Our understanding of human brain function can be greatly aided by studying analogous brain structures in other organisms. One brain structure with neurochemical and anatomical homology throughout vertebrate species is the locus coeruleus (LC), a small collection of norepinephrine (NE)-containing neurons in the brainstem that project throughout the central nervous system. The LC is involved in nearly every aspect of brain function, including arousal and learning, which has been extensively examined in rats and nonhuman primates using single-unit recordings. Sustainable irrigation methods maximize agricultural productivity in Northern African countries like Sudan. This project evaluates soil properties to develop optimized irrigation strategies for Sudan's arid and semi-arid climate. Assessing soil characteristics such as texture, infiltration rates, and nutrient content informs the selection of modern irrigation systems. Data from 3,192 locations, collected using GPS and ring infiltrometers for infiltration rates, alongside laboratory analyses for soil properties, were employed. A ranking system determined suitable irrigation systems for specific soil classes, considering factors like slope, wind, crop tolerance, available water capacity (AWC), drainage, germination, capital cost, labor cost, maintenance, and evapotranspiration (ET). The species-specific migratory patterns and strategies of many songbirds remain unknown or understudied, as research in animal ecology is biased toward the breeding period, with the fewest studies on the migratory period across taxa. Identifying large-scale spatiotemporal migratory patterns is challenging, as individuals within a species may vary in their migratory behavior and strategies. The Yellow Warbler (Setophaga petechia) is a Nearctic-Neotropical migrant that is relatively well studied during the breeding season, but its species-wide migratory patterns remain understudied. Wheat streak mosaic virus (WSMV) and Triticum mosaic virus (TriMV) present a great threat to wheat production in the Great Plains area of the United States due to the loss of photosynthetic area causing reductions in crop yields. Effective control of these viruses is limited to a few available strategies including controlling volunteer wheat and using currently identified resistant genes to both the vector and viruses. While effective, these genes show temperature sensitivity and yield drag, and evidence that the vectors and viruses can overcome them has been found. As species respond to changing climate, distributions and abundances may shift and alter species interactions. Hybridization, a relatively widespread phenomenon becoming more common with climate change, can have beneficial and detrimental effects on population growth rates and genetic integrity. Beneficial effects due to the introduction of advantageous alleles and increased genetic diversity may result from hybridization. Species may also accrue fitness costs associated with changing climates if mismatches occur between environmental variables and phenotypes. I examined the effects of virtual fencing on beef cattle stress and how implementing the technology can aid wildlife habitat conservation at the University of Nebraska Gudmundsen Sandhills laboratory (GSL) near Whitman, NE. First, I examined how virtual fencing influences acute heart rate changes in beef cattle fitted with the virtual fencing collars. No significant difference in the average heart rate of a cow (P > 0.05) was observed between the 30 min control and virtual fenced periods for both trial dates. Mountain lions (Puma concolor) have been studied extensively throughout their wide geographic distribution in the Western Hemisphere, but most research has been conducted at relatively small spatial scales. We used GPS telemetry data contributed by researchers in 16 study areas throughout California to advance understanding of mountain lion spatial ecology. First, we investigated multi-scale resource selection of mountain lions relative to spatially varying risk of intentional human-caused mortality associated with livestock depredation management. Gambusia are likely relatives of guppies and are known as mosquito fish because they largely prey on mosquito larvae. For a century, they have been used locally and globally in mosquito control. Delineating irrigated land supports decision-makers and promotes the sustainable use of this crucial resource. Global agriculture relies heavily on the use of synthetic nitrogen fertilizer to meet the current global food demand. Unfortunately, the average nitrogen-use efficiency (NUE) of maize (Zea mays) is approximately 50 percent. Observing enzyme structures while they are catalyzing a reaction has been a major goal of both enzymology and structural biology for decades. With the advent of time-resolved serial X-ray crystallography using X-ray free electron lasers (XFELs) and synchrotron sources, enzyme reactions can be monitored in real time in crystallo. Here, we use two different approaches to study related enzymes involved in methylglyoxal detoxification. Misophonia is a disorder that elicits an extreme negative reaction to specific auditory and visual stimuli. Triggering stimuli, which are often human produced, provoke an array of symptoms including heightened negative emotions along with autonomic nervous system responses. Despite being a relatively common disorder, misophonia is still inadequately recognized and is often classified under conditions such as obsessive-compulsive disorder and anxiety. No cure for misophonia currently exists, but there are many treatments and coping mechanisms used by people with misophonia to ease their symptoms. As climate change progresses, wildfires will become more common, increasing their impact on soil microbial ecosystems. This research examines how increased fire frequency affects soil microbial communities and whether effects differ for communities associated with different tree species. Soil samples were collected from under five individuals of four tree species within each of four 1-ha plots that had experienced fire from 24 to 111 years ago. Samples were analyzed for microbial DNA using next generation sequencing techniques in order to explore the influence of fire return interval and tree species on microbial diversity and community structure. News media provide an accessible health information source for patients and the public, but are often criticized for overemphasizing benefits and underplaying risks of novel biomedical interventions. Overly optimistic portrayals of unproven interventions may influence public and patient expectations; the latter may cause patients to downplay risks and over-emphasize benefits, with implications for informed consent for clinical trials. Investigating The Antimicrobial Potential Of Bovine Lactoferrin Against The Neonatal Pathogen, Staphylococcus Capitis, Anna Desmond, Lesley Cotter, Des Field, Fiona O'Halloran

Lactoferrin is an antimicrobial glycoprotein that demonstrates a broad-spectrum of activity against a wide variety of clinical pathogens. This study investigated the potential of bovine lactoferrin (bLf) against multidrug resistant Staphylococcus capitis (S. capitis) strains. Growth curve analysis and time-kill curves demonstrated that at 750 µg ml−1 lactoferrin significantly inhibited (50.6%, P < 0.05) the growth of most isolates tested (90%), and this effect was based on a bacteriostatic mechanism. At the same concentration, bLf also significantly inhibited (30%, P < 0.05) biofilm formation in 40% of strains tested. Combinations of bLf with selected antibiotics were assessed for enhanced antimicrobial activity using growth curves. BLf combined with β-lactam antibiotics reduced the growth of S. capitis strains, however, the effects were not significant. BLf displays antimicrobial effects against multidrug resistant S. capitis isolates, but with strain-specific effects.

Clostridium difficile is a major threat to public health and primarily affects at risk groups such as elderly, immunocompromised individuals, and individuals on antibiotics. Many decades of research have been conducted to figure out ways to exclude or reduce C. difficile colonization and toxin production. Two major proposed mechanisms for colonization resistance are production of secondary bile salts and competition for nutrients.

In Chapter 2, we found that antibiotic treatment disrupted bile salt metabolism increasing the levels of the primary bile salt cholate and decreasing the levels of deoxycholate, a secondary bile salt. However, bile salts were not required …

Sphingolipids are pivotal for plant development and stress responses. Growing interest has been directed towards fully comprehending the regulatory mechanisms of the sphingolipid pathway. In this study, we explore its de novo biosynthesis and homeostasis in Arabidopsis thaliana cell cultures, shedding light on fundamental metabolic mechanisms. Microbiomes are a fundamental part of eukaryotic organisms and play a role in both host health and fitness. Although microbiomes are often associated with health and positive effects on the host, certain microbiome compositions are also associated with disease. We used Caenorhabditis elegans and its experimental microbiome, CeMbio to explore the interactions that individual microbiome members have with other microbes, with the host, and how the entire microbiome community interacts with the host. Swine Influenza A Virus (IAV-S) imposes a significant burden on the pork industry and poses a great threat to public health due to its zoonotic potential. Susceptibility to human, avian, and swine influenza make pigs the ultimate mixing vessels for reassorted influenza A virus strains. Modern breeding programs rely heavily on efficiently screening large numbers of genotypes for agronomic traits, such as disease resistance, drought tolerance, and yield. Identifying the genetic loci or genes associated with these traits using GWAS or functional analyses will benefit future plant breeding efforts seeking to incorporate these traits into new crop varieties, whether through conventional breeding or gene editing techniques. Unmanned aerial vehicle (UAV)-based image data has been increasingly used for this task, as it allows entire test plots to be quickly and cost-effectively phenotyped. The work of the dissertation investigated the efficacy of foam-based decontamination treatments to enhance microbial safety in Roma tomatoes, specifically targeting natural microflora and Salmonella contamination. Growing concerns over foodborne illnesses linked to fresh tomatoes and the limitations of conventional decontamination techniques, such as organic matter built-up and limited microbial reduction, are addressed. Cell-cell signaling molecules known as neuropeptide and peptide hormones, distributed in both the central nervous system (CNS) and the peripheral nervous system (PNS), regulate a variety of physiological processes including body temperature, feeding, circadian rhythm, reproduction, energy homeostasis, and more. While a number of active neuropeptides structures with physiological functions have been discovered, advances in peptidomic mass spectrometry are enabling researchers to uncover novel, previously unknown, active neuropeptide structures with potential for bioactivities. Using liquid chromatography coupled with tandem mass spectrometry (MS/MS), it is possible to identify a wide range of neuropeptides in single experiments without preselecting specific peptides. Small extracellular vesicles (SEVs, “exosomes”) are natural nanoparticles that can be endogenously synthesized and also obtained from dietary sources, such as bovine milk SEVs (mEVs). mEVs have been extensively studied and discovered with contributive features that potentially play a role in drug delivery and other therapeutic purposes, such as diagnostic tools. Current studies have shown that mEVs have been used preclinically to carry in targeting drugs, nucleotides, biomolecules, and labels to target metabolic diseases and cancers.
