Longnose sucker had 7% prevalence of infection in both lens and vitreous humor of metacercariae, while Yellowstone cutthroat trout had 3% and lake trout 8%. Recommended Dietary Allowances (RDA's ) reflect that the average healthy American aged 18-24 years should consume 1,200 mg of calcium daily and 10-12 mg and 15 mg of iron for men and women respectively. These RDA's have not been adjusted for those persons involved in endurance sports. Studies by Telford (1992) and Clarkson (1992) show that endurance athletes suffer from calcium and iron deficiencies. There is a general assumption that athletic coaches are more concerned with the "weigh in" of 4 athletes than overall nutrient requirements. Positive nutrition education should be part of the overall training of athletes. Also, studies …

Serum pepsinogen, ui-antitrypsin (ui-AT) and blood groups were studied as genetic markets in 32 patients with endoscopically proven duodenal ulcer and 44 control subjects with no family history of ulcer disease. Serum pepsinogen was detennined by the modified method of Edward et al7, a1-AT by single radial hnmunodiffusion8 (RID) and phenotyping was carried out by isoelectric focusing (IEF)9. Duodenal ulcer patients with hyper- pepsinogenemia (28%) and low serum ui-AT (35%) had a dominant blood group 0, lower mean age, an early onset of disease, a higher frequency of gastrointestinal (CI) bleeding and ulcer perforation. Polycyclic aromatic hydrocarbons (PAHs), are known immunotoxicants in animals, and are suspect toxins to the human immune system. The mechanism(s), however, by which PAHs exert immunosuppression have not been fully elucidated. Previous studies conducted in our laboratory have shown that PAHs, such as 7,12-dimethylbenz(a)- anthracene (DMB A) and benzo(a)pyrene (BaP) may exert their immunotoxic effects by altering intracellular calcium (Ca2+) homeostasis in lymphocytes. Intracellular Ca2+ is an important second messenger in the immune response, and the mobilization of Ca2+ is critical in the transduction of intracellular signals from the plasma membrane to the nucleus. Pinus aristata Engelm. forest vegetation in Colorado was studied to determine vegetation composition and the relationship between vegetation and environment. Species percent cover, tree size class, and environmental variables were recorded for 49 plots. Previously collected data for 4 plots from New Mexico were included. Environmental variables included latitude, longitude, elevation, slope, aspect, topographic position, parent material, percent rock cover, mean rock size, litter depth, estimated plot age class, and evidence of anthropogenic disturbance. Soils were analyzed for texture, depth, and percent carbon and nitrogen. Pinus aristata foliage was analyzed for percent nitrogen and phosphorus. Direct and indirect gradient analyses …

The purpose of this study was to examine predictors of social physique anxiety (SPA). SPA, self-esteem, body-esteem, public body consciousness (PBC) and percent body fat (%BF) were assessed with elite female youth athletes (N = 68) competing in either figure skating, soccer or gymnastics. Stepwise multiple regression analyses, controlling for BF%, accounted for 59% of the variance in SPA. Self-esteem entered first, and BF%, followed by body-esteem and PBC. The psychological variables accounted for 57% of the variance with self-esteem contributing the most (R square change = 45%). Contrary to previous research, BF% did not significantly contribute to SPA. …

Early in S phase, the vacuole (lysosome) of Saccharomyces cerevisiae projects a stream of vesicles and membranous tubules into the bud where they fuse and establish the daughter vacuole. This inheritance reaction can be studied in vitro with isolated vacuoles. Rapid and efficient homotypic fusion between saltwashed vacuoles requires the addition of only two purified soluble proteins, Sec18p (NSF) and LMA1, a novel heterodimer with a thioredoxin subunit. We now report the identity of the second subunit of LMA1 as IB2, a previously identified cytosolic inhibitor of vacuolar proteinase B. Both subunits are needed for efficient vacuole inheritance in vivo …

With respect to the ethical debate about the treatment of animals in biomedical and behavioral research, Harry F. Harlow represents a paradox. On the one hand, his work on monkey cognition and social development fostered a view of the animals as having rich subjective lives filled with intention and emotion. On the other, he has been criticized for the conduct of research that seemed to ignore the ethical implications of his own discoveries. The basis of this contradiction is discussed and propositions for current research practice are presented.

A mannose-binding lectin, termed champedak lectin-M, was isolated from an extract of the crude seeds of champedak (Artocarpus integer). On gel filtration chromatography, the lectin eluted in a single peak at elution volumes corresponding to 64 kDa, SDS-PAGE showed the mannose-binding lectin to be composed of 16.8 kDa polypeptides with some of the polypeptides being disulphide-linked to give dimers. When tested with all isotypes of immunoglobulins, champedak lectin-M demonstrated a selective strong interaction with human IgE and IgM, and a weak interaction with IgA2, The binding interactions of lectin-M were metal ion independent. Various fungal products have immunomodulating activity and some have been studied regarding prevention of transplantation rejection. Prior to this investigation, the mycotoxin, gliotoxin (GT), has never been investigated as an immunotherapeutic drug for autoimmune disease. GT is a fungal secondary metabolite and a member of the epipolythiodioxopiperazine (ETP) family which has been shown to inhibit phagocytosis, induction of cytolytic T cells and the proliferation of T cells following mitogen stimulation. GT also induces in vitro apoptosis in certain immune cell types. More importantly, GT exhibits selective activity towards cells of hemopoietic origin.

A 1994 survey of 10,000 households by the National Sporting Goods Association (1994) found that approximately 40,000 children between the ages of 7-17 years participate in an organized sport or recreational activity annually. In the realm of organized sports, especially endurance activities, there continues to be interest in how cardiovascular fitness is attained and how it can be maximized. Measurement of cardiovascular fitness is an important piece to that puzzle.

Insertion mutations in two Vibrio cholerae genes, cya and crp, which encode adenylate cyclase and the cyclic AMP (cAMP) receptor protein (CRP), respectively, derepressed the expression of a chromosomal cholera toxin (CT) promoter-lacZ fusion at the nonpermissive temperature of 37 degrees C. In the classical biotype strain O395, the crp mutation increased the production of both CT and toxin-coregulated pilus (TCP) in vitro under a variety of growth conditions not normally permissive for their expression. Concentrations of indicator bacteria such as fecal coliform (FC) and fecal streptococcus (FS) are often used to assess the suitability of waters for their intended use(s) and to allocate resources for water quality improvement measures. There is evidence, however, that concentrations of FC and FS can be influenced by variables such as season and flow rate during sampling, which could lead to biased results. The objective of this study was to assess the impacts of season and flow rate on concentrations of FC and FS. The effects on feeding of moving animals from individual- to pair-housing were explored in the present three experiments. Wheel access is known to result in a temporary suppression of feeding. The move to pair-housing was compared with the effect of wheel access. In Experiment l, a group of rats was moved from individual to pair-housing (IP group) and compared to a group of individually-housed rats (IND group) and a group of chronically pair-housed rats (PAIR group). The animals in the IP group showed a temporary (three days) suppression in feeding (initially 23%). When half of the IND and PAIR group …
